Static strength assessment of the weld
Gális, Róbert ; Řehák, Kamil (referee) ; Dlugoš, Jozef (advisor)
This thesis aims to review, summarize and compare the available approaches and standards assessing static strength of welds. In the introductory section, the theoretical knowledge required to assess static strength of welded structures is summarised. In the next section, the algorithms for evaluating static strength of welds using each guideline are reviewed and described in detail. They are also applied to designed T-shaped welded geometry with a two- sided fillet weld. Subsequently, the obtained results are compared and evaluated. Finally, an experiment is drafted to verify the calculations or to determine the most appropriate approach.
History and acoustics of traditional timber joist floor structures
Kalousek, Lubor ; Fišarová, Zuzana
The article is focused on the issue of traditional timber joist floor structures, especially from the end of the 19th century to the first half of the 20th century. In common building practice, we very often encounter these structures in residential buildings that are, or were in the past, the subject of structural changes. Unfortunately, during these interventions, the basic ideas of the original technical solution are not always respected and some physical parameters of building are degraded, of which the requirements of building acoustics will be discussed in particular. However, the problem includes not only the actual composition of timber joist floor structure, but also the floor, which forms an integral whole in interaction with it. The article will describe the principles that were devoted to these types of structures in historical legislative and standard regulations and further in professional literature. The original historical solution will be presented on a practical example. In relation to building interventions in historic timber joist floor structures, current legislative and standard regulations will be described, as well as examples of modern solutions and their limitations.

Characterization of eukaryotic translation initiation factor 3 subunits (eIF3) in A. thaliana male gametophyte
Linhart, Filip ; Hafidh, Said (advisor) ; Retzer, Katarzyna (referee)
From RNA-to-protein, translation initiation and protein synthesis is mediated by trans-acting factors that recognize mRNA features common to almost all eukaryotes. Eukaryotic translation initiation factor 3 complex (eIF3) is a highly conserved protein complex that recognizes 5'-CAP elements of the mRNA to initiate translation. eIF3 consists of nine subunits, three of them having two isoforms: eIF3A, eIF2B1, eIF3B2, eIF3C1, eIF3C2, eIF3D, eIF3E, eIF3F, eIF3G1, eIF3G2, eIF3H and eIF3K. This work deals with functional characterization, expression and subcellular localization of eIF3B1, eIF3B2 and eIF3E in Arabidopsis thaliana male gametophyte and interaction of eIF3E with the Constitutive photomorphogenesis 9 (COP9) complex as a regulatory complex of eIF3E post-translational control. Here we show that depletion of eif3b1 or eif3b2 is not gametophytic lethal and that the two protein might function redundantly, whereas, knockout of eIF3E causes male gametophyte lethality. Interestingly, eif3b1 show post-fertilization defects during embryogenesis, suggesting that its redundancy with eIF3B2 is restricted to the gametophyte. Gene expression studies revealed high expression of eIF3 subunits in actively dividing zones of leaf primordia, root meristem and root elongation zones as well as in the vegetative...
