Characterization of germline alterations affecting genes influencing development and prognosis of specific adult cancers
Jelínková, Sandra ; Kleibl, Zdeněk (advisor) ; Hlaváč, Viktor (referee) ; Hájková, Nikola (referee)
In my dissertation, I studied the genetic predisposition of selected types of cancer that have not been systematically studied in the Czech Republic. We used next-generation panel sequencing to identify germline pathogenic variants. Analysis of 1333 patients with ovarian cancer, 527 patients with endometrial cancer, and 334 patients with hepatocellular carcinoma included sequencing using the CZECANCA panel. A specific CZMELAC panel was prepared for the analysis of 264 melanoma patients. We focused on the identification of pathogenic variants in known predisposition genes. We also evaluated candidate genes and phenotypic characteristics in carriers of pathogenic variants. Analysis of high-risk melanoma patients revealed pathogenic variants in melanoma associated genes in 9/264 (3.4%) patients, and an additional 22 (8.3%) patients carried a pathogenic variant in one of the other predisposition genes. The odds of carrying a pathogenic variant were increased in probands with multiple melanomas and in the presence of melanoma in relatives. The incidence of germline pathogenic variants was highest in ovarian cancer, where pathogenic variants were found in 427/1332 (32.0%) patients, with a predominance of mutations in BRCA1, BRCA2, followed by alterations in other ovarian predisposition genes. Hájková, Nela ; Pekara, Jaroslav (advisor) ; Maláska, Jan (referee)
The Abstract The diploma thesis is a review study in the form of scoping review which deals with non- pharmacological interventions to prevent and manage delirium. The diploma thesis is divided into two parts: theoretical and practical. The theoretical part approaches the issue of delirium and provides an overview of the latest findings on this issue. A search of professional literature was made within the practical part or also known as methodology, according to the recommendations of the Prisma methodological procedure. The included studies also dealt with non-pharmacological interventions to prevent and manage delirium. The search strategy for professional publications was limited to a range of 10 years (2012-2022) and also to Czech, Slovak and English languages and was searched using specific keywords. This definition led to the finding of 8776 professional publications which were then reduced to a final number of 40 studies that met the inclusive criteria. The included articles were analyzed in detail and gathered using codes into individual topics like patient reorientation, sleep care, environmental adjustment, early mobilization, family involvement, analgesia and sedation or package implementation and multifactorial approach and education. The effect of individual interventions against delirium was...

Hájková Paterová, Natálie ; Altová, Blanka (advisor) ; Hájek, Václav (referee)
This thesis is concerned with Jaroslav Seifert's relationship to visual arts and artists, for it is apparent in both his writings and his biography that this relationship was not only deep, but it also served as a powerful source of inspiration for his work. This study focuses on the works of Jan Zrzavý, Toyen and Josef Sudek, since Seifert considered them extraordinary and maintained a lifelong friendship with them. However, I also consider other artists and works that Jaroslav Seifert related to in his writings. The selected topics are followed throughout the poet's life and work, and also set in the context of the given time period and shared culture. This study brings answers to the question of the nature of Seifert's relationship to visual arts and how it is reflected in his writing, and also aims to describe the way the dialogue between Seifert's texts and arts is developed based on comparison of specific visual and literary imagination. Key words: Jaroslav Seifert, Jan Zrzavý, Toyen, Josef Sudek, friendship, dialogue, visual arts, fine arts, photography, poetry, journalism, All the Beauties of the World, imagination, Devětsil, Czech culture of the 20th century.
