Determination of selected bioactive substances in grapes

The subject of this thesis was the determination of selected bioactive substances in grapes and the subsequent comparison of differences between the samples. Two fractions of grapes (skins and pulp with seeds) of 4 grape varieties - Hibernal, Johanniter, Riesling and Aurelius - were analysed. Two methods were used for the determination: for phenolic substances, high-performance liquid chromatography with mass spectrometry and for volatile substances, headspace solid-phase microextraction coupled with gas chromatography and mass spectrometry. For each of these methods, the optimal extraction technique was first selected and then used for the analysis of all samples. The linearity of the external standard method was also verified with a reliability coefficient R2 in the range of 0.9650-0.9989 for high-performance liquid chromatography with mass spectrometry and 0.8571-0.9999 for headspace solid-phase microextraction coupled with gas chromatography and mass spectrometry. 63 volatile and 9 phenolic compounds were identified in the samples. From these, specific compounds of grapes (20 volatile compounds and 5 phenolic compounds) were selected. On the basis of the amount of observed compounds, the two grape fractions and subsequently the grape varieties were compared with each other. There were differences in the content of selected compounds between grape fractions and across varieties, which could probably be due to the geographical origin of the samples.
Viticultural tradition in Bratislava

Viticulture has long belonged to Bratislava. It has been mentioned in archival reports since the middle of the 13th century. The vineyards stretched from Bratislava Castle to Rača and Vajnor. Especially in the last century, large-scale construction of apartment and family houses began to replace vineyards. This trend still persists and developers attract people to live with a view of the city and the proximity of nature, from which, however, they take themselves. The vineyards have been receding for the centuries of the developing city and this historic heritage is falling into oblivion. however, vineyards are not only a reminder of the glorious past, but also of irreplaceable creators of biodiversity and contribute to a better climate for the city. The aim of the diploma thesis is to analyze the retreat of vineyards expanding in Bratislava. Conditions for vine growing and its processing in Bratislava and its surroundings. Subsequently, finding a suitable form and applying the outputs from the analytical part in a specific architectural design.
The stum and its utilize

In a bachelor's thesis are described characteristics of wine texture, wine grapes and stum like a natural products that have a beneficial health influence. In this thesis is briefly described a technology of processing of wine grapes too. The bachelor's thesis is concentrated on the possibility of the use of stum and making products for wholesome nourishment. It is effective in its use with a combination of elderberry juice. A practical part has been made an acceptable testing procedure of taste, with various concentrations of each ingredient (stum and elderberry juice), being used in order to chose an appropriate option of product that will have a healthy beneficial influence.

The bachelor's thesis is based on an architectural study of the subject Atelier's work AG032 and a design study of the subject of the Comprehensive Project AG036. The award contained several requests from the investor. The subject of this bachelor thesis is the design of a new-build winery in the village of Novosedly. Sorted parcels with parcel numbers 5844/2, 5764/29, 5764/55 and 5764/3 on which the building project is located and is located in the cadastral territory of the municipality of Novosedly. The main access to the property is from the main road 414 and further along the asphalt road, which also serves as a cycleway. The building is situated near the property boundary, i.e. its northwest side. From the very situation emerges the basic concept of the project, which is to try to build a new domination of the village, while respecting the surroundings of the vineyards. One of the goals is to combine production, tasting and accommodation into one complex whole. The final design was influenced by the view of long-distance views from the area to the surrounding area (on Pálava) and another equally important one, by the creation of a resting place and meeting of people who go specifically for wine tasting or a selfless view of the surrounding area. The proposed building is two-storey, partially sunk in the terrain, when I took advantage of a large high jump in the terrain. On the first floor there is wine production and storage for subsequent export or preservation of wine. On the second above-ground floor there are areas for visitors, namely tasting and accommodation. A characteristic feature of the building is the saddle roof, which floats on the Old Mountain, thanks to considerable glass and lightening by means of columns. An important point of the proposal is the creation of a closed but open space (courts).
Wine Farmstead in Jaroslavice

Winery of Markel family is located on the border of small South Moravian village Jaroslavice. The area is surrounded by vineyards, fields and an original castle peasantry. The area is heavily influenced by insensitive exploitation of a former brickyard. A footprint of the exploitation apparent at first glance has remained in the relief. On one hand a scar, on the other a fascinating geological spectacle of an up to 10 meters high earthen-loess wall. The spacious and various area becomes a winery once again.

The diploma thesis is focused on determining the influence of transport on the content of heavy metals in white and blue grape varieties and their subsequent products from vineyards near traffic-laden roads and reference, traffic-free locali-ties and its relation to heavy metal content in air and soil in monitored vineyards. The analysis of elements in grapes and their subsequent products is performed in various stages of wine production, air analysis by analysis of weekly sampling and soil analysis by one-time sampling in the monitored vineyards in the period befo-re harvest. The instrumental method of inductively coupled plasma mass spectrometry was used for the measurement.
