Probiotics and prebiotics in food and other products

Theoretical part of this thesis focuses on present state and research of probiotics and prebiotics, their use for microflora modulation of host and their beneficial effects on the health of individuals. Furthermore thesis deals with efficiency of probiotics strains, which depends on the food matrix and other various factors. The experimental part focuses on the identification of chosen bacterial strain, which is contained in probiotics product. It is realized due the isolation of bacterial DNA by phenol extraction and by use of magnetic particles and subsequent analysis of obtained DNA by polymerase chain reaction (PCR).
Importance of probiotics in child's nutrition

With great advances in science, medicine and knowledge of a man at the high level of treatment. However, even today the humanity seeking to return to its roots - the nature and use it to maintain its good health. The term probiotics is meant by living organisms that are capable of resettling in the human intestine and have a positive impact on maintaining healthy intestinal microorganisms, which also reflected the positive effects on human health in general. Company Valosun a.s. provided for testing probiotický food supplement for children, which was tested mothers with children preschool age and pregnant. The aim of our sociological survey was to determine what is the awareness of the issue of probiotics in this group, the perception of the queried the availability of probiotics and what are the most common reasons for their use.
Recombinant probiotics

Theoretical part of this thesis focuses on present state and research of recombinant probiotics and their use in food industry and health care. It also concentrates on their beneficial effects on the health of individuals. The experimental part focuses on the identification of specific bacterial strain in a probiotics food supplements. The DNA was isolated from these products by use of magnetic microparticles and obtained DNA was subsequently analysed through polymerase chain reaction.
Development of probiotic dietary supplement

The thesis deals with a theme of probiotic mikroorganisms which are aplicated as a dietary supplement. Health benefit on the host have been known for a long time, however, there might be some negative effects affecting a consumer. In connection with this observation defining requirements and evaluating of safety of probiotics in food was necessary. The new probiotic dietary supplement can not be expanded on the market without these processes. Every strain must be tested separately on each property. The ability of rezistance against conditions inside human gastrointestinal tract belongs to the most basic tests. The aim of the practical part is testing this ability of rezistance in vitro by simulated conditions. The model of gastrointestinal juices was designed according to the Czechoslovak codex. The form contributes to the higher rezistance of probiotics within implementing to the host. Also a dosage form of probiotics was involved to a in vitro testing.
The used of magnetic microparticles for isolation and prove of probiotic bacterial DNA in meat

This thesis deals with the isolation of probiotic DNA from meat products and its assesment by PCR methods. In this thesis is developed homogenization of samples of sausages with kopist, preparation of sausage cells lysates and isolation of DNA by using of magnetic microparticles. The DNA was isolated from sausage lysates by using magnetic microparticles. Isolated DNA was further amplified in genus and spesies-specific PCR methods. In tested products was proven presence of DNA of domain Bacteria, type Lactobacillus and Bifidobacterium. In one product was proven presence of species Lactobacillus acidophilus and Bifidobacterium animalis.
Probiotics and their importance in human nutrition

A great deal of immune system is located in the colon thanks to a large number of microbs that influence not only digestive tract of a human. The contact of these microbs with the digestive tract of a human is very important for other development of immune defensive mechanisms and the natural immune response, which supports health of a host. That is why in recent years a great deal of attention and efforts have been paid to possibility of human health status improving by a modulation of the intestinal microflora. As probiotics are indicated living microorganisms, that can have positive effects on human health and some species, for example Saccharomyces boulardii, have positive effects in prevention and treatment of many diseases associated with diarrhea. The company Valosun a.s. has provided for testing the probiotic food supplement which is suitable in acute diarrheal disorders. This product has been tried by patients of gastroenterology ambulance. The aim of sociological survey was to map knowledge of respondents in the field of probiotics and probiotic food supplements.
Probiotic foods

This bachelor thesis summarizes the knowledge about problematic of probiotics and their positive effects on human health. Probiotics are live microorganisms, which, at application in appropriate quantity as component of food, have health benefit in humans and animals. Probiotics are already an important component of treatment in gastroenterology, in hepatology, at treatment of urogenital system, at prevention and therapy of cancer and mycoses. The best known probiotics are lactic acid bacteria, of which most often used are Lactobacillus and Bifidobacterium genera. Lactic acid bacteria because of their unique metabolic characteristics are involved in many fermentation processes of milk, meat and vegetables. We can say, that advances in probiotic bacteria research are beneficial for both the consumers and the producers.
Identification of selected probiotic bacteria in food additives

Bifidobacteria and lactobacilli are gram-positive bacteria which belong to the lactic acid bacteria group. They are ordinary constituents of gastrointestinal microflora, play a pivotal role in human nutrition and health, prevent pathogen colonization and maintain normal mucosal immunity. In present days they are used in production of functional food products and pharmaceutical additives. Polymerase chain reaction (PCR) is used to the detection and identification of bacteria which belong to the genus of Bifidobacterium and Lactobacillus. Specific primers for each genus are used in PCR reactions for in vitro amplification of definite part of DNA. In this work, total DNA was isolated from three additives (Pangamin Bifi plus, Biopron Junior and Probiodom) by the method of phenol extraction and amplified using genus-specific PCR primers giving amplicons of 523 bp (Bifidobacterium) and 250 bp (Lactobacillus). Occurrence of bacteria of both genera was proved in all of the three tested probiotic products.
Validation of probotic bacteria presence in fermeted plant food products

Probiotic bacteria are an essential part of human nourishment which provide many health benefits. We consume them not only in traditional dairy fermented foodstuff, but also in the ever-expanding fermented plant products. The theoretical part summarized general knowledge about probiotic microorganisms, individual species used in technology of processing plant and dairy fermented products, and their influence on human health. The following chapters are devoted to various methods of identification of probiotic bacteria with emphasis on molecular diagnostics, specifically by PCR method. The experimental part is focused on the identification of lactic acid bacteria in a plant fermented product – in a fermented cabbage using 2 methods of isolation bacterial DNA and subsequent analysis by PCR method.
Isolation of PCR-ready DNA from probiotic products for baby nutrition

The aim of thesis is focused on isolation of DNA in quality for polymerase chain reaction (PCR) and the identification of probiotic bacteria. From six probiotic supplements for children were isolated PCR-ready DNAs using magnetic carriers P(HEMA-co-GMA). Isolated DNA was amplified by genus-specific and species-specific primers. DNAs of Lactobacillus, Bifidobacterium and Streptococcus genera were identified as: L. acidophilus, L. rhamnosus, L. casei, B. bifidum, B. longum ssp. longum, B. breve, B. longum ssp infantis, B. animalis and S. thermophilus. The identification corresponded with the data declared by the producers.
