Špenát setý-zhodnocení odrůdového sortimentu a možnosti využití v zahradnické produkci
Ondráčková, Lenka
The content of this thesis on the topic Spinach-evaluation of the assortment of varieties and the possibility of use in horticultural production, was the elaboration of a literature search describing spinach leafy vegetables, focusing on ingredients, cultivation technology and varietal assortment in our country and abroad. The practical part consisted of a field experiment, laboratory analysis of selected ingredients, morphological evaluation of parameters of spinach plants and practical tasting. The field experiment was focused on the evaluation of the varietal assortment of spinach and the effect of vermicompost on the quality of production. The results of the experiments were statistically processed and the effect of vermicompost on the yield and nutritional content of substances in the leaves of spinach was not confirmed.
Isolation of photosystem II particles and their characterization
Svitačová, Denisa ; Pekař, Miloslav (referee) ; Venerová, Tereza (advisor)
This master's thesis deals with the isolation and characterization of spinach thylakoid membranes and photosystem II particles. Based on the research, two procedures were chosen for the isolation of spinach thylakoid membranes and photosystem II particles with an emphasis on preserving their native properties. Subsequently, the characterizations were carried out, focusing on the spectroscopic properties of the isolated samples. UV-VIS spectrometry was used to determine the ratio of chlorophyll a to chlorophyll b in samples of thylakoid membranes and photosystem II particles. The success of the isolations was confirmed by fluorescence methods. The excitation and emission spectra of the samples were measured by the steady-state fluorescence spectroscopy, and the fluorescence lifetimes of the samples of thylakoid membranes and photosystem II particles were determined by time-resolved fluorescence. The isolated samples were afterwards visualized with a fluorescence microscope with a CCD camera and the FCS technique.
Diurnal changes of Rubisco activity and its content in spinach leaves
Riegerová, Jana ; Vespalcová, Milena (referee) ; Hrstka, Miroslav (advisor)
The graduation theses theme was to determinate changes of initial activity, total activity and Rubisco enzyme content in spinach leaves. Ascertained activities and Rubisco enzyme content were determined from extracts of spinach leaves, which were collected during two distinctively climatic different days. Both activities were determined by spectrometric method and content of enzyme Rubisco by SDS-PAGE method. The findings were compared with findings determined in earlier done works.
