Web Application for Primer Selection for 16S rRNA Amplicon Sequencing

Main goal of this thesis was an implementation of web application, that can evalutate and recommend primer pairs for 16S rRNA amplicon sequencing, according to users specific needs and by this simplify the procces of primer pair selection. Primer pair evaluation is based on database of primer pairs, which contains data about senzitivity and specifity of each primer pair. Part of the work was also the performance optimization of the primer pair analysis algorithm, that computes senzitivity and specifity data. This optimization helped in an integration of algorithm into the application, which means that users can submit their own primer pair sequences, run analysis and get informations about position of amplified region, sensitivity and specifity.
Assessment of Efficiency of waterborne penetrations for cement substrates

The bachelor's thesis is addressing the issue of using primer coatings for cementitious surfaces. It analyzes reasons of using primer coatings and specific features of application. There are mentioned different species of surface treatment for cementitious surfaces which is followed by methods of surface treatment by variety cementitious screeds, and self-leveling flooring systems. Then it is looked into general characterization of polymer dispersions (especially water-based dispersions) which have typical attribute for primers. In experimental part of thesis are compared ten different waterborne primers. Testing is briefly described and evaluated.
Dynamic Analysis of Selected Firing Mechanisms of Firearms

Bachelor thesis engages in analysis of firing mechanisms of chosen firearms. In introductory part of the thesis, there is a research made which involves gun problems, theoretical basis of gun theory is summarized and detailed description of firing mechanism of some types of guns is given. Alternatives of solution of the set assignment are analyzed at the end of the theoretical introduction. Practical part of the thesis deals with firing mechanisms’ analysis of four different types of firearms and subsequently with theoretical calculation of initiation energy and impact speed of the firing pin. This calculation is in conclusion applied on some specific guns. Final output is the determination of initiation energy, impact speed of the firing pin and safety in safe firing of the bullet's primer.
Evaluating the Quality of Combination Gun's Firing Mechanism

This Master’s thesis deals with evaluation of quality of Brno Combo combination rifle shotgun rifled barrel’s firing mechanism. It contains a proposal of criteria for evaluation of quality of firearm firing mechanisms. It further focuses on using measurement of primer deformation as a means of assessing the quality of a firing mechanism. Based on measurements made using a Focus Variation microscope, various geometrical parameters of fired and failed primers were analysed. The measurement results were processed using ANOVA and regression analysis. Based on output of these analyses, practical recommendations and further research proposals were formulated.
Analytical model of gun firing mechanism dynamics

This bachelor thesis is devoted to creation of the analytical model of the dynamics of gun firing mechanism, that is used for design of individual parts of the mechanism, allowing the primer to be initiated properly. This analytical model has been verified by experimental measurement of the real mechanism operation from Brno Combo shotgun using a high speed camera Vision Research Phantom v710. In addition, the minimal energy, needed for initiation of FOCCHI 616 (.209 TYPE) primer, was experimentally measured using the test device and the high-speed camera. The results of experimental measurement of minimal initiation energies were compared with the experimental data from Sellier & Bellot primer producer.

Detekce bakterií rodu Xanthomonas patogenních pro rajče a papriku metodou loop-mediated isothermal amplification

The subject of this thesis is the design of specific LAMP primers and LAMP analysis and its optimization. The purpose of this analysis is to specifically distinguish Xanthomonas gardneri from other pathogenic xanthomonads. Two sets of LAMP primers from the DNA sequences of hrpB and atpD genes were designed. These LAMP primer sets are highly sensitive and the detection limit of LAMP assay was found to be 0.01 l / mg of DNA. The reaction temperature of the LAMP was optimized to 64 °C to reach the maximum of amplification. LAMP can be applied to detection and identification on pathogens in plant tissues. LAMP takes less time than conventional PCR to detect bacteria. This LAMP assay has a great potintial to be applied to detection and identification of Xanthomonas genus bacteria pathogenic for tomato and pepper.
