Processing of sauerkraut juice to enhance its stability and consumer acceptance

This diploma thesis focuses on the development possibilities of fruit and vegetable juices containing sauerkraut juice, the characterization of their nutritional and sensory qualities and their stability, as well as the potential for enriching selected juices with probiotic culture of Lactobacillus plantarum. The best combinations of various juice mixtures were selected based on sensory analysis and were stored for 30 days at 8 °C. Changes in selected parameters were monitored during the storage. Antioxidant activity, total flavonoids and polyphenols content and content of total water-soluble and reducing saccharides were determined by spectrophotometric methods. HPLC was used for lactic and L-ascorbic acid determination, flow cytometry was used for probiotic cell content determination, culture test was used to observe viability of cells, pH was monitored. Contents of antioxidant activity and of total polyphenols content did not drastically change during the storage. Major changes were observed in total and reducing sugar content and total flavonoids content. It was observed that probiotic viability was preserved for 14 days, decrease occurred afterwards. In order to achieve as high content of probiotics as possible, it would be advisable to store probiotically enriched juices for maximum of 14–19 days at temperature of 8 °C. The highest viability was observed in JHM sample (containing apple, pear, carrot and sauerkraut juice in the same ratio). Even through viability of probiotic cells was determined as low, these presented functional beverages remain nutritionally, visually and taste-wise interesting products, which can help popularize the consumption of non-traditional local foods.

This bachelor work addressed the issue of the use of types of carrots in carrot breeding and development of new varieties. It describes the theoretical part of this issue, mainly dealing with the basic description of the types of carrots and carrot breeding. Furthermore, an attempt was founded varietal carrots, which were classified in different experimental variations, which constituted the main representative of the types of carrots. They were evaluated and compared with each other types of carrots, depending mainly on market demand for varieties of root vegetables and a suitable extension type and varietal range carrots with us. According to various breeding parameters were determined by the overall suitability of particular types of varieties of carrots for use in breeding process.
Ověření vlivu použití biostimulačních látek a mykoparazitických hub při inkrustaci osiva

In the master thesis The verification of the influence of biostimulants and mycoparasitic fungi used in the seed incrustation technology were evaluated species parsley (Petroselinum crispum convar. radicosum) and carrot (Daucus carota L. subsp. Sativus) grown on 2 locations in 4 variants of incrustation and control variant. Total yield, marketable roots yield and percentage of marketable roots yield from total yield were determined. Research has shown that there has been a significant impact especially in plant health condition. An increase in yield was proved in variant using treatment by Clonoplus combined with CAPTAN 80WG, and also treatment by Clonoplus and Sunagreen-M combined with CAPTAN 80WG. An influence on increase of percentage of marketable roots yield from total yield was more significant in carrot. No phytotoxicity was proved in the germination test even during long-term storage of the seed.

The diploma thesis is focused on the use of the ISSR and SSR methods, with the intention of genetic differentiation of 7 varieties of carrot (Daucus carota L., subsp. sativus (HOFFM.) SCHUBL & MART.). The literary part of the thesis is focused on the basic characteristics and history of carrots. The following part of the thesis is focused on the description of molecular methods based on PCR, which are RAPD, AFLP, ISSR, and SSR, and also the description of the PCR method itself. The purpose was to find suitable ISSR primers and SSR markers that could distinguish individual carrot varieties. Based on analyses and their results, dendrograms of the genetic affinity of observed varieties were created.

Bachelor thesis Options for improving the quality of seeds using stimulants in seed technology (seed treatment, incrustation) describes possibilities of increasing seed quality in general, especially the seed plants in the family Apiaceae, which has long and difficult germination. There are described various factors affecting the quality of seeds and ways to improve seed quality. In the practical part were evaluated two kinds of plants: parsley (Petroselinum Crispum (MILL.) NYM. ex A. W. HILL convar. radicosum (ALEF.) DANERT) and carrot (Daucus carota L. subsp. sativus (HOFFM.) SCHÜBL & MART.) grown on two locations in 3 variantions of seed treatment and also control variant. Roots were weighted for evaluate total yield, yield of roots for markets and percentage of market roots from total yield.
Optimizing of weed control in carrot anf lettuce

The main aim of this work was to made literary review about possibilities of weed control in carrot and lettuce. The experimental part of this work was focused on optimization of herbicides weed control in lettuce and carrot. Small plot field trials were carried in 2015. Literary review was focused on the summarizing of information about the particular vegetables. Futhermore, the available literature about weed control in these vegetables were complied, specialy with a focus on mechanical and chemical (herbicide) control. The last section of review is dedicated to optimizing use of herbicides that are not registered in tested vegetables. In lettuce, herbicidal efficacy was assessed on Chenopodium album, Amaranthus retroflexus, Mercurialis annua and Echinochloa crus-gali and selectivity of herbicides to crop. In carrot, herbicidal efficacy was assessed on Amaranthus retroflexus, Solanum physalifolium, Chenopodium album, Echinochloa crus-gali and Mercurialis annua and selectivity of herbicides to crop. Lettuce was treated by herbicide combinations with active ingredients: befor planting or postemergence pendimethalin, postemergence propyzamide, postemergence phenmediphan, postemergence dimethenamid and postemergence pethoxamide. Carrot was treated by herbicide combinations with active ingredients: preemergence aclonifen, preemergence clomazone, preemergence dimethenamid, preemergence pendimethalin, postemergence linuron and postemergence metribuzin. In lettuce, were on the plots, where were used pendimethalin and dimethenamid. In carrot, best weed control were on the plots, where were used aclonifen, clomazone and dimethenamid. However, after application these herbicides, highest phytotoxicity was found. The lowest selectivity to both tested vegetables showed dimethenamid.

The work deals with testing sowing treatment of seeds , methods of Bio- osmopriming and hot water treatment (HWT) on seed pea (Pisum sativum) and carrot (Daucus carota) and the subsequent evaluation of the success of probing methods of sowing treatment for individual crops in field conditions to test germination in the laboratory. Investigation and evaluation parameters are the number of germinated seeds in laboratory conditions , germination rate (how many seeds germinated in each day) , the number of emerged plants in the field, plant health field (plant infestation and plant health), composure stand and yield evaluation of test cultures. final results are photographically documented and processed in tables and graphs (statistically) and compared.
