Aminocarbonydrates as part of cyclic polyamines

Magnetic resonance imaging is one of the imaging methods that are needed in medicine for diagnostic purposes. This method neither exposes the patient to X-rays, nor any other negative effects on the human organism are known by then. Another significant advantage of MRI is its ability to achieve high contrast even between structurally very similar tissues. The contrast agents commonly used with the magnetic resonance imaging help heighten this ability. Part of these contrast agents is a metal ion, the ion gadolinium (GdIII) is the most commonly used nowadays. But it must be complexed by a suitable ligand. This work deals with the synthesis of a number of potentially suitable model ligands. The saccharide component is even a part of these ligands. This component should enable to achieve a greater solubility of the complex or the possibility of linking other additional saccharide units, another ligand or a detecting group. Several theoretical paths leading to the final fabric were created. Not all of them led to the target, only one was successful. The produced macrocycle is the potentially applicable ligand for complexation with GdIII or other metal ion, it means a good part of the new potential contrast agent.

ANGLICKÝ ABSTRAKT Functional study of the putative nucleotidase encoded by spr1057 gene in Streptococcus pneumoniae, a likely homolog of Escherichia coli protein YjjG. Bacterial cells are constantly exposed to innumerable toxic substances, either in their external environment or by by-products of their own metabolism. For these reasons, the bacterial cells evolved several mechanisms to cope with this challenge. These mechanisms are represented by: blocking the uptake, export by specific transporters as well as specific inactivation of these substance by enzymes. A particular group of these toxic substances are noncanonica nucleotides, which can directly inhibit bacterial cell DNA replication or can result in increased mutation rate. Enzymes recognizing these modified derivatives are known as "house-cleaning" nucleotide phsphateses, which can inactivate the potentially mutagenic nucleotides and prevent their incorporation into DNA and RNA. Some of the "house- cleaning" enzymes belong to a group of haloacid dehalogenase enzymes (haloacid dehalogenase-like hydrolase superfamily), which are found in many bacterial species. Study of Platinum Cytotoxic Drug Interactions with ABCG2 Membrane Transporter

Petr Bouška Consultant: PharmDr. Martina Čečková, Ph.D. Title of Thesis: Study of platinum cytotoxic drug interactions with ABCG2 membrane transporter Platinum cytotoxic drugs pertain among most frequently used cytotoxic drugs. However, their usage is complicated with development of resistance which can be caused by few mechanisms. The aim of this study is to evaluate possible interactions of cisplatin, carboplatin and oxaliplatin with ABCG2 membrane efflux transporter which causes resistance to many cytotoxic drugs. Hoechst 33342 assay was performed on cell lines MDCKII (parent cell line) and MDCKII-ABCG2 (cell line genetically modified for expression of human ABCG2 gene). Intracellular concentration of Hoechst 33342 was not increased in presence of any tested cytotoxic drug; significant change of Hoechst 33342 intracellular fluorescence was observed only in presence of ABCG2 control substrate mitoxantron and ABCG2 inhibitor fumitremorgin C. Final results did not demostrate interactions of ABCG2 with any studied platinum cytotoxic. It shows insignificant interaction between ABCG2 and platinum cytotoxic drugs and so improbable effect of this transporter on toxicity reduction...
The coal mining and its influence on the environment from the legal point of view

ENGLISH SUMMARY Coal mining is an important activity which significantly contributes to the economy of the Czech Republic. Coal is currently still indispensable material for many industrial branches, but coal mining has serious and wide-ranging consequences for the environment. Coal mining influences negatively especially the appearance of landscape, the soil conditions and the water system in the affected area. It causes air pollution and it leads to the destruction of fauna and flora, too. Furthermore the problem is that coal is a non-renewable material, which will eventually be exhausted. Due to these negative impacts on environment and the importance of coal as the source of energy, the coal mining requires state regulation. Legislation regulation of mining activity in our country can be found in these three acts: The Mining Act No. 44/1988 Coll., The Act of Mining Activity No. 61/1988 Coll. and The Act of Geological Works No. 62/1988 Coll. These three acts are supplemented with number of subsidiary regulations. This thesis attempts to describe and analyze basic legal instruments of environmental protection, which are exercised in mining. The main sources for this thesis are the three above-mentioned Acts and Acts regulating particular elements of environment for example the Water Code, the Forest Code...

