Abstrakt: This bachelor thesis focuses on extracellular vesicles (EVs), which are small transport units derived from membranes and released by a variety of cells. Based on their biogenesis, two main classes were observed. First class, referred to as exosomes, are vesicles formed during an endosomal pathway and released by multivesicular bodies. The second class, microvesicles, are formed by a direct budding of the plasma membrane. We focused on EVs of different life stages of the tapeworm Schistocephalus solidus (Cestoda: Diphyllobothriidae), a parasite of fish-eating birds with a three-host life cycle. For our experiments, a full life cycle of S. solidus was established under laboratory conditions. EVs secretion was studied in procercoids fom copepods, plerocercoids from fish and adults from the cultivation medium. We have observed biogenesis of EVs on the surface of procercoid and plerocercoids of S. solidus using transmission electron microscopy and successfully purified EVs from the cultivation medium. Furthermore, we observed differences in secretory activity during the maturation of plerocercoids and a difference in releasing EVs from different parts of the the tapeworm surface.
