Dependence of derived fibrinogen on values of DH from APTT and QUICK(PT) methods

Hemocoagulation, blood coagulation, is an important indicator of hemostatic balance in the human body. There are many ways how to investigate blood clotting. In practice, next to the tests investigating time of coagulation cascade from view of internal way (APTT – activated partial tromboplastin time) and external way (PT – prothrombin time) is often used determining of fibrinogen concentration by Clauss method. Derived fibrinogen method determined fibrinogen concentration, too, by subtracting form the clotting curve in PT test. The reaction for Clauss method is not necessary here. Derived fibrinogen is not used much in practice. This is the reason, why the thesis related to this project will try to find relationship between concentration of fibrinogen and standard tests APTT and PT. Clinical data will be used for this.
The PCR method optimization for analysis of the gene PAI-1 polymorphism

The plasminogen activator inhibitor (PAI-1) is an important biomarker of fibrinolysis. This glycoprotein acts as a principal inhibitor of plasminogen activators and its high plasma level is associated with various types of diseases, such as cardiovascular diseases or malignant tumors. The PAI-1 gene is located on chromosome 7 in region 7q22.1. In 1993 4G/5G polymorphism was discovered in the gene promoter. The 4G allele increases PAI-1 gene expression which results in a higher plasma level of this inhibitor. That's why this polymorphism has become the subject of many studies. The main aim of my bachelor thesis was to optimize the ARMS-PCR method for analysis of the gene PAI-1 4G/5G polymorphism. Other goals were to write a specialized research on this subject and to learn the techniques used in the genetic laboratory (DNA isolation, DNA concentration measurement, Real-time PCR). In the theoretical part I describe the PAI-1 gene and its product. I deal mainly with the role of this inhibitor in the mechanism of fibrinolysis and its influence on various diseases. Furthermore, I showed an overview of methodological approaches used for genetic analysis of 4G/5G polymorphism in PAI-1 gene. In the practical part there are presented procedures of optimization of the ARMS PCR method and the results of the genotypes of the examined group obtained by the Real-time PCR method, when a commercial kit was used. The optimization of the ARMS-PCR method was not successful.

D-dimer concentration in pregnancy

D-dimers belong to fibrin degradation products (FDP) which we can find in the blood when the blood clot is dissolved. This process is named fibrinolysis. The levels of D-dimer are higher in all conditions connected with increased coagulation activation like deep venous tromboembolis, pulmonary embolism or disseminated intravascular coagulation. So the negative results, when the D-dimer concentration is below predetermined threshold, are one of the useful ways how to rule out tromboembolic disease The problem is that the elevated D-dimer levels are observed in other conditions like infection, trauma, acute cancer, recent surgery or pregnancy. The thesis should confirm that the concentration of D-dimer in pregnancy is elevated and the D-dimer levels were established in particular trimesters. These are compared with the D-dimer levels described in contemporary literature. The levels are determined for two different tests which were used in Hematology laboratory in České Budějovice hospital at different times. Besides the viewpoint of D-dimer and gravidity the thesis also follows up different results of the two tests and their possibility of comparison.
