Getting old at home - getting old in a town.
Dušková, Eva ; Lejsal, Matěj (advisor) ; Bareš, Pavel (referee)
This thesis discusses dignity of aging and elderly citizens and the ways in which it is supported within a village community. Dignity is seen as the most important and universal human need, and not only for elderly people. Its significance and protection is anchored in international and national legal documents which are also binding at lower levels of state administration including the community and municipality level. Municipalities are public institutions obliged to create social policies. Citizens need to be involved and to participate in constructing these policies that touch upon the needs of specific groups of them, including that of the elderly. Using several methods, the analytical part describes the situation in a particular village community. The method of social-demographic analysis is based on statistical data and literature. Participant observation and close study of documents provide additional information to the analysis of secondary data. The results show up-to-date age structure and the present state of coexistence of different people in the community. Judging from ways of participation in political life of the community we can infer that close relationships among citizens are the driving force of the development of the area as far as meeting the needs of the elderly are...
Regulation of alternative splicing
Dušková, Eva ; Staněk, David (advisor) ; Trejbalová, Kateřina (referee)
Alternative splicing is an important cellular mechanism. It allows to produce multiple protein isoforms from a limited number of genes. Regulation of alternative splicing involves cis-acting elements on pre-mRNA and trans-acting splicing factors (SR and hnRNP proteins). Because splicing occurs co-transcriptionaly, chromatin structure appears to have a role in the regulation of alternative splicing. We have studied the effect of histone acetylation on alternative splicing. We have prepared splicing reporter for alternative EDB exon, which is part of the fibronectin gene. We have shown, that the inhibition of histone deacetylases affects splicing pattern of EDB exon from the reporter in the same way as the splicing of the endogenous EDB exon. Furthermore, we have shown, that the structure of the promoter affects splicing of alternative EDB exon from splicing reporter. Currently we have found out, that the structure of the promoter influences the degree of histone H4 acetylation. Inclusion of alternative EDB exon in mRNA was inversely proportional to histon acetylation on the reporter. This work might explain why various promoters have different splicing patterns of alternative exons.

The Case Study: The Reflection of the Civic Sector Organizations in Chomutovské noviny between 2006-2008
Dušková, Eva ; Dohnalová, Marie (advisor) ; Haken, Roman (referee)
The communication with the public is really important for the existence and work of the civil sector organizations. The reason is that the public includes possible users of products and services, backers, donators and associates. One of the possible channels how to communicate and keep up with the public is promotion and presentation in the media. Chomutovské noviny is a printed matter that is run by the local administrative body. This type of media should give the opportunity to as many local organizations as possible. The study is aimed at selected civil sector organizations within one particular period. The results of this study show that the presentation of the civil sector organizations was not too numerous in Chomutovské noviny. The possible reasons arise from the interviews.

Addiction issue and development of child's personality
Dušková, Eva ; Koťa, Jaroslav (referee) ; Krykorková, Hana (advisor)
This bachelor work deals with addiction and the possibilities of its prevention (the possibilities of preventing its origination). In the introductory chapter, the terms habit and addiction are defined, addiction characteristics are listed, and the types of addiction are specified according to its possible appearance with children and teenagers. The next chapter presents the biological, sociological and psychological addiction-start hypotheses. Then the reasons of abuse of addictive substances by children and teenagers are given, as well as some special kinds of pathological behaviour. Prevention of addiction, especially in school and family environment, is elaborated on in the following chapter which covers also the organization of primary prevention in the Czech Republic, the prevention programmes bias and the responsibilities of prevention methodologists in schools. At the family level, important prevention rules are presented. The final sections are focused on the ways leading to acquiring sound confidence, mental resistance and responsibility for one's own health.
