The principle of participation and application this principle to the school youth hostel.

This Diploma thesis presents participation as an important part of the educational process of school youth hostel as a tool for empowering youth for active and responsible attitude to the life. First part deals with definitions and meanings of participation in school youth hostel environment. Next part examines participation in five school youth hostels in South Bohemia. The last part introduces data from questionnaire survey and the data are presented in the context of theoretical knowledge and practical examples.
Recombinant expression of rat NK cell receptor Clr-b in the presence of fluorinated analogues of monosaccharides

NK cells are part of innate immunity that, besides eliminating damaged cells, also produce chemokines and cytokines, which affect the cells of adaptive immunity. NK cells express activating and inhibitory receptors on their surface. The balance between them keeps the NK cells inactive. When the balance is disrupted, the cytotoxic mechanisms of the cell are activated. Receptors NKR-P1B and NKR-P1D are two rat NK cell inhibitory receptors whose ligand is protein Clr-b, a receptor belonging to the C-type lectin-like receptor family. This work aimed to recombinantly produce Clr-b in the presence of seven fluorinated analogues of monosaccharides as potential inhibitors of N-glycosylation. The protein was successfully expressed in the HEK293T cell line as a construct containing the extracellular part of Clr-b, the Fc fragment of human IgG, and a histidine tag multiple times, each time in the presence of one of the compounds. As glycosylation plays a major role in the functionality of many proteins, inhibition of glycosylation appears to be a promising way of treatment of different diseases such as cancer or multiple sclerosis. MUDr. JEHLIČKA SUMMARY This study has focused on a new method for the assessment of endothelial dysfunction (ED) using the Deceleration Index (DI). A total of 242 ultrasonographic examinations were performed in 92 children (FH 32, DM1 30, healthy controls 3O). The average DI values were 21.8%(±9.4)in healthy children, 19.9%(±9)in FH and 17.6%(±14)in the DM1 group. There were statistically insignificant differences between the study groups. This was most probably due to the high biological variability of the method employed. The coefficient of variation of DI was 1.02 (0.68 -1.53). Therefore, the clinical usefulness of DI is limited with regards to the individual assessment of ED and thus for the prediction of cardiovascular risk. The association of ADMA and oxLDL with ED and their superior sensitivity compared to DI and FMD has been confirmed in our study. Folic acid supplementation did not lead to a statistically signifiant improvement of DI and FMD. Similarly, plasma levels of ADMA and oxLDL remained unchanged despite a decrease of homocysteine and hsCRP in these children. This finding demonstrates a complicated relationship between ADMA, homocysteine and folic acid. The protective effect of folic acid on ED in high risk children was not confirmed in this study.

Introduction: Early childhood is a critical phase for shaping basic eating behaviors and food preferences that impact an individual long-term physical and mental condition. A little child takes over eating habits from his or her parents and their improvement later in life is difficult. Methods: An analysis of eating habits of Czech children aged 12-23 months. The data was obtained in a cross-sectional retrospective questionnaire study. The questionnaires were completed by children's mothers addressed in public places. Results: Almost a quarter (24.4%) of mothers still breast-fed a child after his or her first year of life and the toddlers were fed at average 4.3 times a day. The average age of child when his or her mother stopped its breast-feeding was 8.5 months. The most frequent reason of weaning the child was a paucity or loss of breast milk. More than third (38.5%) of children received the non-milk fluids before the end of the 6th month. And approximately one-third (27.8%) of children obtained the non-milk food before the end of the 6th month. Toddlers got the meals at average 5.3 times a day. Most (83.3%) of mothers had no difficulties with children's feeding. The most frequent eating problem was pickiness. Development of cardiovascular system function in patients with a functionally single ventricle. Abstract Total cavopulmonary connection (TCPC) is the only possible surgical palliative procedure in patients with a functionally single ventricle. It consists of the stepwise anastomosis of both caval veins with pulmonary arteries, creating a unique situation of so-called Fontan circulation, where the separation of the pulmonary and systemic circuit is traded for chronically elevated systemic venous pressure and impaired cardiac output increase during exercise. We aimed to evaluate long-term results and development of exercise capacity in the population of patients with TCPC in the Czech Republic, possible predictors of their survival and functional outcome, as well as analysis of permanent cardiac resynchronization as one of the new treatment options for dysfunction of the single ventricle. We present a retrospective study of all consecutive patients in the Czech Republic identified in the nationwide single-centre institutional database and matched with the National Death Registry and Cardiovascular Intervention Registries. Quote Attribution and Character Networks in Novels

This thesis focuses on extracting information from literary works using tools for language analysis. Our goal is to automatically extract a conversational network of the characters in a novel. We divide the work into three subproblems and solve them separately: Character Extraction, Quote Attribution and Network Creation. The result is an end-to-end tool that gets a text of a novel in English and outputs a visual representation of the character network. Our work is based on existing literature. Zuzana Urbanová The Function of Direct Forms of Presentation in the Generic Structure of Newspaper Reports Abstract The thesis deals with the employment of direct forms of presentation in the genre of hard news. The texts for the analysis were excerpted from the main British broadsheet newspapers. The classification of forms of presentation is based on Semino and Short (2004) and focuses on their deictic and syntactic properties, interpreted in terms of the pragmatic concepts of perspective, faithfulness claims and the role of the reported and reporting speaker. Attention is paid to various direct forms, including direct speech, free direct speech and combined forms, i.e. non-direct forms appearing with a partial direct quote. The thesis draws heavily on the work by White (1998), whose approach to genre is informed by the ideas proposed by the Sydney School and Systemic Functional Linguistics. Hard news is characterised by the orbital generic structure, consisting of the nucleus and a number of specifying satellites (White 1998). Searching for and Evaluating the Severity of Endothelial Dysfunction in Children with Chronic Autoimmune Disease

We aimed to evaluate the endothelial function by combining RHI measurements and specific biochemical markers in the children with possible risk of premature manifestation of atherosclerosis and in the control group of healthy children. In all, 124 children (of which 106 patients divided into five groups according to diagnosis - type 1 diabetes mellitus, Crohn's disease, cystic fibrosis, familial hypercholesterolemia and acute lymphoblastic leukemia and 18 healthy controls) were enrolled in the study. During the study, we measured RHI using a new plethysmographic method and further evaluated biochemical markers of endothelial dysfunction (ADMA, E-selectin, hsCRP and VCAM) and lipidogram in individual groups of children. The primary objective of our study was the determination of RHI and biochemical parameters in healthy subjects and in selected risk groups of children (type 1 diabetes mellitus, Crohn's disease, cystic fibrosis, familial hypercholesterolemia and children after successful treatment of acute lymphoblastic leukemia). Self-reflection of a student, who study leisure studies

This Bachelor thesis examines the method of self-reflection in the reflective practise as a tool for improving the professional and personal skills of students of leisure studies. First part deals with definitions and meanings of self-reflection and connection with process of reflection and the improvement of the reflective thinking. Next part examines also the charakteristics which a teacher of leisure studies should have and what education he must have to be able to work in the state organizations. The last part introduce practise in high school education and examies where is possible to impriove self-reflection skill.
