Úpravy rostlin a jejich využití v historickém interiéru

The subject of this final thesis was to study the techniques used to extend the life of plants, in practice to test and evaluate them. The obtained material is used to design an arrangement for specific historical interiors of Znojmo castle. The third task was to measure the temperature and humidity in one of the rooms where the arrangememnt was placed. The thesis includes a supplement with photographs of plants preserved with various techniques and examples of period interiors with installed floral arrangaments.

α-N-Acetylgalactosaminidase as a tools in the synthesis of complex oligosaccharide immune stimulators Introduction: Alpha-N-acetylgalactosaminidases are exoglycosidases specific for the hydrolysis of terminal α-linked N-acetylgalactosamine in various sugar chain. They occur widely in microorganisms, plants and animals, and have considerable potential in various industrial application. Stability and activity at high temperatures are important properties of α- N-acetylgalactosaminidases. A large screening study for extracellular α-N- acetylgalactosaminidase activity in a library of filamentous fungi (42 strains), led to the identification of the best constitutive producer, Aspergillus niger CCIM K2. The enzyme from Aspergillus niger has certain unique properties, and it was chosen for further investigations. Methods: Alpha-N-acetylgalactosaminidase from Aspergillus niger CCIM K2 was partially sequenced by Edman degradation and MALDI MS. Degenerated PCR primers were designed based on the obtained N-terminal and internal sequences. A gene fragment encoding a putative part of the α-N-acetylgalactosaminidase was amplified using cDNA prepared from Aspergillus niger CCIM K2. Non-stacionary thermal field in the nozzle at stream mixing

Nuclear energy is already well known area of energy generation. However, it is necessary to eliminate the potential risks of failure. That places special emphasis on safety in production and operation of nuclear power, especially its primary circuit. This work is focused on thermal damage to the injection nozzle, which is part of the pressurizer in the primary circuit Dukovany plant.The aim was a comparative analysis of two geometries nozzle which were loaded by varying temperature loading trought time and by static preassure loading. Main attention was placed on the thermal shocks in the material.

Ichtyofauna of selected streams of Bohemian Forest

Presented work is directed to summarize the results of ochtyomonitoring of five stream in Bohemian Forest in autumn 2010. The processing status is evaluated fish communities, especially their species composition, diversity, dominance, evenness and other parameters. They have been evaluated also physico-chemical properties of the sites such as air and water temperature, conductivity and turbidity. Last but not least were evaluated the characteristics of flows and in particular morphological characteristics, types of the bottom substrate and its contours etc. The evaluation of all obtained data showed that fish communities surveyed Bohemian flows are balanced, they are not burdened with invasive species and rand indicators correlated with the state of flow, geographic location and natural conditions. It was caught nine fish species in the total number of 963 individuals. Abundance was at all locations in the range 201,6 to 409,7 individuals per kilometer of flow, showed the diversity of communities from 0,372093 to 2,166085 (Shannon and Wiener index) and from 0,301804 to 0,866948 (Simpson index), then evenness of plant 0,372093 to 0,783906. The most dominant species was the brown trout, the second most frequent species was bullhead.

The production of secondary metabolites in the Trifolium pratense L. suspension culture (red clover) is low and therefore there was an attempt to increase it by elicitation. Elicitation is a method making use of protective mechanisms of plants to increase the production of secondary metabolites in plants and in vitro cultures. The jasmonic acid and its precursors and derivates are some of the endogeneous signal substances of the plants' defensive responses. In the exogenic application case, they may also function as elicitors. This diploma thesis examined the effect of jasmonic acid and the effect of jasmonic acid in combination with calcium ions and verapamil on the production of flavonoids and isoflavonoids by the Trifolium pratense L. suspension culture. The culture was cultivated in Gamborg medium to which 2 mg.l-1 of 2,4-dichlorophenoxyacetic acid and 2 mg.l-1 of 6-benzylaminopurine were added, at the temperature of 25řC, 16-hr light/8-hr dark period. Natural Swimming Pool for a Small Municipality

Bachelor thesis deal with natural swimming biotope. In the first part concetrates on the principle of water purification natural swimming pool, a device used for the operation, properties of water, right sort of plant and regulations dictated by the state. In the sekond part presents free selected natural swimming pool in southern Moravia, compares selected indicators in bathing water and source water in natural biotope. At first depots graphic progres sof temperature, transparency and pollution in a particular biotope and then compares individual biotope between them with respect to the notice of the Ministry of Health.

Study of cytokinin role in abiotic stress response

Plants had to evolve, due to their sessile growth habit, a complex system of defence against adverse environmental conditions. Plant abiotic stress responses are regulated by plant hormones, especially by cytokinins (CKs) and abscisic acid (ABA). In order to evaluate the effect of enhanced stress tolerance on the dynamics of hormones (CKs, auxin and ABA) as well as polyamine levels during the drought and/or heat stress (HS) progression, response of tobacco plants with enhanced level of osmolyte proline was compared with the corresponding wild-type. HS (40řC) caused a transient increase in bioactive CK content, accompanied by decrease of ABA, which indicated stimulation of transpiration, important mechanism to cool down the leaf temperature at the early phase of HS. Simultaneously, a transient increase in spermidine and spermine levels was found, more profound in proline-over-producing transformant. The activities of particular polyamine metabolic enzymes correlated well with their content. Drought stress resulted in the establishment of a gradient of bioactive CKs in favour of the upper leaves, crucial for their preferential protection. Application of HS at the end of drought period strongly enhanced the stress severity, mainly due to additional water loss caused by enhanced transpiration....

Based Sorbents for Dry Sorption of Acidic Compounds from Flue Gas from Municipal Solid Waste Incineration Plants.

The paper compares sodium and calcium based sorbents for the dry treatment of flue gas from waste-to-energy plants with respect to their surface, reactivity to individual acid compounds in the flue gas and process conditions. The emphasis is on the dry flue gas treatment at temperatures below 250 °C. Commonly used stoichiometric dosages of sorbents are compared in relation to the mass of sorbent used. In addition, an experimental unit for testing of dry flue gas treatment is introduced.

Heat hardening of Ni-P based coatings prepared on wrought magnesium alloys

This bachelor’s thesis is focused to hardening of nickel-phosphorus coatings on magnesium alloys. In the theoretical part are listed current knowledge about magnesium alloys and electroless deposition of Ni-P coatings for which is also stated mechanism and function of components contained in the bath. Theoretical part is ended by review aimed to the research in areas of chemical coating. Experimental part describes individual steps of pretreatment on magnesium alloy AZ31 and then planting of Ni-P coating on thus prepared alloy. In the next part is stated range of used hardening temperatures and mechanical repercussion to that. Composition and morphology of deposited coating and magnesium alloy were studied using scanning electron microscopy.
