Preclinical models of brain metastases

Malignant diseases are the second most common cause of death in the Czech Republic. One of the serious complications of these diseases are brain metastases, the incidence of which is increasing due to the improvement of diagnostics and advances in the therapy of the primary tumors. However, the treatment of brain metastases is complicated by their localization and the presence of the blood-brain barrier, which affects the efficacy of many drugs. Preclinical models are essential for better understanding of the biological properties of metastasizing cells, their relationship to the unique microenvironment of brain tissue, and for testing new therapeutic approaches. This bachelor thesis focuses on in vitro and in vivo systems modelling key aspects of brain metastasis biology and their use in testing new therapeutic approaches.
Cytotoxicity of selected naphthoquinones on prostatic cell cultures

This master´s thesis discusses cytotoxicity of selected naphthoquinones on prostatic cell cultures. The introductory part is dedicated to general characteristic of naphthoquinones with focus on their cytotoxicity, testing of cytotoxicity and mechanisms of cytotoxicity. This part is followed by chapters about cytotoxicity, characteristics and biological activities of selected naphthoquinones; plumbagin and naphthazarin. The last part of this thesis’ theoretical section speaks about fluorescence microscopy and its use in research of naphthoquinones cytotoxicity. The practical part is dedicated to evaluation of cytotoxical tests’ results and to analysation of pictures of cells obtained by fluorescence microscope. At the end of thesis, all finding are summarized and put in the context.
Use of cell cultures to test cytotoxicity of natural substances

This thesis is aimed on study of cell cultures, primarily on introduction of techniques for cultivation of human cells and optimization of their usage in cytotoxicity tests. The thesis is aimed preferably on natural substances and liposomes that are suitable to be used in cosmetics. Theoretical part contains literature review focused on overview of cell cultures, appropriate cultivation conditions for in vitro testing and summary of cytotoxicity tests. Experimental part contains characteristics of natural extracts with antimicrobial effect in terms of content of polyphenols, flavonoids and antioxidant activity. Further, these extracts were encapsulated into the liposomes. Encapsulation effectivity was determined. Distribution of particles size by dynamic light scattering and finally also stability of prepared particles were also determined. Long-term stability in water and gel environment was tested too. This thesis also contains study of optimization of cell culture of human skin keratinocytes cultivation and use of those cells for testing cytotoxicity of selected natural substances by MTT method.
Interaction of animal cells with organic semiconductors

This bachelor´s thesis was focused on study of biocompatibility of organic semiconductors. In introduction, organic semiconductors and their properties, benefits and features of selected representatives were described. In the following part biocompatibility definition and approaches for its assays including tests of cytotoxicity, material stability were presented. The last part was focused on overview of cell cultures, appropriate cultivation conditions and compounds of media for in vitro testing of biocompatibility. Experimental part showed the result of biocompatibility testing of materials. For this purpose murine 3T3 fibroblasts were used.

The bachelor thesis is devoted to testing the toxicity of chemical substances. In the current chemical and pharmaceutical industry, thousands of new substances are produced annually, which brings about the need to ensure sufficient consumer safety and environmental protection. In vitro testing is becoming an increasingly important research tool in this issue. This work verifies the theoretical foundations and proposes an algorithm developed by Lonhus et al. from the Laboratory of experimental complex systems in Nové Hrady and its possible future applications in in vitro testing. This new approach uses the principles of microrheology and tracks Speed-Up Robust Features (SURF) in a digital image and converts them into vectors of trajectories of intracellular mass particles. Microrheology is a relatively new field of research in life sciences and deals with the study of flow and deformation of materials at the microscopic level. The algorithm separates two main components of the movements inside the cells: direct movement of organelles as larger particles and random Brownian motion. The advantage of this method is its potential of easy use and sensitivity. The method makes it possible to obtain important primary information about the effect of chemical substances on cell cultures.
Purification of water soluble bioactive polysaccharides from biomass of heterotrophic mutant alga Chlorella vulgaris G11.

Water extract obtained from biomass of mutant alga Chlorella vulgaris G11 was deproteinized and then purified with AEC on DEAE Sepharose FF and SEC on Sepaphadex G-75 superfine columns. Two high MW (>1000 kDa and 98,3 kDa) polysaccharides (CPS1 and CPS3) were eluted with water and 0.5M NaCl on DEAE column, and heteropolysaccharide with MW of 15 kDa (CPS2) was also eluted with water on DEAE. The former fractions were identified as branched rhamnans, the latter one as branched xylogalactofuranan of unusual structure. CPS1 shown properties of unstable polimer: according to GPC analysis and by using light scattering and RI detectors it was found, that some fraction of polymers (260-800 kDa) have self-assembling properties in aqueous solution, and MW of this fraction decreased during some time while polimer with initially bigger MW (>1000 kDa) became bigger. CPS3 was eluted with 0.3-0.5 M of NaCl on DEAE Sepharose FF column and have MW of 98,3 kDa. Cell toxicity of the products were tested in vitro on cell cultures for different concentrations

Syntéza a vlastnosti chirálních acyklických nukleosid bisfosfonátů s otevřeným kruhem

A new type of acyclic nucleoside phosphonates (ANP) is derived from 2,4-diamino-6-hydroxypyrimidine. These 6-[2-(phosphonomethoxy)ethoxy]pyrimidine derivatives significantly inhibit replication of retroviruses and herpesviruses in cell cultures. This novel subclass of pyrimidine ANPs can be considered as analogues of 2,6-diaminopurine with an open imidazole ring in the purine moiety.
