Determination of polybrominated diphenylethers in matrices from fireplaces

Polybrominated diphenyl ethers belong among the persistent compounds, which have been classified as priority organic pollutants. In environmental compartments are observed in the past decade. Polybrominated diphenyl ethers are detected in abiotic and biotic matrices. For this dissertation were chosen the matrixes from seats of fire, taken in various localities of the Czech Republic and Slovakia. Their analysis should demonstrate whether they in these specific matrices remain. There were examined following polybrominated diphenyl ether congeners: BDE- 28, 47, 99, 100, 153, 154, 183. For their isolation from the matrix were used three different extraction techniques, namely ultrasonic extraction, microwave extraction and pressurized solvent extraction. For the determination was chosen method of GC / ECD. In this dissertation are also described basic chemical, physical and environmental properties of BDE, including other analytical methods that can be used to determine polybrominated diphenyl ethers in environmental compartments.
Developement and optimalization of methods for extraction of selected compounds from Hypericum perforatum

Bachelor's thesis deals with the optimization of processes for extraction of phenolic compounds from St. John´s Wort (Hypericum perforatum). The theoretical part of the work focuses on characterization of this herb, its botanical classification, traditional use, chemical composition and curative effects. Increased attention is further devoted to a theory of preparation of samples for extraction of phenolic compounds and suitable techniques for extraction and characterization of phenolic compounds from extracts of St. John´s Wort. The experimental part of the thesis deals with application of theoretical knowledge. When optimizing the emphasis was on influence of solvent (water acified to pH 4, pH 5, pH 6), influence of extraction technique (ultrasound extraction, subcritical water extraction, maceration) and influence of extraction period (10, 20, 30 min). Extracts were subjected to determination of selected parameters (total phenolic content, total flavonoid content, antioxidant activity) in leaves of St John´s Wort harvested in June 2016 using UV-VIS spectroscopy. The used extraction techniques are significantly different from each other.
The monitoring of agent BZ after intramuscular administration

The compound 3-quinuclidinyl benzylate (agent BZ) belongs to the group of incapacitating warfare agents with anticholinergic activity. Today, for its ability to induce functional cognitive impairment (i.e. coordination and memory disorders), the agent BZ is used mainly for scientific purposes in the study of Alzheimer's disease. Despite this fact, its pharmacokinetics has not been fully examined yet. In order to determine the agent BZ in biological material, LC-MS / MS method and sample preparation procedure for body fluids (plasma, bile) and tissues (brain, liver, kidneys) were developed, optimized and validated. The sample preparation procedure for body fluid employed solid phase extraction using a C18 column eluted with methanol and for body tissues it was precipitation with acetonitrile. The chromatographic separation was performed on Gemini NX-C18 reverse phase column (150 × 4.6 mm, 5 μm). The mobile phase consisted of a 10mM solution of ammonium acetate at pH 11 and methanol in a ratio of 30 : 70. Elution of the analytes was performed under isocratic elution. The total analysis time was 5 minutes. Mass spectrometric detection was performed by a linear ion trap using electrospray ionization. Recovering of Viticulture Wastes Into Value-added Products.

Viticulture has a very rich tradition in the Czech Republic, especially in South Moravia. During the production of wine, as in any other industry, different types of wastes are produced. These are, for example, the remains of must-pressing, the so-called grape marc, which is usually used on the compost or the wood mass produced by the pruning of the vineyards, called grape cane, which are pressed into wooden briquettes. Within our project, we are dealing with the possibilities of combining extraction techniques for the treatment of grape marc and grape cane in order to obtain extracts rich in biologically active substances (resveratrol, viniferine).
