Influence of the temperature on hydrophobic domain quality in a biopolymer-surfactant system.

The temperature dependence on polarity has been investigated by fluorescence spectroscopy with pyrene as a probe. Pyrene was chosen thanks to its unique sensitivity to polarity of the medium. As a biopolymer was chosen 73 kDa and 1,4 MDa hyaluronic acid. Emission spectra of pyrene were measured as a function of temperature in aqous solution of CTAB in a presence and absence of sodium chloride. The results show, that the addition of specific concentration of hyaluronic acid to the sample with CTAB, mostly doesn´t have effect to change increasing/decreasing slope of polarity with the temperature.
Study on Sol-gel Process of Agarose by Classical Rheology and Dynamic Light Scattering

The main aim of the bachelor thesis was characterization of hydrogels from rheological point of view and by dynamic light scattering. Rheological part of research relates with the determination of the influence of temperature on mechanical properties, gelation temperature and temperature of solidification as well as the effect of aging on mechanical properties of hydrogel. The influence of the particle size on diffusion coefficient was investigated by scattering techniques. The positive correlation between the data obtained by both types of measurement and the potencial usage of dynamic light scattering method for the characterization of hydrogels was discussed as well.
Microrheology in study of biopolmer colloids.

A new method for determining the viscoelastic properties of materials was introduced and investigated. Results of three groups of samples obtained using one particle microrheology method, DLS microrheology method and classical rheology method were compared to be sure of correctness of measurements. As a model system were chosen mixtures of glycerol of different viscosities. In case of samples containing glycerol, results were also compared with tabulated values. Hyaluronan of various molecular weights was used as a biopolymer and polystyrene particles were used for microrheology. It was confirmed, that viscosity values of biopolymer samples obtaining by each method are comparable.
DLS microrheology to study gelation of polymer

This bachelor thesis deals with the study of gelation process on model polymer through DLS microrheology. As the model polymer was used agarose and for microrheological measurement were used polystyrene particles. Agarose samples of various mass concentrations were studied through dynamic light scattering. Measured data were used to create graphs of MSD against time which were compared to MSD curves and error bars. The values of diffusion coefficients for all samples and each temperature were also compared.
Microrheology of Biocolloids

The main aim of the doctoral thesis was study of passive microrheological techniques as advanced methods for characterisation of viscoelastic properties of soft material. These techniques are able to provide certain advantaged or additional information on classical rheology. Several techniques from the wide range of passive microrheological group such as one particle video – microrhelogy with IDL data procesing, microrheology based on light scattering – DLS or FCS microrheology have been applied in practice during my PhD studies. New Matlab script has been also invented to simply obtain information about viscosity of low volume samples. Aplicability of techniques were studied on several various bicolloids such as solutions or gels of hyaluronic acid, on study of properties at the interfaces or on gelation proces studies of agarose. Obtained data were compared classical rheology results. Suitability of each technique for investigated systems or appropriate reccomendations for further measurements were didcussed based on obtained data. Main advantages and limites of passive microrheology techniques were also described in comparison wih classical rheology method.
Nanocarriers based on hydrophobized biopolymer

This work studies the properties of hydrophobically modified biopolymers for potential use in targeted drug delivery. Two samples of hydrophobically modified hyaluronan were selected. The critical micelle concentration (CMC) of these samples was measured using fluorescence spectroscopy, wherein it was found that micelle formation occurs over a wide concentration region. Environment of 0,15 M NaCl causes the decrease of CMC only slightly or not at all. Using the method of dynamic light scattering, particle size was measured. One of the samples exhibited a double size distribution. DLS results are consistent with the results of fluorescence spectroscopy at issue in aggregates forming near the CMC point. Zeta potential measurements provide information about stability of the samples. The results of this study confirm that one of the studied samples seems to be a suitable candidate for the post of drug carrier for targeted delivery.
Influence of particle properties on microrheological measurements of biopolymer solutions

This bachelor‘s thesis is focused on the evaluation of particle influence on microrheological measurements of biopolymer solutions. Hyaluronic acid was the biopolymer and glycerol solutions of different concentration were chosen as model system. Polystyrene particles of different diameters (0,5 m, 1 m a 2 m) were used for the measurements. Results of passive microrheology were compared with classic rheology and with tabulated values in case of glycerol.
