Migration of pharmaceuticals in soil systems

The increasing use of drugs leads to their frequent occurrence in the environment, which poses a risk not only because it can lead to the proliferation of antibiotic-resistant bacterial strains, but also because it can threaten human health. Therefore, this work focuses on the issue of the occurrence and behaviour of two antibiotics in soil, specifically tetracycline and sulfamethoxazole. Due to the presence of soil organic matter, it is believed that the soil can partially prevent the spread of drugs that enter the environment. The diploma thesis involved conducting diffusion experiments in soil columns under real conditions to determine the effective diffusion coefficients of tetracycline and sulfamethoxazole in three extraction agents. Sorption and desorption processes were also conducted in the individual layers of the soil column. The concentrations of the leachates were measured using UVVis spectrometry and liquid chromatography, while FTIR spectrometry was used to confirm the presence of the bound drug in the soil column.
Study of selected antibiotics sorption on polyhydroxybutyrate

The bachelor thesis deals with the issue of sorption of antibiotics on microplastics. Antibiotics and microplastics are now a highly studied topic, due to the positive and negative properties. Antibiotics with groups of sulfonamides and quinolones were selected for the experiment. From sulfonamides, sulfamethoxazole and trimethoprim were selected, and from the quinolones, enrofloxacin and ciprofloxacin, which have already been the impetus for some microplastic studies. Biodegradable polyhydroxybutyrate was used as the microplastic. Two experiments were performed in the laboratory. The first aimed to monitor the time dependence of antibiotic sorption on polyhydroxybutyrate, the second the concentration. The concentration dependence was subsequently evaluated using the Freundlich and Langmuir isotherms. The method of liquid chromatography with mass spectrometry was used for analyzes.
Removal efficiency of selected drugs by various sorptive materials from water

This thesis deals with sorption of selected drugs from model water by various sorption materials. Contamination of water resources by the pharmaceutical industry is a major problem today. Wastewater treatment plants, whose technological processes are unable to completely remove them, have a significant share in the penetration of these substances into the environment. At present, attention is paid to alternative materials that are capable of eliminating these substances. One of the potential sorption materials is biochar as one of the main pyrolysis products. This work focused on the assessment of the sorption properties of the different types of biochar and commercially used active charcoal. The sorption properties of the individual materials were compared with respect to the non-steroidal anti-inflammatory substance ibuprofen and the sulphonamide antibiotic sulfamethoxazole. The results of vial experiments were analysed on a liquid chromatograph with mass detection.
Interactions of pharmaceuticals with humic acids

The bachelor thesis is focused on interaction of drugs with humic substances, which are part of organic soil matter. The results would help in understanding the behavior of drugs in soil and other natural systems. Three drugs were selected. Humic acid was selected as a sorbent, which was extracted from lignin, the lower one is assumed to be the same as that in organic soil. Based on a comparison of the measurement results using the UV-VIS method, it was determined how much the acid of the given drug is able to absorb and how much it is released back into the water. The theoretical part deals with the information about soil organic matter, with the information about humic substances, with the information about drugs such as ibuprofen, sulfamethoxazol or sulfapyridine. Furthermore, the theoretical part deals with methods of analysis.
Removal of sulfonamide antibiotics using advanced oxidation processes

This thesis deals with a present problem of increasing concentrations of pharmaceuticals and other micropollutants in aquatic environment, because these micropollutants are not treated sufficiently in conventional wastewater treatment plants. It is necessary to develop new technological processes, that are able to treat these pollutants sufficiently. One of the options is that conventional methods would be complementary with advanced oxidation processes (AOP), which would work as a tertiary cleaning process on wastewater treatment plants. The actual knowledge, principle of the plasma discharge and principle of analytical methods is described in theoretical part of the thesis. In experimental part of the thesis, the plasma discharge is used for removal of sulfonamide antibiotics (sulfamethoxazole), as the representative of AOP. Samples were taken during the plasma treatment of the model solution of sulfamethoxazole. These samples were analysed by LC-ESI-MS as the appropriate analytical method. An efficiency of degradation of sulfamethoxazole in different solutions (neutral, acidic and alkaline) was evaluated in this bachelor thesis. A suitability of plasma application on degradation of sulfonamide antibiotics was reviewed by comparing with other AOP.

The bachelor thesis focuses on a problem associated with diffusion processes of antibiotics in agarose hydrogels enriched with humic acids. The soil model system simulates the transport of these pollutants and demonstrates the ability of the humic acids to bind and retain these substances, thereby preventing their subsequent contamination at unintentional places. The theoretical section encompasses an overview of the occurrence of pharmaceuticals in soil and water systems; it also provides information about their behaviour, effects, degradation, and potential outcomes in nature. It further clarifies basic knowledge of soil components, in particular humic acids. Furthermore, the mechanism behind the diffusion processes is explicated. Within the experimental part, the diffusion of the sulfomethoxazole and sulfapyridine in two types of agarose hydrogels was examined. Firstly, the pure agarose hydrogel was used and secondly, the addition of the active agent in the form of humic acids was performed. The method of diffusion from continuous source was utilized, followed by measuring through UV-VIS spectrometry for 21 days. The study aimed to compare the results and to establish the diffusion coefficients of the pharmaceuticals in both types of hydrogels.

The Flow Injection Analysis of Procaine and Sulfamethoxazole with Spectrophotometric Detection

A derivatization reaction, based on a copulation reaction of diazonium salt of an analyte with a reagent (1-aminonaphthalene or NEDA) producing colour, spectrophotometrically detectable product, has been used for determination of two analytes (procain hydrochloride, sulfamethoxazole) containing amino group in the molecule. The conditions of batch spectrophotometric determination were studied. It was found, that the batch determination is applicable only for procain hydrochloride analysis, because sulfamethoxazole gives time-unstable colour product. FIA arrangement was proposed and effects of selected parameters were optimized using two levels factorial design. The calibration dependences were measured for both analysis with both reagents, and subsequently were applied on determination of analytes in medical forms.
