[cze][eng] Lenka Zdražilová Produkce sekundárních metabolitů v suspenzní kultuře Trifolium pratense L. Souhrn Elicitace je metoda využívající obranných mechanizmů rostlin ke zvýšení produkce sekundárních metabolitů v rostlinách i v kulturách in vitro. Kyselinu jasmonovou můžeme zařadit mezi biotické elicitory, protože v rostlinách působí jako induktor exprese obranných proteinů. V experimentu byl sledován vliv čtyř koncentrací kyseliny jasmonové a vliv kyseliny jasmonové v kombinaci s vápenatými ionty a verapamilem na produkci flavonoidů a isoflavonoidů suspenzní kulturou Trifolium pratense L. (varieta Sprint). Kultura byla kultivována při teplotě 25řC a světelné periodě 16 hodin světlo / 8 hodin tma na živném médiu podle Gamborga s přídavkem 2 mg.l-1 2,4-dichlorfenoxyoctové kyseliny a 2 mg.l-1 6-benzylaminopurinu. U elicitovaných i kontrolních vzorků bylo provedeno fotometrické stanovení flavonoidů dle Českého lékopisu 2009 a stanovení isoflavonoidů metodou HPLC. Z výsledků vyplývá, že optimální vliv kyseliny jasmonové na produkci flavonoidů a isoflavonoidů se projevil po 24hodinové aplikaci koncentrace 50 µmol. Stimulace elicitace extracelulárními vápenatými ionty se nejvíce projevila u nejslabší koncentrace kyseliny jasmonové (5 µmolů) po přidání chloridu vápenatého o koncentraci 1 mmol a 10 mmol. Přidání...Lenka Zdražilová The production of secondary metabolites in the Trifolium pratense L. suspension culture Summary Elicitation is a method making use of protective mechanisms of plants to increase the production of secondary metabolites in plants and cultures in vitro. Jasmonic acid we can include into biotic elicitors, because it acts in plants as an inducer of the expression of protective proteins. In the experiment examined the impact of four concentrations of jasmonic acid and the impact of jasmonic acid in combination with calcium ions and verapamil on the production of flavonoids and isoflavonoids by the Trifolium pratense L. suspension culture (variety Sprint). The culture was cultivated in Gamborg medium to which 2 mg.l-1 of 2,4- dichlorophenoxyacetic acid and 2 mg.l-1 of 6-benzylaminopurine were added, at the temperature of 25řC, 16-hr light/8-hr dark period. The elicited and the inspection samples underwent the photometric determination of flavonoids in accordance with the Czech Pharmacopoeia 2009 and the determination of isoflavonoids via the HPLC method. The results show that the optimal effect of jasmonic acid on the production of flavonoids and isoflavonoids was manifested after a 24-hour application of the 50 µmol concentration.
