[cze][eng] Neuropatogenní motolice Trichobilharzia regenti (Schistosomatidae) infikuje centrální nervovou soustavu ptáků a savců. Během své migraci míchou poškozuje nervovou tkáň a vyvolává zánět, který je při infekci savců patrně zodpovědný za eliminaci parazitů. Tato práce s využitím imunohistochemického značení detekovala v postižené míše myší C57BL/6J během infekce prozánětlivé cytokiny IL-1β a IL-17. Zároveň však byly identifikovány i IL-4 a IL-6, které se patrně podílely na regulaci zánětu a poškození tkáně. Tyto cytokiny byly produkovány mikrogliemi a astrocyty, ale i dalšími neidentifikovanými buňkami. Reakce periferní imunitní odpovědi byla charakterizována na základě fenotypizace T lymfocytů ve slezině infikovaných myší pomocí průtokové cytometrie. Produkované cytokiny v době kulminace zánětu v míše vypovídaly o smíšené imunitní odpovědi (Th1/Th2). Výsledky této práce prohloubily znalosti o buněčné imunitní odpovědi myší při infekci T. regenti a potvrdily vliv zánětu v míše na polarizaci periferní imunitní odpovědi. During its migration through the spinal cord, the parasite causes tissue damage and triggers inflammation which is likely responsible for the elimination of the parasite. In this thesis, the proinflammatory cytokines IL-1β and IL-17 were detected by immunohistochemistry in the affected spinal cord of C57BL/6J mice during the infection. Additionally, IL-4 and IL-6, participating in the regulation of the inflammation and tissue repair, respectively, were noticed. Astrocytes, microglia and other, yet unidentified cells, produced these cytokines. Furthermore, splenic T-lymphocytes were phenotyped by flow cytometry to characterize peripheral immune response. At the peak of nervous tissue inflammation, mixed (Th1/Th2) cellular immune response was observed. Taken together, this thesis extended the knowledge of cytokine immune response of mice infected with T. regenti and also confirmed that inflammation in the nervous tissue influences the polarization of peripheral immune response.
