This bachelor's thesis deals with a business plan processing for a private nursery school establishment in Brno. The partial goal is to design this business plan funding. The first part summarizes initial theoretical knowledge needed to work out the business plan. The practical part analyzes the environment, and describes a specific business plan processing - suggests possibilities for funding and evaluates the best option.
In vitro 3D organization of the mammalian testes

A testicular organoid that would sufficiently recapitulate the architecture of the testicular tissue and at the same time be able to provide the complete process of spermatogenesis in vitro has not yet been created. Achieving this goal would mean the development of a 3D model of the testis, which would mimic the in vivo situation in terms of cell arrangement and could thus contribute to a deeper understanding of the physiological functioning of the testicular microenvironment. Among other things, such a model has a great potential for clarifying the causes of male infertility and finding treatment options. This thesis dealt with the generation of organoids from mouse or human testicular cell suspensions, which can also be used, for example, to study de novo organogenesis. A total of four 3D culture systems were tested, of which the soft agar culture system (SACS) achieved the best results. Furthermore, as part of this thesis, the procedure for preparing testicular organoids for light sheet microscopy was successfully introduced and optimized, enabling the evaluation of their internal structure. Folate and its role in early embryonic development of higher vertebrates.

Folate or vitamin B9 is an essential water-soluble nutrient that takes part in important cellular processes. These include amino acid metabolism, methylation, and nucleotide synthesis, the last two of which play a key role in early embryonic development. It was approximately 30 years ago when folic acid supplementation was confirmed to help prevent embryonic neural tube defects and since then an intake of 400 µg of folate a day has been recommended to all women of childbearing age. Despite this, folate deficiency is a relatively common phenomenon and during pregnancy can lead to congenital malformations and pregnancy complications. This bachelor thesis summarizes the latest findings on the association of neural tube defects and miscarriages with single-nucleotide polymorphisms in genes encoding folate metabolism enzymes in humans, focusing mainly on the gene encoding the enzyme 5,10-methylenetetrahydrofolate reductase (MTHFR). Furthermore, this thesis deals with potential mechanisms of the folate's protective effect which could participate in the prevention of neural tube defects and are studied mainly on model organisms. Education to reading through work with book in pre-school age

Baccalaureate work attends to problems of the relation of pre - school child to book. Theoretic part deals with reader's literacy, possibilities of activities of pre - school arrangements fuelling the reader's literacy of childern and complexity of activities supporting aesthetically development of child in pre - school age. In practical part was realized thematic oriented project built on arranged meetings with a fairy tale. The purpose was to create and strengthen the relation of child to book. In work are further presented results of controlled talks and singhtings, on whose base was analysed present - day (nowday's) reader's level of families of hereof sample.
