Broadcast coverage of the coronavirus epidemic 2020 as a possible world
Bergerová, Michaela ; Jirák, Jan (advisor) ; Křeček, Jan (referee)
This diploma thesis explores the theory of possible worlds and its relation to broadcast during the coronavirus pandemic. It perceives news, especially broadcast, as a possible world. Using narrative analysis, this thesis describes the characteristics of two possible worlds that arose during the first wave of the coronavirus pandemic on Czech Television and television Prima. The main goal of this research is to examine what kind of possible worlds have these two TV stations constructed in their main news programs and to point out how these two possible worlds differed. My diploma thesis should primarily contribute to a clear comparison of how the depiction of the coronavirus pandemic differed by individual television channels in the first half of 2020.
Determination of indican on carbon paste electrode and on carbon composite electrode
Bergerová, Michaela ; Dejmková, Hana (advisor) ; Schwarzová, Karolina (referee)
This thesis is focused on the application of two working electrodes of graphite suitable for determination of oxidizable tumour biomarkers. It deals with the study of electrochemical behavior of urinary indican. For the determination the technique of differential pulse voltammetry (DPV) was used on carbon paste electrode and film composite paste electrode in Britton-Robinson (BR) buffer. In the first step optimal conditions were determined to find the lowest limit of determination (LQ) of this substance. BR buffer of pH 3 was chosen as the optimum medium for determination on a carbon paste electrode and BR buffer of pH 2 was used as the optimum medium for the determinaton using a film composite electrode. Performance for both electrodes was observed in a concentration range of from 1 · 10-6 to 5 · 10-5 mol·l-1 . The limit of determination for measurements on a carbon paste electrode was 0.7 µmol·l-1 and the film composite electrode was 1.7 µmol·l-1 . Possibility of the acummulation of indican was examined in order to increase sensitivity and decrease of limit of quantification. However, after 5 minute sorption, no increase in response was observed and the accumulation step was not inserted. Developed method was tested on the determination of urinary indican in human urine matrix after solid phase...
Voltammetric determination of 2-amino-3-nitrotoluene on silver solid amalgam electrode
Bergerová, Michaela ; Fischer, Jan (advisor) ; Vyskočil, Vlastimil (referee)
This thesis is focused on development of electrochemical method for determination of 2-amino-3-nitrotoluene. The technique of DC voltammetry (DCV) and differential pulse voltammetry (DPV) were used on a silver solid amalgam electrode modified by mercury meniscus (m-AgSAE) with the Britton-Robinson (BR) buffer supporting electrolyte. In the next step the suitability of developed method was tested for drinking and river water model samples. BR buffer of pH 12.0 was used for determination of 2-amino-3-nitrotoluene by DCV method and BR buffer of pH 7.0 was used for DPV method. Both methods were used for concentration monitoring of studied compounds in the range from 1·10-6 to 1·10-4 mol·l-1 . Limit of quantification of DCV method in BR buffer with deionized water was 2,7·10- 6 mol·l-1 whereas 1,2·10-6 mol·l-1 in drinking water and 1,5·10-6 mol·l-1 in river water. Limit of quantification of DPV method was 1,5·10-6 mol·l-1 in BR buffer, 7,3·10-7 mol·l-1 in drinking water and 1,4·10-6 mol·l-1 in river water. Stability of stock solution of the studied compound of 1·10-3 mol·l-1 concentration in water was measured by spectrometry at 3 wavelengths 222 nm, 293 nm, 416 nm. After one month there was no significant decrease in absorbance so the sample was considered to be stable and it was kept in the dark at...
