Study of organs of mice in inhalation experiments

The diploma thesis is devoted to study potential risks for living organisms resulting from inhalation of nanoparticles of chosen heavy metals. The main aim of this work is the investigation of organs from experimental white mice after their guided inhalation of lead nanoparticles. The samples of liver and lungs from these animals are taken in time periods during the inhalation experiments and processed by specific methods for their evaluation. After pressurized microwave assisted decomposition of selected samples of mice organs in high purity concentrated nitric acid the content of lead is determined by atomic absorption spectrometry with electrothermal atomization. Furthermore, the changes are monitored using the microscopy of histological slides stained with hematoxylin-eosin. The results of both approaches are evaluated and compared within the frame of this work.
Carotid Artery Stenosis

The basis of the doctoral thesis consists of three studies focused on the diagnosis of carotid stenosis. The first study examines the histological structure of carotid plaques and their relationship to clinical manifestations. The study included 269 patients with 280 analyzed plaques. Multiple logistic regression demonstrated a significant association between symptomatic stenosis and foam cells (OR=1.9, p=0.03) as well as plaque thrombosis (OR=3.5, p=0.02). A significant correlation was also found between symptomatic plaque and unstable plaque according to the comprehensive AHA plaque classification (OR=1.8, p=0.03).The second study addresses the accuracy of carotid stenosis diagnosis using digital subtraction angiography (DSA). Carotid stenosis was measured on DSA and histologically processed plaques obtained during carotid endarterectomy in 644 patients. The study did not establish a correlation between the findings on the histological plaque and DSA (correlation coefficient of 0.22 for ECST and 0.20 for NASCET). The last study focuses on the analysis of the biomarker of oxidative stress (malondialdehyde, MDA) as a predictor of unstable carotid plaques. The study involved 35 patients. The present thesis investigated the impact of a ten-week oral exposure to BPS (50 ?g/kg live weight/day) in female outbred Wistar Han rats. The effect of BPS exposure on live weight, absolute and relative kidney and liver weights, morphometric parameters of kidney and renal corpuscles, histopathological changes of kidney and liver were evaluated. In addition, the concentration of total protein, triacylglycerols, cholesterol, urea and alanine aminotransferase activity were determined in blood serum.
Diagnostics of Helicobacter pylori using histological and immunohistochemical examination

The main topic of this bachelor thesis is the bacterium Helicobacter pylori, its occurrence in the gastric mucosa, and the metod of its diagnosis in the laboratory using histological and immunohistochemical examination. In the theoretical part, I describe the bacterium itself, its morphology and mainly its diagnostics. There are many ways to detect the presence of this bacterium, most commonly divided into invasive tests, which include histological examination, cultivation and urease test, and non-invasive, where we can use serological and breathing tests. I also describe the anatomy and histological structure of the stomach in this part. Then I follow up on stomach pathology, where I focused on the most common diseases caused by Helicobacter pylori, acute and chronic gastritis and stomach tumors. The over-all histological examination is also included, its parts being receiving the sample to the department through fixation with examples of various fixation agents, to embedding, cutting and staining, where I also describe the types of staining that are used in the practical part. I also deal with the topic of immunohistochemistry. One of the goals of the practical part is to master the basic routine Hematoxylin eosin staining, special Giemsa staining and immunohistochemical examination of the antigen using the Anti-Helicobacter pylori polyclonal antibody. Another goal is to map the occurrence of Helicobacter pylori infection based on age groups, sex of examined patients and season. I set two research questions that I would like to answer. Results obtained in the year 2022 will be statistically processed and supplemented with photomicrographs of selected samples.
Dopad různé alimentární expozice bisfenolu na játra potkanů

Bisphenol S (BPS) is an organic compound widely used in industrial production as a substitute for bisphenol A (BPA) for which negative impacts on the human body have been demonstrated and have therefore been significantly regulated. However, as time progresses, the negative properties of BPS are also shown, especially for the endocrine and reproductive systems. The liver is an organ in which xenobiotics are detoxified and thus BPS and BPA. The aim of the bachelor thesis was to verify whether 10-week oral exposure to BPS (4 g, 50 g, 100 mg/kg bw/day) and BPA (100 mg/kg bw/day) will lead to a negative impact on body weight, weight gain, weight and morphological liver structure and on the concentration of triglycerides, cholesterol, total white-vinas, urea and alkaline phosphatase activity in blood serum. The experiment involved 24 rats. BPA and BPS were administered daily by gastric tube in sunflower oil. The results of our research clearly did not show a negative effect of BPS and BPA in rats on body weight, absolute gain, absolute and relative liver weight in rats. There was also no evidence of histopathological changes in rat liver. In contrast, bio-chemical parameters were altered compared to the control group. Decreases in trigly-cerides, total protein content, urea, alkaline phosphatase activity and increases in serum cholesterol were noted. In the 50 g/kg body weight/day group of rats, the highest and lowest decreases in biochemical parameters were observed, respectively. The exposure dose of 100 mg BPS and BPA in our experiment did not cause liver histopathological changes or changes in biochemical parameters according to the hy-pothesis. A possible explanation could be given by the carrier for which sunflower oil with natural antioxidant content was used.
Pathology of adnex

