Jiří Langer - The try of Monography

The goal of this diploma work is to introduce the life and the work of Jiří Langer - one of the most interesting Czech Jewish authors. Langer did not use only Czech to express his ideas, but also Hebrew and the German language. I introduced him as a Czech author because I am convinced that it was mostly the Prague environment that formed him. The second important influence was his experience of Hasidism. Jiří Langer entered the world of literature thanks to the study Die Erotik der Kabbala and other Judaic works - in this period under the influence of Freud. He published in Czech his master work Devět bran (Nine Gates) in 1937 - an interesting collection of Hasidic legends, which is not comparable to any other works. In 1938 he succeeded to publish Talmud. Ukázky dějiny. (The Talmud. Excerpts and History) - a Judaic publication destined for Czech clerks in Galicia. He proved his translation and poetic abilities by publishing the anthology of Hebrew poetry from the 10th to the 19th century called Zpěvy zavržených (Songs of the Rejected) in 1938. The last two books, he managed to publish, was the Hebrew poetry. The first Piyutim veShirei Yedidut appeared in Prague in 1929, the second Me'at Tzori was published after his death in Palestine in 1943. Among his friends were Franz Kafka, Max Brod and Dov Sadan....

The main aim of presented work is to evaluate the importance of standardized echography in the diagnosis of uveal melanoma and in terms of determining the prognosis important morphological features in vivo.

Antimicrobial activity of bacteria isolated from Pectinatella magnifica

The thesis is comprised of two parts. The first part is dedicated to the literature search, overview, which is focuses on the summary of information that was previously found not only about the life and reproduction of freshwater bryozoan Pectinatella magnifica, but also the importance of marine species bryozoans. Furthermore, importance of antimicrobial active substances and most commonly used laboratory methods for determination of microbial activity are shown in the report characteristics. The aim of the review was to summarize the most important facts underlying the experimental hypothesis described in the second part of this work. The reason for testing the antimicrobial activity of symbiotic bacteria isolated from Pectinatella magnifica is that antimicrobial activity of symbiotic bacteria and also bryozoans´ extracts were demonstrated in the marine bryozoans. Because of this, deeper knowledge of these largely unknown animals has become of high importance. Bacteria isolated from Pectinatella magnifica colonies collected in 2012 and 2013 in South Bohemia were tested for their antimicrobial aktivity against many strains of bacteria. Diffusion method was chosen for testing of the antimicrobial activity. Instead of application by paper disks, method using holes into solid agar was used. Into these holes, precisely selected volume of the supernatant of bacterial isolates has been dosed. In total, 170 supernatants were tested against 80 strains of bacteria. In the case of supernatant containing antimicrobial substances produced by the tested bacteria, an inhibition zone around the hole would be created. The antimicrobial activity has been confirmed in three cases while testing supernatant of symbiotic bacteria Aeromonas veronii and Pseudomonas moraviensis against strains of clostridia. Furthermore, it has also been confirmed, that Aeromonas veronii inhibited the growth of Pseudomonas moraviensis. In the remaining studied isolates, antimicrobial activity suppressing the growth of test bacteria has not been confirmed.
Effect of prebiotics on survival of bifidobacteria in digestive tract

The gastrointestinal microbiota of livestock is a complex and diverse ecosystem, which strongly depends and correlates with animals health. One of the many ways, how to improve the composition of this microbiota in the gastrointestinal tract, is application of probiotics, prebiotics or an appropriate combination called as synbiotics. Using subtle interventions can be achieved health benefits, increase of some indicators livestock performance and improving the economic situation of farming. The aim of the thesis was to find out the effect of prebiotics on the currently applied probiotics and the composition of the intestinal microbiota. Three groups of calves were studied. In the first one the mixture of five strains of rifampicin resistant variants of Bifidobacterium (RRBif): Bifidobacterium animalis ssp. animalis (2 strains), B. choerinum, B. thermophilum and B. longum ssp. suis was fed to 2 days old calves in a form of fermented milk. In the second group probiotics were adminestered together with prebiotics mixture contains fructooligosaccharides and galactooligosaccharides everyday. The third group without synbiotic treatment was used as a control. The numerous of main bacterial groups in gastrointestinal tract were determined by cultivation. Our results showed that applied bacteria were able to survive the passage through the gastrointestinal tract of calves. Administered bacteria persist in counts more than 107 CFU/g in the group of calves, that were fed only by probiotics and in numbers 108 CFU/g in the group of calves, that were treated by synbiotics. Furthermore a slow decrease of bifidobacterial numbers to 104 CFU / g was observed in both groups. Significant difference (p = 0.05) between the count of RRBif in probiotic and synbiotic group were determined only two days after administration of that additive. Administration of probiotics and synbiotics to calves increased the count of naturally occurring bifidobacteria, but these differences were not significant.
Bifidobacteria of intestinal tract of exotic animals

The main topic of the bachelor thesis is description of occurence of bifidobacteria in the gastrointestinal tract of exotic animals and insect. Afterwards was described a role of probiotic bacteria and influence of nutrition on their occurence. The goal of the practical part of the bachelor thesis was genus-level identification of bifidobacteria in feces samples of exotic animals. After the bacteria cultivation on selective agars, isolates were collected and detection of the enzyme fructose-6-phosphate phosphoketolase were made. Bifidobacteria were found just in two samples from analyzed animals.
