All my beginning identities

1 ABSTRACT In which situations have I found myself as an aspiring and later as a beginning teacher? How do I experience my affiliation with this profession today? What roles shape me as an educator? How do they influence my views on education? How do they impact my professional identity? The beginning of a professional career is a transitory and transformative period, crucial for further development, and builds upon various past experiences that date back to childhood. The theoretical part of this study navigates between social science, pedagogical, and artistic discourse. It draws on contemporary art that offers an approach to questions of identity and subjectivity, exemplified by works from authors such as Margit Titlová, Rineke Dijkstra, Marlene Dumas, Cindy Sherman, among others. The research section maps, through the analysis of autobiographical records, various situations and roles I encountered at the beginning of my career, as well as during the period leading up to this career. The autobiographical research oscillates between an artistic approach and an approach based on qualitative research, emphasizing the tension between the internal and external dimensions of identity. Refractometric study of very low density lipoprotein (LDL)

Knowledge of the chemical composition of lipoproteins has a broad meaning. In determining lipoprotein particle size fractions is necessary to preliminary separation by ultracentrifugation in density solutions. Usually the determination is performed by electrophoretic or gel filtration with a suitable detection, which distorts the results. A better solution offers a new detector, called Light Scattering Detector, which detects the bend of laser beam on molecules or particles in solution. To calculate the required particle size dependence on the concentration of the refractive index dn / dc. Concentration is determined gravimetrically and refractive indices by refractometric method. Dependence suits polynomial function. If it correlates with the chemical composition of VLDL lipoprotein can be determined by studying a larger number of samples.
Lipoprotein study by SEC/LLSD method

The disorders of the lipid metabolism have to be monitored for a number of socially significant diseases. For the diagnosis of these disorders, it turned out in the last 10 years as very beneficial to determine the particle size of the individual lipoprotein classes, in particular the low-density lipoprotein. Among several known methods, we have chosen gel filtration with the light scattering detector. Lipoprotein fractions were separated by ultracentrifugation from the plasma of four donors. Three biochemical parameters - total cholesterol, triacylglycerol, and total protein - were routinely determined for plasma and for each fraction. The silica column BioBasic SEC 300x7,8 mm was calibrated by the proteins thyroglobulin, ovalbumin and angiotensin II with UV detection (280 nm). These proteins are not suitable (due to the size) for the calibration method SEC-LLSD. Therefore, latex beads of various modifications and sizes were tested. The only usable measurement was obtained for the deep-blue latex beads of the size 55 nm, the others remained retained in the column. The lipoprotein fractions of VLDL and LDL were examined by the SEC-LLSD method at different pH (7.3, 7.5, 7.7, 8.0) and ionic strength (0.1M phosphate buffer, 0.1M phosphate buffer and 0.9% NaCl, 0.05M phosphate buffer and 0.9% NaCl)....
Determination of niacin and its metabolites by HPLC-MS

Nicotinic acid (NA) is a hypolipidemic agent with pleiotropic effects on plasma lipoproteins. Its medicamentous form with delayed secretion leads to pyrimidine metabolites, which make increased risk of hepatotoxicity and should be monitored. The aim of the presented study was to introduce HPLC-MS method for the determination of NA, nicotinamide (NAM), nicotinuric acid (NUA), 1-methyl-nicotiamide (MNA), nicotinamid-N-oxide (NNO), 1-methyl-2-pyridon-5-carboxamide (2-Pyr) and 1-methyl- 4-pyridon-5-carboxamide (4-Pyr) in blood plasma useful for the monitoring of hypolipidemic therapy. We have compared calibration dependences of individual metabolites using two columns - Hypercarb (grapfite carbon) and Hypersil Silica (silicagel). Both columns revealed linear calibration dependences for all mentioned analytes except MNA in the concentration range 10-2000 ng/ml for chemical standards; calibration dependence in human blood plasma measured with the column Hypersil Silica was linear in the range 20-4000 ng/ml for all tested analytes. Correlation coefficient varied between the value 0,9400 and 0,9997. Analysis time was 15 min with the column Hypercarb and 27 min with Hypersil Silica. Determination of non-esterified fatty acids in plasma

The determination of non-esterified fatty acids (NEFA) in plasma is possible by many routine biochemical methods. For more detailed metabolic studies it is required to analyse the fatty acid (FA) profile. The NEFA profile determination is usually performed by the capillary gas chromatography (GC), but the sample preparation is not uniform. NEFA may be isolated by the extraction process or by the preparative thin-layer chromatography (TLC). The aim of this study was to compare these two separation procedures. The samples of pooled plasma from volunteer donors were analyzed by capillary GC after previous separation procedure by the TLC and the liquid-liquid extraction. The results were compared by the t-test for both the absolute concentration of individual FA provided by the internal standard (margaric acid) method and the relative abundance (Rel%). The reproducibility of the results was significantly better for the liquid-liquid extraction method than the TLC. The relative standard deviations (RSD) of the FA groups (unsaturated, monosaturated and polysaturated) were from 4.3 to 11.3 % vs. 8.6 to 33.8 % for the relative abundance and from 16.5 to 25.5% vs. 15.4 to 47.4% for the absolute concentration. Influence of methods RWCT on improvement of pupil's comunication skills

The thesis is focused on the programme Reading and Writing for Critical Thinking (RWCT) and its influence on the pupil's communication skills. The theoretical part provides factual information relating to the topic. It describes communication, communication skills, the status of communication in the Framework Education Programme for Primary Education and summarizes the information about the programme RWCT, its objectives, content and methods. The practical part deals with the influence of methods RWCT on improvment of pupil's communicative skills. It compares lessons at two elementary schools in terms of time, which is spended with pupil's communication, at which they have to express their own thoughts and opinions.
Advanced lipidomics in selected clinical conditions

Abnormalities of lipid metabolism are considered risk factors for cardiovascular, metabolic, nephrologic diseases amd some malignancies, as well. Nowadays, a lot of effort is devoted to study new risk factors and surrogate markers of conditions mentioned above to improve their prognosis and decrease mortality. The aim of this thesis was to provide a comprehensive survey of lipid metabolism, characteristics of different lipid compounds in health and diseases and of possibilities of utilization of selected lipid parameters in the diagnostics of pathological conditions listed above. Selected lioid parameters were observed in several studies, focused on specific pathological conditions. Besides conventional lipid analytes, the composition of fatty acids in plasma lipid pools was studied in healthy controls, and in the patients suffering from metabolic syndrome, chronic pancreatitis, and pancreatic cancer, as well. Markers of an oxidative stress (oxidatively modified LDL particles and conjugated dienes in precipitated LDL) were assessed in healthy controls, patients with metabolic syndrome, chronic pancreatitis, pancreatic cancer, and in the patients with different concentrations of plasma apoB-48, too.
