Slinivka břišní vylučuje řadu látek podílejících se na regulaci fyziologických procesů v organismu. Po dlouhou dobu se vědělo, že beta buňky slinivky vylučují insulin. Spolu s ním vylučuje ale i další látky, mezi nimi mnoho peptidů, jako jsou insulinu podobné růstové faktory IGF, kratší peptidy jako amylin a jiné. Ve směsi těchto peptidů produkovaných slinivkou byl v roce 2001 skupinou Ch. Buchananové z Univerzity v Aucklandu identifikován peptid dlouhý 34 aminokyselin, které sekvencí přesně odpovídají části proformy insulinu podobného růstového faktoru 2 (pro-IGF-2). Tento peptid byl nazván preptin. Následně bylo zjištěno, že preptin má pozitivní vliv na proliferaci osteoblastů a že zesiluje sekreci insulinu z beta buněk v přítomnosti glukosy. V současnosti je aktivně studován zejména jeho vliv na proliferaci kostních buněk za účelem možného využití v léčbě osteoporózy. Tato práce se soustředí na přípravu a charakterizaci lidského a potkaního preptinu a také jejich dvou fragmentů obsahujících aminokyseliny 1-16 a 17-34 syntézou peptidů na pevné fázi. Další část je zaměřena na testování vlivu potkaního preptinu a jeho fragmentů na sekreci insulinu v linii potkaních beta buněk INS-1E. Součástí práce jsou také vazebné testy lidského preptinu a jeho fragmentů na insulinový receptor a IGF-1 receptor... in English Pancreas is an endocrine tissue which affects many physiological processes in organism. It has been known for a long lime that the pancreatic beta cells secret insulin. However, there are more hormones and substances secreted from the beta cells, mainly peptides such as insulin-like growth factors IGF, shorter peptides like amylin and many more. Some of them have not been even identified. Among these pancreatic products Ch. Buchanan's group from Auckland University, New Zeland identified 34 amino acids long peptide called preptin in 2001. Its amino acid sequence is identical with part of the preform of insulin-like growth factor 2 chain. It has been discovered that preptin has effect on osteoblast proliferation and insulin secretion. Nowadays preptin is being studied like a potential osteoporosis cure thanks to its increasing effect on osteoblasts proliferation. This thesis describes preparation and characterization of human and rat preptin and their two fragments (which contain amino acids 1-16 and 17-34) using solid-phase synthesis. The aim of the thesis is to test effects of rat preptin and its two fragments on insulin secretion in pancreatic beta cell line INS-1E.
