Optimization of a loop-mediated isothermal amplification method for a detection of Haemophilus influenzae infection.

Haemophilus influenzae is a gramnegative, rod-shaped and predominantly human pathogen. Infection caused by this microorganism is especially dangerous for children under five years of age, who are mainly at risk of pneumonia and meningitis.Althought vaccination is available against its most dangerous type b, it still causes problems, especially in developing countries. Due to the insufficient vaccination, strains that are more resistant to the antibiotics are more spreading in these countries. Equipment requirement of current detection options for H. influenzae infection(e.g. polymerase chain reaction) causes, that reliable detectionis a problem in poor countries. Loop-mediated isothermal amplification could be a cheap and reliable method. The aim of this bachelor thesis was the design of sets of primers and their applicationinthe LAMP reaction. The designed primers targetedthe ompP6 gene, which was alreadythe target of LAMP reaction in some studies and is also a frequent target in PCR detection.As a part of the optimization, different volumes of the reaction mixtures and multiple detection options were tested -colorimetric detectionusinganacid-base indicator and fluorescence detection using different dyes monitored in real time. Games in logopedic prevention

The bachelor thesis deals with the topic of Games in logopedic prevention. Its aim is to show the role of games in speech therapy prevention and present their offer, which could serve as an inspiration or a help for kindergarten teachers. In the theoretical part I first focus on ontogenetic speech development which is divided into individual periods and accompanied by preventive recommendations. The second part deals with the speech therapy prevention and its implementation in preschool facilities. I mention the importance of language and speech development within RVP PV, further supported areas of speech therapy prevention such as an auditory and visual perception, general rules for proper development speech and methodology of speech therapy prevention, which includes a game. In the third part I deal with the game itself its meaning, characters and their division. In the practical part there are ten games, which were tested by children in kindergarten. Individual games are focused on the development of skills that are of interest to speech therapy prevention. Based on the observations, the games are described and evaluated. I observed especially the interest of children in the games, but I also focused on the possibility of using games individually or in groups or in different parts of the day. There is a book of games and photographs of the aids I have created for the games.

The bachelor thesis consists of a translation of a part from a French publication called L'Environnement and of a commentary on the translation. The source text deals with different aspects of general ecology and environment protection. The translation contains a part of the first chapter concerning general ecology and the fourth chapter treating animal and plant protection. In the commentary, the source text is analysed and diverse translation problems and possible solutions are examined.
The puppet and its influence on a child's personality development

The bachelor's thesis deals with the influence of puppet theater in the personal development of preschool child. The aim of the work is to find out which puppets preschool children prefer and what influences their choice - material, color, symmetry. The work examines childrens reactions to a particular type of puppet theater. The theoretical part defines the terms of the puppet theater, introduces readers to the puppet types, the historic context of the development of puppet theater. It deals with the developmental traits of preschool age children and using puppets as an educational resource. The practical part is a form of interview and observation investigated action puppets and puppet theater for children.
