Support of the development of tourism potential in the Olomouc Region

The object of my thesis is to analyse projects supporting tourism from the SROP resources in the Olomouc region, mostly their impact on the development of the region. In the first part of the work I analyse preconditions for the development of tourism in the Olomouc region. At the end of the first part the tourism is assessed, there are expressed proposals to increase the efficiency of its management in the future. The main part of my thesis is based on qualitative research focus on lokalization of the projects, the real impacts of the projects to develop the region and tourism in Olomouc region. The role of human mitochondrial YME1L protease in the biogenesis of the oxidative phosphorylation system

Mitochondria are found in virtually all eukaryotic cells where their main function is the production of ATP in the oxidative phosphorylation system (OxPhos). OxPhos is build-up of both nuclear and mitochondrial encoded protein subunits. Due to the potential function threatening defects, the quality of these protein subunits is constantly under tight control by specialized proteins. The recognition and selective removal of defective mitochondrial proteins is carried out by specific mitochondrial ATP-dependent proteases. So far, four such proteolytic complexes active within distinct mitochondrial subcompartments were identified. Both i- and m-AAA protease complexes are found in the inner mitochondrial membrane. Whereas the i-AAA protease is active in the intermembrane space, the homologous m-AAA protease functions on the matrix side of the inner membrane. The aim of the present work was to characterize cellular function of the human orthologue YME1L of the yeast i-AAA protease subunit YME1 using human HEK293 cell model. We found that human YME1L is an integral membrane protein with molecular weight of approx. 600-1100 kDa, exposing the carboxy-terminal domain to intermembrane space. Functional characterization of LACE1 APTase and mitochondrial AAA proteases YME1L and AFG3L2 in mitochondrial protein homeostasis.

Mitochondrial protein homeostasis is crucial for cellular function and integrity. It is ensured by many specific mitochondrial proteases with possible chaperone functions located across the various mitochondrial subcompartments. In the first part, we have focused on characterization of functional overlap and cooperativity of proteolytic subunits AFG3L2 and YME1L of the mitochondrial inner membrane complexes m- and i-AAA in HEK293 cells. The double AFG3L2/YME1L knockdown cells showed severe alteration in OPA1 protein processing, marked elevation in OMA1 protease and severe reduction in SPG7. Our results reveal cooperative and partly redundant involvement of AFG3L2 and YME1L in the maintenance of mitochondrial protein homeostasis and further emphasize their importance for mitochondrial and cellular function and integrity. The aim of the second part was to characterize the cellular function of LACE1 (lactation elevated 1) in mitochondrial protein homeostasis. LACE1 protein is a human homologue of yeast Afg1 (ATPase family gene 1) ATPase. The Economics of Happiness: Are the busier employees the happier ones?

The thesis analyzes the relationship of the perceived happiness level and the level of work busyness of full time employees. For this purpose, a questionnaire was built which mapped happiness and busyness with considering both subjective and objective variables. 205 employees participated in the questionnaire survey. The sample showed above-average education, a high level of busyness and above-average salary (or wages). It has been shown that the work happiness level depends on the level of workload: the unbusy employees are the least happy ones. Influence of ambitions or influence of the importance of work area to the relationship of happiness level and workload level has not been confirmed.
Migrační rozhodování

Tato práce se zabývá migračním rozhodováním jedince. Pomocí ekonomické teorie vysvětluje migrační chování člověka. Ten musí zhodnotit veliké množství proměnných, které do jeho rozhodování vstupují, a rozhodnout, zda je pro něj optimální migrovat či zůstat v rodné zemi. Protože na rozhodování člověka může mít vliv i státní migrační politika, je v práci teoreticky popsané migrační chování zúčastněných států. Takto vyložený teoretický rámec je v druhé části aplikován na reálný svět, kde je nejprve sestavena reálná kalkulace individuálního migračního rozhodování. Následně je na základě empirických zjištění na příkladu České republiky demonstrováno, kteří lidé se nakonec rozhodnou do země imigrovat a z jakého důvodu se tak rozhodnou. Nakonec bude tato práce opět na příkladu České republiky ilustrovat konkrétní státní imigrační politiku a zhodnotí, zda je vůči imigrantům spíše otevřená nebo jejich příchod blokuje.
