Artificial Immune Systems for Spam Detection

This work deals with creating a hybrid system based on the aggregation of artificial immune system with appropriate heuristics to make the most effective spam detection. This work describes the main principles of biological and artificial immune system and conventional techniques to detect spam including several classifiers. The developed system is tested using well known database corpuses and a comparison of the final experiments is made.

iNKT cells are a subset of T cells, they express the same phenotypic markers usually found on T and natural killer (NK) cells. They comprise a lineage of CD1d-restricted glycolipid-reactive T lymphocytes which have an important role in imunity. They play critical role in the regulation of many different types of immune responses, ranging from self-tolerance and development of autoimmunity to responses to pathogens and tumors. These cells are gaining increasing importance in health care, particularly in hematooncology. In this study we examined the essential features and functions of Invariant Natural killer T cell (iNKT cells).iNKT cells were isolated from mononuclear cells (MNCs) of healthy donors. Then they were cultured under appropriate conditions in vitro. After reaching sufficient concentration and purity the iNKT cells were used for cytotoxic and immunomodulatory assays and immunophenotype determination. Our results showed that iNKT cells have varying qualities across donors and that they have a heterogeneous immunophenotype, cytotoxic and immunomodulatory potential.
Adaptive immune system in patients with primary immunodeficiencies

(ENG) This thesis summarizes the results of a project dedicated to adaptive immune system of patients with partial DiGeorge syndrome caused by deletion of 22q11.2. The introduction sets the DiGeorge syndrome into a broader context of international pathophysio- clinical classification of primary immunodeficiencies and goes into detail describing its history, causes, clinical phenotype, therapeutic options and changes of the immune system. The attached manuscripts illustrate the premature aging of the T cell population, but also impaired development of B cells with low class-switched memory and high naïve subpopulations, along with high serum levels of BAFF, a B cell survival factor. The surprising lack of T independent marginal zone- like (MZ-like) B cells is reflected in decreased natural anti-α-Gal antibodies. The faulty B cell maturation and imperfect germinal center response is not caused by a deficit of follicular helper T cells, which are in fact increased in DiGeorge syndrome patients, and in most cases doesn't lead to hypogammaglobulinaemia. Mutační status a jiné chromozomální změny u chronické lymfatické leukémie

This thesis is focused on mutation status and other chromosomal changes in chronic lymphocytic leukemia. CLL is a lymphoproliferative illness with a low malignancy based on clonal proliferacy of malignatly transformed B lymphocytes. Although this is the most frequent malignant illness in western population, the cause leading to its origin has not been yet clearly given. Identifying prognostic markers has a significant meaning in identifying the illness prognosis, the choice of treatment strategy, and it influences general survival of patients. The aim of this thesis is to describe the frequency of the most usual mutations in patients suffering from CLL. All the results obtained from CLL patients were acquired from the Hospital in České Budějovice from 2016 to 2018.