The topic of my thesis I chose because for a long time I meet women damage to health caused by work . In his practice to rehabilitation department during the study physiotherapy, and then in daily practice , I have been in contact with women working in the past Rukavičkářských races Dobříš or currently working in smaller establishments engaged in the manufacture of gloves. The rehabilitation was running mostly for long-standing problems with musculoskeletal system . According to personal communication with the seamstresses were damage to health caused by work . I wondered which of musculoskeletal disorders really impairment of work are . Musculoskeletal disorders in fact appeared in women working in other professions. I deal with the question of what the seamstresses work gloves and How can it affect the emergence of musculoskeletal diseases. I focused mainly on carpal tunnel syndrome, as one of the occupational diseases (according to the current list of diseases occupational ) , in which the incidence in the last few years have seen a rising trend.
Esophageal achalasia - etiology, pathophysiology and treatment.

Background: Achalasia is a primary esophageal motility disorder that can be classified into three types (I-III) based on high-resolution manometry (HRM). Exact pathogenesis is unknown, but immune-mediated processes and genetic predisposition play a role, which is supported by finding of a genetic risk variant (rs28688207 insertion) in HLA-DQB1 gene that is strongly associated with achalasia. Per-oral endoscopic myotomy (POEM) has become a standard treatment for achalasia, but the long-term efficacy, safety and impact on esophageal physiology are not fully understood. The aims of our studies were to perform the first genotype-phenotype analysis investigating the frequency of rs28688207 accross three HRM types of achalasia, to evaluate the efficacy and safety of POEM and to assess the post-POEM esophageal motility patterns. Patients and methods: These were three retrospective studies of prospectively collected data. Genotyping of the rs2868827 insertion was performed using real-time PCR in 347 patients from Czech Republic (n = 163), Germany (n = 114), Greece (n = 70). The efficacy and safety of POEM were evaluated in 133 patients treated in our center. The post-POEM esophageal motility was assessed using the Chicago Classification in 237 patients in whom HRM was performed prior to and after POEM....
Effect of Barefoot shoes on the footprint loading

Zuzana Vacková Title: Effect of Barefoot shoes on the footprint loading Objectives: The aim of this experiment is to evaluate if barefoot shoes affect the size of vertical component of contact force during walking on a smooth non- abrasive surface during normal conditions. Furthermore to find out which parameters can be used to monitor the degree of convergence foot behaviour during the stance phase of gait cycle with the barefoot and barefoot shoes. Methods: Seven probands (4 women, 3 men) aged 23 to 25 years participated at the study. The dynamic gait parameters (reaction forces from the pad) were measured using Kistler force plates in the extreme load laboratory at FTVS UK. Probands after introduction to the experiment went repeatedly over the power plate subjectively comfortable speed. The first measurement were done while walking in the barefoot type of shoes. The second measurement was barefoot walking. It was a one-time measurement. The obtained data were exported from BioWare® program into MS Excel. The final data processing was carried out in the MatLab program, where the necessary graphs of reaction forces and their evaluation were created. Water quality development in Mladoticky Brook catchment area

Water quality was one of the biggest problems of environment in beginning of 90. of 20th century in the Czech republic. Since 90. the level of water quality was in czech rivers rapidly better. Mainly in big rivers have accomplished great changes. The small rivers haven't notice that rapid changes. The Czech republic, after entrance to the European Union have committed to comply with Directive 91/271/EEC reduce urban waste water and which should have big influence to water quality also in small rivers. During 90. industry technology, agriculture, cleaning technology of waste water etc. reached big development. Therefore the presumption is higher quality of water in the Czech republic since 90. Goal of the thesis is comparison of results from 1999-2000 with results from 2012- 2014 from Mladoticky brook catchment, which is lined up to small river catchments (79,77km2 ). From this comparison there is obvious trend, if conditions of the water quality are really better or not since 90.
