The use watercourses and waterbodies within an urbanised countryside area

Water is an integral part of our life and waterways and areas were and still are an essential part of the public space of our cities and municipalities. We can find public space in every seat and we meet with the water on many of them. The water in public spaces appears in a different form. Whether these are minor elements, such as fountains, drinking water features, or larger water flows and water area. The water in the public space has its indispensable role. It is well known that its presence has a great impact on the well-being of man. Despite the fact that water is really important in the Interior of human settlements, over the years, water has disappeared on many places from public spaces, due to the technical development and development of settlements. The development of human settlements and public spaces takes place constantly from the time they were established. In recent years, the viewpoint of their importance is significantly changing and lot of the seats are aware of their value. The public space is like a living room of the city, thus a representative place. That´s why, the reconstructions of public space are quite often in recent years with use of water elements. The phenomenon of water surfaces is a factor which affecting the sustainable development and quality of life. Public space is not only an important functional element in the big cities. It has a great importance for smaller municipalities too. And just on them the thesis is focused. Their public space is often neglected. As well as water features in the surrounding area, which are uncared-for or extinct. Water in the form of water courses or water bodies have a great potential, in which a lot of possibilities is hidden, with which you can work in the urbanized areas. The goal of this thesis is to find a new role for the watercourses and water areas to be integrated into public space and public space, thanks to this, become a representative and functional part of the village once again.
Multipurpose Town House on Křížová Street, Brno

The aim of the thesis was to design the urban town house in blank space on Krizova street. The blank space is situated on the edge of the Mendel's square, an important transportation node of the city Brno. Spatial and conceptual solutions of the house is based on urban studies of Mendlel's square and its surroundings. The house is designed on a long parcel, which in the urban environment is equivalent to about four houses. Therefore, the house is divided vertically to keep the urban scale. At the same time at both ends it is directly connected to the existing buildings, therefore, different height levels of each mass are designed. Parterre of the house is commercial, as is the same for neighboring houses, in the other floors modern large single deck and maisonettes are designed.

This work includes an elaboration of the project documentation on a small family house with minimum acquisition costs for a family of four.The project documentation (the covering and technical report, situation, digs, foundations, section, ground-floor plan, ground-roof plan, views,{\dots}..) is worked out in the extent for the publication of the building licence. Within this work there is solved a choice of the proper locality, wheres at the disposal the real town-planning documentation. The object (a bungalow, cellarless, 3+kk house) is situated in a new housing project in the village of Hoštice which is 3 km for away from the town Volyně and c. 10 km from the district town of Strakonice. The next procedure of work is an elaboration of 2 different dispositional variants, that fulfil the binding and directive parts of the local plan. There was chosen one of the variants and elaborated in more details for the publication of the building licence. This work is also focused on suitable ways and possibilities of financing the building-up {--} the building savings mortgage credits, and their benefits. At processing the project documentation there are observed the legal rules and regulations, as well as the local plan and rules for the execution of the design documentation for the given building to construct.
Genetic and Proteomic Screening in Patients with Cardiovascular Disease.

The aim of this study is to analyse a genetic and proteomic aspects that could play an important role in development of chosen cardiovascular disease. Matrix metalloproteinases are enzymes that contribute strongly to the degradation of extracellular matrix components. In this study the serological levels of MMP-2 and MMP-9 were investigated using immunological testing in patients with aortic valve disease and in patients with myocardial infarction. Significantly higher levels of MMP-2 and MMP-9 were determined in both above mentioned groups of patients. Association of serum levels of MMP-2 and MMP-9 and development of concomitant aortic dilatation was not confirmed in patients with aortic valve disease. Changes in serum levels within 24 hours and after 6 months post myocardial infarction were characterized. About 10 % of patients operated for aortic valve disease suffer simultaneously from ascending aortic dilatation. The current study did not reveal any significant genetic variation in TGFBR2 gene and in chosen exons of FBN1 gene in these patients. Further genetic research is needed to identify the cause of the pathology in aortic wall. The diploma thesis focuses on the sustainability of social enterprises founded by NGOs after the end of their projects supported by the Call no. 30 HREOP "Social Economy". The first part contains the theoretical definition of the concepts of social economy and social entrepreneurship and conception of social enterprise in the Czech environment. More attention is paid to work integration social enterprises. The concept of sustainability of social enterprises is presented from the perspective of international research and also the only one Czech survey on this subject - Evaluation of Supporting Social and Inclusive Entrepreneurship in HREOP. The research examined the ability of 11 work integration social enterprises to operate their business after the end of financial support from HREOP. The research is also analysing other support possibilities for social enterprises. All surveyed enterprises operate after the end of the project and most of them are financially self-sufficient or at least heading to it. Every seat, whether it is a town or village, has in its own area space that can be used by the inhabitants of the settlements as well as the general public. The question in many cases is the quality and size of the public space. The most important public space in the seat is usually considered to be square or in the village the village square. In the seats there are also other public spaces, which are more or less used and more or less meet the purpose for which they were constituted.
