Microclimate and aircondition in spaces the depository

This bachelor´s thesis describes the design of air conditioning in space for the depository. The device is design to provide a suitable microclimate for storage a local mixed collections. The main task of the equipment is ensure a constant temperature and relative humidity in area depositary. Depositary space is served by one air handling unit, which is part of the proposed project. The theoretical part solves different kinds of inventory and their storage requirements. Calculation and project part contains design of air-conditioning units for operate depository. The result of this work is the detailed design work of this equipment.
Optimization of DNA amplification isolated from buccal swabs for sequencing purposes

The topic of my bachelor thesis was the issue of taking primary DNA samples from the buccal mucosa, determination of the ideal length and optimal conditions and subsequent analysis of these samples. In the theoretical part, I focused on the preanalytical phase of the laboratory examination. It was about, the methods of sampling and the type of material that is most often used used for DNA isolation in the molecular biological laboratory. Subsequently, I focused on the methods themselves, which are used for DNA analysis. These included DNA isolation, DNA amplification methods, nucleic acid electrophoresis and DNA sequencing. In the practical part, I processed 42 samples. First, I isolated DNA from the samples and measured its concentration and purity. Then I performed electrophoresis in these samples and based on its results I decided which samples I would process further by PCR. Subsequently, I determined by electrophoresis whether DNA was amplified in the samples. Finally, I selected samples that I sent for sequencing. The aim of my bachelor thesis was to determine the optimal length and storage conditions of samples from the buccal mucosa for further processing. Then I evaluated how these parameters affect the quality, purity and quantity of isolated DNA as well as the course of the polymerase chain reaction. According to the obtained results, the best option is to store the samples in a refrigerator temperature and perform their analysis in the shortest possible time, or to store samples in a refrigerator temperature and process them within weeks to three months.
Optimalizace uskladnění produktů prvovýroby v podmínkách ZD Ločenice

This thesis focuses on the storage of cereal and rapeseed grains in the company ZD Ločenice, which has been dissatisfied with the state of its storage facilities for a long time. The aim of this work is to objectively describe the current state of storage and to propose specific projects that will lead to an improvement. The information on the current state was obtained from consultations with the management of the company and from a tour of the storage buildings. As a result of the work, three specific proposals for improvements have been consulted with and agreed by the management. The company AGRICO Třeboň s.r.o. was contacted for the purpose of obtaining a price estimate and design options. The first proposal is for a new outdoor silo with a capacity of 1 000 t, the design of which is elaborated in two posible variants. The second proposal is a new silo located inside one of the existing storage buildings, also in two versions. The third proposal is a new conveyor linking the post-harvest line building to the two main storage buildings.
Handling and physical preservation of historical sound carriers: conditions and procedures of long-term physical preservation focused on phonograph cylinders and shellac discs

The presented methodology focuses on selected aspects of the preservation of historical sound carriers. Text aims on reflecting the physical conditions, basic assumptions and procedures related to the handling and care of phonograph cylinders and shellac discs. The methodology provides a brief historical excursion on technical parameters with a more details about material composition of these types of sound carriers. Furthermore, it discusses the approaches to storage conditions both on the scale of entire collections and individual sound carriers. Concerning this topic it deals with the possibilities of carrier assessment and appraisal of degradations and damages. Finally, the text is devoted to elementary methods of handling and care of both types of historical sound carriers.

Effect of different descaling/skin removing methods on sensory properties and shelf life of common carp

The present bachelor thesis studied microbiological quality, sensory attributes and customer preferences of common carp filet (Cyprinus carpio) descaled by different methods under two storage conditions. Fillets were processed by four descaling methods, namely: without skin, without scales, undercut scales and fillets of mirror carp (with skin). Moreover two types of storage conditions, air packed and vacuum packaged, were tested at 3.8 +- 0.5 ° C. Microbiological analyses were determined by total viable counts in logarithm of colony-forming units per gram (log CFU.g) on storage days 0, 3, 6 and 9. Furthermore, sensory analyses of fillets was classified and customer preferences were evaluated by using an anonymous questionnaire. In microbiological analyses, an increase of the total number of microorganisms with increasing storage time was observed. Microbial communities grew to between 2.3 - 2.69 log CFU.g-1 on the first day of fish processing (day 0) and 6.35 - 8.28 log CFU.g-1 on the last day of storage (day 9). Microbiological analysis showed that skinless fillets reached the lowest total number of microorganisms, regardless the type of packaging (air / vacuum packaged). Also sensory analyses showed that consistency and odour of carp fillets were influenced by packaging conditions as well as descaling methods. The best results in sensory evaluation achieved fillets without skin and undercut scales. The survey on consumer preferences of carp found out that 58.9 % of customers preferred scaly carp with undercut scales and processed to fillets. The results of present study show that different types of descaling methods combined with air / vacuum packaging can significantly prolong the shelf life of carp fillets.
Study of quality of different vegetable oils

The aim of the diploma thesis was determined the particular fatty characteristics of various vegetable oils in dependence on their storage life and storage conditions. Storage conditions were defined by room temperature and coolroom temperature. The fourteen oils were used for analyses, namely linseed oils, castor oil, grape seed oil, almond oil, olive oil, jojoba oil, avocado oil, apricot kernel oil, corn oil, rice bran oil, macadamia nut oil, soya bean oil, pumpkin seed oil and sunflower oil. All tested oils were supplied by firm M+H, Míča a Harašta s.r.o. The fatty characteristics included the saponification value, the acid value, the iodine value and the thiobarbituric value. The ester values and the percentage glycerol content were counted on the basis of the saponification value and the acid value. Time dependences of all fatty characteristics were figured to graphs and they were compared on various storage conditions. The fatty characteristics indicated lower values in storage of oil samples in coolroom than at laboratory temperature.
