Kohezinové geny hrají důležitou roli v buněčném dělení. Zajišťují správnou segregaci chromozomů během mitózy i meiózy. Tato práce se zaměřuje na roli kohezinových genů při meióze u modelového organismu myši domácí (Mus musculus). V práci jsou nejprve uvedeny klíčové procesy savčí meiózy. Dále je popsána struktura kohezinového komplexu. Ten se skládá z heterodimeru SMC proteinů - SMC3 a SMC1α nebo SMC1β, který je uzavřen do kruhu štěpitelnou podjednotkou RAD21, RAD21L nebo REC8. S ní pak dále asociuje čtvrtá podjednotka - STAG protein (STAG1, STAG2 nebo STAG3). Dále jsou podrobně rozebrány funkce čtyř proteinů specifických pro meiózu (SMC1β, RAD21L, REC8 a STAG3) odvozené od projevů deficiencí jejich genů u myších samců. Všechny čtyři geny jsou nezbytné pro mnoho procesů v rámci meiózy - např. udržení koheze sesterských chromatid, synapsi nebo rekombinaci. STAG3, SMC1β, a REC8 jsou nutné k udržení centromerické koheze sesterských chromatid. STAG3 a RAD21L jsou důležité pro asociaci ostatních kohezinových podjednotek. Nejdůležitějším projevem deficience těchto čtyř genů je meiotická zástava v profázi I. Proto je výzkum kohezinů důležitou součástí studia příčin sterility u savců. klíčová slova: kohezin, meióza, spermatogeneze, myš, Smc1b, Rec8, Rad21l, Stag3 Cohesin genes play an important role in cell division. They ensure proper chromosome segregation during mitosis and meiosis. This study is focused on the role of cohesin genes during meiosis in male house mouse (Mus musculus). At first, this study introduces key processes of mammalian meiosis. Next, the structure of cohesin complex is described; it consists of a heterodimer SMC proteins - SMC3 and SMC1α or SMC1β, which are enclosed to the ring by cleavable subunit RAD21, RAD21L or REC8. Fourth subunit - a STAG protein (STAG1, STAG2 or STAG3) associates with the cleavable subunit. Meiotic function of specific cohesin proteins (SMC1β, RAD21L, REC8 and STAG3) as deduced from the phenotypes of the deficiencies of their genes in male mouse is depicted. All these four genes are necessary for many processes during meiosis, - e.q. sister chromatid cohesion maintenance, synapsis and recombination. STAG3, SMC1β, and REC8 are necessary for centromeric cohesion. STAG3 and RAD21L are important for the assembly of the remaining cohesin subunits. The most important phenotype of deficiency of all four genes is the complete meiotic arrest in male prophase I.
