Cell inclusions in microscopic eukaryotes

This thesis reports, for the first time, a systematic study identifying various cell inclusions across the broad diversity of all major eukaryotic supergroups. Raman microspectroscopy technique was employed as a powerful technique, which is becoming the method of the first choice for such studies. In Chapter 1, the spatial reconstruction of chloroplasts of the unicellular zygnematophyte Cylindrocystis sp. (Streptophyta) revealed "empty spaces" inside. Subsequent analysis showed massive accumulations of polyphosphate, which is not located in the cytoplasm as in other species, but inside the chloroplasts. This newly discovered ability of the zygnematophytes, a sister group of terrestrial plants, to accumulate polyphosphate leads us to speculate about a possible adaptation to extreme Arctic conditions or even a preadaptation leading to plant terrestrialization. Chapter 2 summarizes the serendipitous discovery of purine crystalline inclusions in dinoflagellates and other microalgae. Investigated dinoflagellate species comprised zooxanthellae, the endosymbionts of reef-building corals, and the species causing toxic algal blooms. Proposals of Practical Excercises in Zoology Based on Protists and Invertebrates from Freshwater Aquaria

This thesis deals with proposals for practical exercises using freshwater aquaria in teaching elementary schools and junior high schools that focus on protozoa and invertebrates. The main aim of the thesis is to propose practical exercises using elements of inquiry-based science education. Using identification cards of protozoa and microscopic invertebrates, pupils can learn about these organisms during laboratory exercises with microscopes. A total of fourteen cards with representatives of protozoa, invertebrates, algae and cyanobacteria typical of freshwater aquariums are presented. Each of the identification cards contains a basic description, as well as a procedure for the pupils to observe and draw the organism. They are accompanied by a photograph, a pen drawing and a QR code that links to a video of the observation of the representative under the microscope. Another proposal is a practical exercise focusing on the food chain of standing water. Also included is a worksheet focusing on independent laboratory work by students on water quality analysis using the Pasco laboratory technique. Interactions among free-living prokaryotes and protists in microbial communities

Protists and prokaryotes live in the natural environment in complex microbial communities whose members interact with each other. These communities consist of a few to many species that interact with each other in many ways such as predation/prey, ecto/endosymbiosis, parasitism, modulation of development or behaviour and harmful algicidal/bactericidal interactions, resulting in a complex network of interactions that influence structure of the microbial community as well as metabolism and behaviour of present species. This bachelor thesis is an overview of the interactions between free-living protists and prokaryotes within microbial communities, excluding well-studied interactions such as predation, parasitism, and ecto/endosymbiosis, while focusing on the diversity of lesser- known interactions, highlighting their importance and future research opportunities.

Communities of protists and invertebrate animals in freshwater aquaria and possibilities of their use in teaching

This bachelor thesis focuses on the hidden but very diverse community of protists and microscopic invertebrates in freshwater aquaria. Samples of these micro-organisms were obtained from microscopic slides submerged in aquaria for several weeks. The main output of this thesis is a multimedia didactical atlas of characteristic protists and invertebrates inhabiting freshwater aquaria. In total, eleven members from two phyla of protists and six phyla of invertebrate animals are presented using own original micrographs and short videosequences. The atlas was prepared in two forms - a text document and a web page. Potential of the atlas for use in biology teaching and extracurricular education is discussed. 4 Abstract The main goal of my thesis was to create our own proposals for laboratory work in biology of unicellular organisms (Protista). I designed the laboratory tasks, so that they can be mastered by the pupils of sixth year of primary school. Creating the proposals for laboratory exercises was preceded by the work in the laboratory at Charles University in Prague, Faculty of Education, the Department of Biology and Environmental Studies. In the laboratory, I verified the methods of cultivation of selected representatives of ciliates (Paramecium caudatum), amoebae (Amoeba proteus) and euglenids (Euglena gracilis) and selected methods of dyeing their different organels. Parasitic diseases of Australian parrot species

Australian parrot species can host many species of parasites. Parasitic diseases affects both parrots kept in captivity and living wild. This work is compiled as a literary review and provides a summary of most parasites with confirmed presence at the Australian species of parrots. Many parasites species have been detected recently. The presence of new species is confirmed also for parrots kept in Central Europe (coccidia Sarcocystis calchasi) including the Czech Republic (parasitic protozoa Leucocytozoon spp., roundworm Ascaridia platyceri). Parasitic infections may be detected trough examining samples from living parrots (feces, blood, tissues) or trough necropsy of died individuals (histopatology). In psittacines, protozal infections have been described (phylum Metamonada and Apicomplexa). Giardiasis, coccidiosis, cryptosporidiosis and blood parasites (Plasmodium spp., Leucocytozoon spp., Haemoproteus spp.) are among the most frequent. Australian parrot species are among the most frequent hosts of nematodes roundworms (Ascaridia spp.) and capillaries (Capillaria spp.). Regarding the strain Fungi, presence of Encephalitozoon hellem which causes microsporidiosis which primarily occurs in humans with acquired immunodeficiency has also been identified in psittacines. This makes it a potentially zoonotic disease. As of the ectoparasites, those most important representatives from the variety of species of parasitic mites and insects are mentioned. The life cycle of the parasites, their pathogenicity, ways of transmission, clinical signs in affected individuals, diagnosis and treatment options have been described.
