Peroxizomy jsou malé eukaryotické organely, známé zejména pro svou schopnost vytvářet a degradovat peroxid vodíku. Jejich enzymy však zastávají mnoho dalších významných funkcí, podílí se na beta-oxidaci mastných kyselin s velmi dlouhým nebo větveným řetězcem, prostřednictvím alfa-oxidace umožňují zpracování molekul, které nemohou být degradovány v beta-oxidačním cyklu, také se účastní počátečních kroků syntézy éterických lipidů nebo formování žlučových kyselin. Výše zmíněné aspekty činí peroxizomy potenciálně zajímavými pro jejich možný vliv na metabolismus srdce, který je založen především na oxidativním zpracování mastných kyselin, avšak dosud o této problematice není mnoho známo. Práce se zaměřuje jednak na vznik a funkce samotných peroxizomů, ale také srdeční metabolismus společně s možnou rolí peroxizomů v něm. Peroxisomes are small eucaryotic organelles, mainly known for their ability to create and break down hydrogen peroxide. However their enzymes play other significant roles, they participate in beta-oxidation of fatty acids with very long or branched chains, through alpha-oxidation they enable to process molecules that cannot go through beta- oxidation cycle, they also participate in early steps of synthesis of ether-lipid or bile acides. Above mentioned aspects make peroxisomes potentionally interesting for their possible influence on heart muscle metabolism, that is dependent on oxidative degradation of fatty acids, although not very much is known about this issue. The thesis focuses on biogenesis and function of peroxisomes, but also on their possible role in heart muscle metabolism.
