Rainfall-runoff regime in the headwater part of the Bystřice River basin

Rainfall-runoff regime in the headwater part of the Bystřice River Abstract This bachelor thesis deals with the rainfall-runoff regime and analysis of extreme situations in the headwater part of the Bystřice River basin. At first, the area of interest is described in physical and geographical terms and the influences of individual factors on the runoff are mentioned. The text content is documented by maps. The characteristic of rainfall-runoff regime is evaluated by means of runoff variability parameters. The variability is assessed by means of collected data using charts illustrating exceedance probability of average daily discharges in years 2009 and 2010. The flow variations are evaluated on the basis of decile deviations. The distribution of monthly discharges in the Bystřice River is displayed and the monthly flow variability is described in pie graphs separately for hydrological years 2009 and 2010. The monthly balance of the runoff is also evaluated by means of statistical measures of Kr and Cv coefficients. Finally, the runoff regime evaluation is completed with exceedance probability calculation of the mean annual discharge. Eva Novotná, Ph.D. Title of diploma thesis: Cloning, expression and purification of mycobacterial dihydrofolate reductase Dihydrofolate reductase is an enzyme essential for the metabolism of folic acid - it catalyzes the reduction of dihydrofolate to tetrahydrofolate. Tetrahydrofolate is an important cofactor involved in one-cabron transfer reactions. Dihydrofolate reductase plays a key role in the synthesis of DNA, RNA and proteins. Dihydrofolate reductase was also found in M. tuberculosis. This bacterium is the most common causative agent of tuberculosis in humans. Thus dihydrofolate reductase could be a potential target for the design of new antituberculotics. The recombinant protein dihydrofolate reductase was prepared in several steps. The coding sequence of the protein was first amplified by polymerase chain reaction. A recombinant plasmid, obtained by the ligation of an amplified segment of DNA with plasmid pET-28b(+), was transformed into competent cells E. coli strain BH101 by the heat shock method. Title: Application of the Snowmelt Runoff Model for snow accumulation and snowmelt modelling in experimental catchments Bystřice and Zlatý Brook in the Krušné Mountains Modelling of hydrological processes is a dynamically developing part of hydrology. The Snowmelt Runoff Model (SRM) was applied for modelling the runoff in two experimental catchments Bystřice and Zlatý Brook. The aim of this thesis is to set up and calibrate SRM model and to evaluate methods and procedures used for runoff simulations. The SRM model was used for modelling snow accumulation and snowmelt in two selected catchments in the Krušné Mountains. The snow depths and snow water equivalents are measured since 2009 at selected locations situated in catchments. Calibration and validation of the model was based on continual time series of precipitation, air temperature and discharge measured 2009. Hydrological years 2009 and 2010 were used for model calibration and hydrological years 2011 and 2012 were used for model validation. Sensitivity analysis, which quantifies the effect of individual model parameters on the simulating proces, was carried out based on results.
