Kvasinky jako součást mikrobioty osidlující potravinové suroviny a potraviny

In the literary research this thesis summarizes general information about the yeast genera, including cytology, morphology, and physiology of the cell. Yeasts are being classified within taxonomy. Some of the most important genera found and used in the food industry are characterized. Theses then describes both the cultural and contaminating microbiota of main food groups. Lastly, the paper describes some commercially used preservation techniques. The experimental part of this thesis is focused on assessing total microbial count (TMC) and yeast and mold count in two samples of spontaneously fermented beer as well as two samples of wine with different treatment. This is to capture the natural microbiota of these alcoholic beverages. In the case of yeast and mold count, samples were inoculated in chloramphenicol yeast glucose agar, in the case of TMC the plate count agar (PCA) was used. Found yeast was reinoculated in order to obtain a pure culture strain. Yeasts were identified using a CANDIDA-screen set (Erba-Lachema). C. glabrata, C. krusei, C. lipolytica and S. cerevisiae were found. Molds have been found on PCA. They were isolated and identified under microscope based on their morphological features. 3 Penicillium molds and 1 Aspergillus mold were identified.
Construction of foundry core boxes and their use for the production of silicone parts

The thesis deals with the design of a mold for use in casting of plastics and rubbers. The product is two different gaskets used in the testing of tubes for automotive industry. The part has a cylindrical shape with a diameter of 19 mm and a length of 25 mm, where two holes with a diameter of 4 mm are additionally drilled in one of them. Depending on the service life, the batch size is approximately 200 pieces per year. The material was not specified, so after research two options were chosen. The first was RTV-2 condensation silicone rubber LUKOPREN N 1522 from Lučební závody a.s. Kolín and the second one was RTV-2 additive silicone rubber ELASTOSIL M 4642 A/B from Wacker-Chemie, s.r.o. The second material came out better from preliminary testing. By gradually improving the design of a simple single-piece steel mold, a final version was created which is double in size and consists of three parts. It is made by 3D printing using FDM method from PLA material. It was found that in order to achieve a defect-free casting, the mix must be vacuumed before casting.

Production of casting cylinder for engine modification of motorcycle Jawa 50. This bachelor thesis deals with the design of an air-cooled cylinder for racing modification of the JAWA 50 engine and its subsequent implementation using the rapid prototyping function. The designed part was cast and the finished casting was subjected to analysis.
Machining of vulcanizing molds from aluminum alloys

This diploma thesis is focused on the issue of deep drilling of vulcanization molds from various aluminum alloys. The work will describe a brief description of machined materials and tools for deep drilling, including the production of the vulcanization mold itself. Based on experimental measurements, the analysis of data and knowledge gained from deep drilling of aluminum alloys with different silicon contents will be performed. In the end, the individual components of force loads for both machined materials will be compared
A proposal and production of vacuum mold for serial production of plastic forming

This diploma thesis deals with the proposal and production of a mold for series production of plastic parts by vacuum forming. The thesis is divided into 2 main chapters. The theoretical part includes analysis of chip machining technology. The experimental part focuses on solving the design of the mold in PowerShape software, a detailed analysis of making of the model for casting and about making of the mold themselves. In the last part the manual completion and verification of the mold is discussed. Thesis is terminated by technical-economic evaluation, in which is discussed about production costs.
Combination of numerical mathematics and neural network for the model of breakout prediction

This thesis deals with creation of breakout prediction model implemented via artificial neural network in MATLAB, hence it contains theoretical description of artificial neural networks and also presents how the networks were used for the breakout prediction problem. In addition, models of heat conduction through the mold wall were created and used for generating neccesary data for proper training of artificial neural network. The thesis contains proper description of used numerical methods for solving the heat conduction problems. In the thesis are presented partial results and also results of testing created artificial neural network with usage of real data.
Heat transfer solution of solidifying steel system with phase change with moving edge conditions

Cílem diplomové práce je vytvoření 2D numerického modelu pohybujícího se řezu s proměnnými okrajovými podmínkami skutečné geometrie plynulého odlévání a chlazení předlitku v prostředí MATLAB. Model se zabývá vysoce nelineárními termofyzikálními podmínkami oceli během tuhnutí a chlazení. V práci je simulovaná nejen nelinearita termofyzikálních podmínek, ale také nelinearita při fázové změně. Fázová změna je modelovaná pomocí metody entalpie, metody zdánlivé kapacity a metody teplotního zotavení. Všechny výsledky práce jsou porovnány z více hledisek, jako např. z hlediska přesnosti, rychlosti výpočtu, nebo vhodnosti časového diskretizačního kroku pro nelineární problémy, a paralelizace.
The issue of biotic attack of ETICS

This bachelor thesis deals with the issue of biotic attack of ETICS contact insulation systems. The theoretical part is focused on the individual components of ETICS, both from the point of view of materials and from the point of view of individual steps. In addition, there are organisms that cause biotic attack on facades. The practical part deals with the possibilities of testing the resistance of the surface treatment of ETICS against biotic attack, both in laboratory conditions and in the exterior.
Proposition of casting technology of aluminium manifolds cast by tilt method

Description of the theoretical factors for casting into metal molds. For thin-walled castings it has turned out that the most advantageous method is tilt casting. Tilt casting reduces the turbulence of the melt and reaches a directed solidification. Two molds were tested and measured for the experiment. During measuring the temperature regime was established lower temperature of the pouring basin than the other parts of the mold, which could cause a many of defects. The verification of mold technology has been performed. To achieve the accuracy of the casting, it is necessary to respect the different shrinkage according to the shape of the casting.
A proposal of production technology of a mold for plastic covers

The bachelor thesis deals with machining of mold for blowing plastic packaging in cooperation with VBF Ltd. It focuses on 5-axis milling technology. It also contains descriptions of mold materials and injected materials. It analyzes the current process of mold production. It also contains its own proposal to improve (to make more effective) the production of a given block. The difference in technology used and the time difference are described in the conclusion.
