Influence of plasma activated water on seed germination and quality of corn

The bachelor's thesis deals with the effect of plasma-activated water on corn germination. After water comes into contact with plasma, the water changes its chemical composition and thus, it acquires new properties that can be used in agriculture. The theoretical part is divided into two basic parts. At the beginning of the first part, plasma is briefly characterized, and then this part deals with plasma-activated water, its formation, physical and chemical properties, and at the end of this part, its use. The second part of the theoretical part is devoted to the germination of plants, external influences affecting germination and statistical evaluation of germination. At the end of the theoretical part, this thesis deals with the ecotoxicity of plants. The content of the experimental part was the preparation of PAW, its subsequent application to corn seeds and the study of its effect on germination and plant quality. PAW was created from distilled or tap water using three different plasma systems and the effect of each PAW on the seeds was compared with that of each control sample. The obtained results indicate a positive effect of PAW on seed germination, but the final hypothesis would require more extensive experiments in different environments, especially in soil.

Czech maize hybrids are grown by the company CEZEA Čejč. OSEVA Bzenec is an exclusive producer of Czech corn hybrids of the cultivation plant CEZEA Čejč.The Central Agricultural Inspection and Testing Institute (CAITI)checks this for quality. If the seed does not meet the standards, it is not recognized. To avoid all of the processes from the delivery up to dispash of the dressed seeds are supervised by a laboratory of Oseva.My disertation monitors qualites of the maize seed. This seed was harvested in 2008 and 2009. After several years since the harvest, should be this seed decrease your germination capacity and incrase sensitivity to early sowing. Farther I evulate diference in the germination capacity by the small and bigger kalibrs by the cool tests. The evaluace is for the store seed. I suppose, that bigger grain has more energy to the germination capacity and is resisted to the ageing.

ABSTRACT The objective of this study was to compare the germination of cultural and weed variety of selected species of the family Apiceae. There were selected species: Daucus carota and Pastnaca sativa. For this research were selected three stocks of Carrot: Daucus carota -- wild, Daucus carota -- seeded from stored root in the garden - variety Olympus and Daucus carota -- originated from the propagation crop species -- variety Cider. For Pastinaca sativa were selected two populations: Pastinaca sativa - from the propagation crops and Pastinaca sativa - wild. For analyzed species the germination of achenes was determined under laboratory conditions. Average germination detected in Daucus carota - wild was 51%, for carrot Olympus 74% and for carrot Cider 89%. Average germination detected in Pastinaca sativa -- wild was 2%, for long white parsnip varieties 87%. The final germination was entered into tables and was processed to charts. Results were compared, in the discussion there were expressed some conjecture what effects might have contributed to the observed differences. There was also mentioned the possibility of affecting the quality of propagation material impurities of free growing stock.

The aim of this thesis was to find out the influence of temperature on germination seeds of Galium aparine. There were suggested labour experiments. In the first experiment seeds were exposed to various temperatures. In the second experiment was monitoring the influence on germination of seeds which were exposed to frost for a specific time. Best germination had seeds at 6 °C. In the second experiment the best germination had seeds that were exposed to frost for five days. Average total germination of all seed were 65.8%. Results have been written to the tables and processed to graphs. Evaluation of the work is adduced in discussion.
The influence of external factors on the growth of selected species of genus \kur{Digitaria}

Fonio is native to West Africa and is one of the world's oldest grains. It is a great source of vitamins, minerals, fiber and sulfur amino acids - methionine and cysteine. The aim of the work was to determine the optimal conditions for germination and seed growth of a selected representative of the genus Digitaria in laboratory and field conditions, and to evaluate the possibility of cultivation in the Czech Republic. Under the laboratory conditions were evaluated the effect of drought, salinity,presence of aluminium, waterlogging, light intensity, and storage temperature on seed germination of fonio. Furthermore, laboratory emergence of plants in different substrates and from different sowing depths was also evaluated and a field experiment was performed with different sowing dates. The most suitable temperature for germination of fonio seeds is 30 ° C in the dark. Fonio seeds were resistant to salinity to a concentration of 200 mM sodium chloride. The presence of aluminum had no significant effect on germination. Fonio exhibits very good resistance to waterlogging. Seed germination of fonio was not affected by drought stress up to a PEG solution concentration of 15%. Germination was not reduced by storing the seeds at 21°C for 3 months. The germination of fonio seeds was not significantly affected by the type of substrate or the sowing depth.
Effect of year on field germinability of opium poppy seeds

The aim of this thesis was to perform an experiment with opium poppy control and drought stressed plants. Gained seeds were tested for germinability in normal conditions and during simulated drought stress induced with PEG 6000. It was found plants for seed production provides less seeds with lower quality, but the vigour and germinability of these seeds are better. In this thesis I also tested the method of surface plasmon resonance as potentialy suitable method for dehydrin accumulation analysis. I managed to verify method for termostable proteins isolation and at the same time I confirmed, the SPR method is potentially usefull method for dehydrin accumulation analysis in opium poppy seedling plants.

The success of seedling survival was studied in microhabitats of a pasture. Seed germination of three species Trifolium pratense, Achillea millefolium and Carlina acaulis was observed in different types of microhabitats i.e. molehills, anthills, wild boar disturbances, and artificial gaps. Also, the environmental factors were measured. The observed microsites varied greatly in their soil conditions and seedlings of Trifolium pratense performed the best, particularly in wild boar disturbances.
