Kinetics of hydrophobic organic compounds uptake in the earthworm Eisenia andrei in soils with different content of organic carbon

This thesis is concerned with the fate of hydrophobic organic pollutants (HOCs) in soil. The theoretical part summarizes the current knowledge about bioavailability, factors influencing the bioavailability and methods for its measurement. Attention is paid to the organic carbon content, which is considered to be one of the key factors and to the influence of microorganisms that can degrade or interact with the substances, thus changing their bioavailable ratio. The practical part of the thesis uses bioaccumulation test with earthworm Eisenia andrei for evaluating the uptake kinetics HOCs in three soils with different organic carbon content. Soils were laboratory contaminated with mixtures of selected HOCs (phenanthrene, pyrene, lindane, p,p'-DDT and PCB 153) and the concentrations of substances in these soils were followed for 217 days. Subsequently, bioaccumulation test was conducted; earthworms were exposed from one to twenty-one days. The collected data were used to model uptake kinetics curves for individual substances. Peak-shaped bioaccumulation were found for pyrene, for lindane a steady-state concentration during exposure was observed, whereas for p,p'-DDT and PCB 153 growing concentrations after twenty days of exposure were detected. The reason for the different behavior of the substances was probably their diverse sorption on the organic material, which together with the different speed of elimination influenced final concentration in earthworms. Phenanthrene was not detected in some samples and therefore the modeling of uptake kinetics was not possible.

This diploma thesis deals with the topic of the notice of termination of employment in the Czech labour law. The thesis sets out multiple primary goals. The first goal is to introduce the reader to the context of the legal framework of the notice of termination of employment, including an analysis of selected case law relevant to the topic. The second goal is to provide a comparative outline via the exploration of Belgian labour law and its legal regulation of the notice of termination of employment. Thirdly, the author reflects the findings gathered throughout the thesis and offers some amendments de lege ferenda. The diploma thesis is divided into eleven parts. The first three parts relate to the general description of the matter - the first is devoted to the labour law in the context of the legal system of the Czech Republic, the second is devoted to specific characteristics of an employment relationship and its subject and object. The third part discusses the termination of employment in general and it also takes into account the strategy of flexicurity. The fourth part aims at enlightening the effect of the international law and the law of the European Union on the legal regulation of the notice of termination of employment. Analysis and mapping of binding sites of gene expression regulators in the genus of Streptomyces.

Streptomyces are medically important soil-living bacteria that undergo morphological changes from spores to aerial hyphae and are important producers of bioactive compounds including antibiotics. Their gene expression is tightly regulated at the early level of transcription and translation. In the transcriptional control, sigma factors play a central role; the model organism Streptomyces coelicolor possesses astonishing 65 sigma factors. The expression of sigma factors themselves is controlled on the post-transcriptional level through the action of sRNAs that modify their mRNA level. However, only several sigma factors in Streptomyces have known regulons and also their sRNAs-mediated regulation has not been studied so far. According to previously measured gene expression data, we selected several highly expressed sigma factors. Using mutant strains with HA-tagged sigma factors, regulons of two important sigma factors, SigQ and HrdB, were analyzed by ChIP-seq procedure. Other sigma factors were further studied to see if they possess asRNAs, using 5' and 3' RACE method and northern blotting. This thesis first introduces the theoretical background of host care, social and legal protection of children, and theories about the identity of the child. The initial chapters will deal with the principles of social and legal protection of children and characterize the available forms of family care. The third chapter is devoted to host care itself; defines it and its purpose in detail describes the so-called "hard-to-place" groups, for whom foster care is especially appropriate. Reference is made to the non-profit organizations that provide foster care and the process of service delivery is described. This research investigates the experiences and opinions of people who grew up in children's home. Analysis of discussions and conversations brought out some unexpected findings. We obtain views on the possible overuse of host care and also gain an understanding of the lives of children living in children's homes.