The term adnex comes from the latin word nexus or connection and i tis used in gynecology and dermatology. Gynecological adnexa involves the internal genitals of woman without uterus, so it involves the fallopian tubes and ovaries. In every woman´s life, there can be conditions when the woman feels abdominal pain. These abdominal pain can´t be serious, however if the pain persists, it needs to be examinated their origin. During diagnostics, it is also important to keep in mind that the woman is prone to various diseases associated with gynecological adnexa. This bachleor thesis deals with pathological conditions od gynecological adnexa, therefore inflammation of the fallopian tubes and ovaries, ovarian cysts, adnexal endometriosis and benign but also malignant neoplasms located in the fallopian tubes and ovaries. Because every woman is at risk of an ectopic pregnancy when she is of childbearin age, this type of pregnancy is considered as pathological, so it is included in this bachleor thesis as well. Forty samples of gynecological adnexa are processed for this thesis. A selected part of the adnex sample is documented in this thesis. Samples are processed in all sections of the histological laboratory. After that histological preparations are made and the diagnosis is made too. Histological preparations are stained with basic histological staining Hematoxylin-eosin, but also with special methods Hematoxylin-van Gieson and Goldner´s trichrome. The thesis also contains a statistical evaluation of the incidence of gynecological adnexa for the period from 2015-2019, which were diagnosed at the pathological department. Female sterilization is included in this evaluation too, because its number has increased over the period. The sterilization included in the evaluation was always performed with the patient´s consent and at her own reques, for example it is performed during the planned caesarean section.
Histological examination of the colon

This bachelor´s thesis deals with histological examination of the colon, namely the laboratory diagnosis of colorectal cancer. Colorectal cancer is a highly relevant issue today not only in the Czech Republic, not least because of its continually growing incidence worldwide. Laboratory analysis of this disease is not only important from the aspect of diagnosis per se. Determining the type and invasive stage of the disease is also crucial for the therapeutic strategy in a given patient. Immunohistochemistry testing of the colon also aids in the diagnosis of Lynch syndrome, a hereditary disease with a high risk of colorectal cancer. The first part of the thesis is theoretical and aims to introduce not only the subject of colorectal cancer but also the anatomical, histological and pathological features of the colon. This section also describes the methods used to obtain tissue samples from the colon and to process these in a histopathology laboratory. This information regarding the histopathological processing of colon tissue is then applied and described in detail in the methodology section of the thesis. This describes the process of routine staining methods such as haematoxylin-eosin and Alcian Blue. It also details the manually performed immunohistochemical methods and related reagents, including the use of antibodies. The next section presents a statistical analysis of a sample of patients with colorectal cancer treated at the Jihlava Hospital between 2018 and 2019. These data confirm the generally accepted fact that there is a higher incidence of colorectal cancer in men and that this cancer predominantly affects patients aged between 60 to 70 years. Furthermore, this work shows that defects in the MMR proteins associated with Lynch syndrome are not detected in even one half of the patients tested.
Immunohistochemical and histological correlation of melanoma lesions

The bachelor thesis deals with the problematic of melanoma lesions. This theme is actual because melanoma is the most serious type of skin cancer and its incidence is moving to the younger generation. Because melanomas occur primary due to ultraviolet light exposure, one of the best possible ways to reduce the risk of developing melanoma is to protect yourself from ultraviolet radiation by using the sunscreen, wearing clothing that covers arms and legs and avoid using tanning beds. The prognosis of early melanoma is usually very good, so the detection and treatement is very important. In the theoretical part the current knowledge of melanoma, the characteristic, the occurence, the clasification, the prevention and the current possibilities of treatment were summarized. In the practical part the Hematoxylin eosin staining and the immunohistochemical method was described. There is also comparation of positive HMB-45 marker between each histological groups. The HMB-45 marker is absolutly specific for melanocytic tumors and has great utility fot the pathologist in differentiation of tumors with uncertain origin. From 2012 to 2018 were imunohistologicaly examined at the Departement of Pathology Nemocnice Písek a.s. 61 samples. Out of total 61 samples were 58 HMB-45 positive. The thesis has been focused on the imunohistological profile of each histological group. The imunohistologichemical profile was proven in 92%.
Postnatální změny ledvin králíka

The kidneys are the main excretory organ that completes morphological and functional development in a species-different postnatal period of an individual's life. This bachelor thesis dealt with morphometric and histological changes in the kidneys of rabbits. The study included 42 New Zealand white rabbits at the age of 0, 5, 10, 14, 19, 27, 32 days. During this period, the length, width and weight of the kidneys and histological changes were determined, and at the age of 14, 19, 27, 32 days also the length, width, area and number of renal corpusles. In newborn rabbits, the average kidneys length was 9.75+-0.66 mm and the width was 5.88+-0.78 mm. Over 32 days, length increased by 117.44% (21.2+-1.71 mm) and width by 144.90 % (14.4+-1.61 mm). The most significant increase in kidneys length and width occurred in the first 19 days of life (length 110.26 %, width 144.56 %). The average kidneys weight of the newborn rabbit was 0.25+-0.03 g and 2.40+-0.83 g on day 32. A nephrogenic zone and nephrons type I and II were observed in the kidneys up to 10 days of age. By day 19, nephrons type III were present. The onset of marrow differentiation into inner and outer zones was noted at 10 days and at 14 days the zones were fully distinguishable. In 14-day-old individuals, the average length of renal corpusles was 55.10+-6.32 um, width 67.51+-8.91 um and area 3125.25+-778.99 um2. In 32 days, the length reached 76.90+-6.26 um, the width 93.96+-6.10 um and the area 5963.06+-851.05 um2. High correlation coefficients ranging from 0.886 to 0.972 were found between body weight and renal morphometric parameters. High correlation coefficients were found also between the weight of the kidneys and the morphometric parameters of the renal corpuscles. A negative coefficient was found between the area and the number of renal corpuscles (rxy = -0.997).
