Microbiological analytical methods suitable for dairy industry

The theoretical part of the thesis is focused on probiotics in dairy products. Thesis deals with molecular genetic methods, which are used to analyze DNA. Especially are discussed methods used for isolation bacterial cells belonging to genus Lactobacillus. The polymerase chain reaction (PCR) is the basic technique at present time. Short chain of oligonucleotide primers are used to amplification specific parts of DNA molecule chain. The practical part of the thesis is focused on the DNA from pure bacterial culture and probiotic dairy product. DNA was isolated using methods of phenol-chloroform extraction and magnetic separation. For magnetic separation was used magnetics particles covered with carbonyl functional groups. Quality of the DNA was confirmed by PCR amplification. Primers specific for domain Bacteria and genus Lactobacillus and Bifidobacterium was used.
Melody Harmonization

This bachaleor's thesis is focused on an automatic harmonization of melody. The implemented system is using a machine learning approach and learning the harmony principles from the database of MIDI files. The input of the system could be monophony songs in MIDI or ABC format. The output of the system are polyphony songs in MIDI format harmonized in the style of protestant chorale.
Residential Construction as a Development Project

This thesis is concerned with developer project Rezidence Austerlitz in Slavkov u Brna realized by KALÁB company. It describes managing this project from buying the grounds to selling the real estate to clients. At the same time the thesis deals with the project's financing, time schedule and assesses market and construction risks. Part of the work is also analyzing the estate market which adverts to evolution of housing construction across the whole Czech Republic in the past ten years. Progress of real estate construction in Jihomoravský kraj is described in detail, including the progress of estate valuation.
Differences in the Driver's Behavior when Driving through a Pedestrian Crossing at Night and in the Daytime

The theoretical part of the diploma thesis focuses on the drivers reacting on a pedestrian crossing during the day and the night. A particular attention is paid to all the factors that can influence drivers' behaviour while driving through a pedestrian crossing. The influence of the behaviour by these factors often causes car accidents with pedestrians. One of the basic techniques is to analyze the behavior of the driver in the optical perception of stimuli that affect the optical response. Eyetracker is used to measure the optical responses. This device monitors changes of the angle of a driver's view, navigation of a driver and the driver's reaction on different stimuli while driving. The practical part of the thesis deals with an analysis of the drivers' behaviour while driving through a pedestrian crossing during the day and during the night. Twelve drivers took part in the day measurement of driving through the pedestrian crossing in Brno. Night measurement was taken place in Břeclav and Lednice with the attendance of seven drivers. During the ride are monitored the optical reactions of drivers on various impulses with the use of a special device Eyetracker.

This thesis deals with the development of a client-server application for Red Hat company. Client participates in testing process of JBoss platform and gets user-defined performance data which sends during testing to the server application by platform independent communication. The server application allows to analyze the received data which can be also compared by several perspectives. These services for data analysis and comparison are accessible for server users using the web client. The server application supports the import of the performance data stored in the XML file and also their export for qVue portal. The client part of the application is implemented in Java and the server application is based on Java EE platform.
The separation efficiency of air pollutants on pilot air scrubber

This diploma thesis deals with the separation process of air polutant on air scrubber. Absorption of carbon dioxide was carried out into 1 % solution of sodium hydroxide. Theoretical part is aimed at mass transportation, theory of absorption and examples of scrubbers. Experimental part focuses on some operating characteristics of scrubber and on optimisation of two-phase flow. The influence of hydrodynamic conditions on the absorption efficiency was found.
The reasons for cooperation with external consultants

The objective of this diploma work was to form hypothesis about external consultant's cooperation. The hypothesis was based on personal practical experience working on projects with external consultants and also being in role of consultant personally. The research with limited extent was performed related to the theme of diploma work. Evaluation of formed hypothesis was performed and the results were linked to the theory of organization. In the theoretical part of the work the history of the sociology of organization and management consulting have been listed and described. Based on personal practical experience cooperating with external consultants and working in the role of external consultant as well I have suggested possible reasons for hiring external consultants. I have performed research with objective to identify subjective opinions about suggested reasons hiring external consultants.
