Study of the Role of Casein Kinase I epsilon in Breast Cancer Using Drosophila as a Model
MATULOVÁ, Jana
A particular allele of the Drosophila discs overgrown locus called dco3 leads to hyperplastic overgrowth of imaginal wing discs. Since high rate of mutations in human homologue of the dco gene coding Casein kinase I epsilon was found in breast cancer tissue samples, I attempted to recreate the observed mutations in the Drosophila genome.
Influence of posttranslation modifications on protein structure: development of methodology for isotopic labeling of proteins in \kur{Leishmania tarentolae} expression system
MATULOVÁ, Jana
We have established a cost efficient approach for isotopic labeling of proteins using Leishmania tarentolae expression system. The method is based on cultivation L. tarentolae in a rich complex (BHI) medium supplemented with labeled amino acid(s). This approach leads to low level isotope enrichment, which implies the decreased sensitivity of NMR experiment. In the same time this approach leads to a two to three fold increase of the yield of the recombinant protein. Importantly, it leads to 5 {--} 10 fold reduction of the overall costs as compared to the recently established protocol (Niculae et al., 2006). In parallel, we have attempted to estimate the suitability of L. taretolae for in-cell NMR spectroscopy.
