Effect of chromatin on the repair of double-strand DNA breaks after cleavage by CRISPR/Cas and other programmable nucleases in plants

Plants are highly resistant to ionizing radiation, also thanks to a high-quality repair system for repairing double-stranded breaks. Double-strand breaks in plants are repaired by four repair pathways. Most often, double-strand breaks are repaired by non-homologous end joining (NHEJ), which joins the broken ends without further processing. More accurate but slower and more complex is repair through homologous recombination (HR), which repairs the break using a homologous sequence. HR repair takes place preferentially in a region with active transcription and during the S and G2 phases of the cell cycle. Alternatively, repair further proceeds through single-strand annealing (SSA) or Theta mediated end joining (TMEJ). Both pathways are based on short homology between the overlapping ends of the double-strand break. An often neglected part of repairs is the overcoming of repressive chromatin, which protects the genome from DNA damage and prevents access of nucleases but also acts as a barrier for repair proteins. The Life and Work of the Pastors Alfred Kocáb and Jiří Doležal between years 1968 and 1989

This thesis deals with the life and work of two pastors of the Evangelical Church of the Czech Brethren, Alfréd Kocáb and Jiří Doležal, between 1968-1989. For this research the oral history method was used. The thesis contains a selection of interviews, which the author conducted with the pastors in spring 2013, and an interpretation of them according to the rules of the discipline of oral history. Preceding to the interviews available materials related to both pastors were studied, which are mainly documents in church and state archives, but also interviews with contemporaries of both pastors. The aim of this thesis is to create a platform to the pastors to speak, so that they can present their life and work according to their memories and comment various events, which constituted them. The two pastors represent two different wings in the ECCB in that period. In this way, these two concepts and accents of life and theology are entering into a mutual dialog. Through two life testimonies we can pars pro toto observe the life of the church and the country in the difficult years 1968-1989. The second aim of the work was to create new historical sources in the form of recordings of interviews and transcriptions of them.
Myths of the Contemporary Man: An Application of the "Mythic Reading" Method to the Selected Works of the World of the World Literature of the 20th and 21st Centuries.

Present paper tries to offer an approach to the myth which would characterize the currentness of myth for the contemporary man. The paper begins with an attempt to grasp meaning(s) of the Greek term μυθος, its relationship to the term λογος in the course of history and a critical evaluation of definitions and interpretation methods on case studies of selected scientists in the field of myth theory. Subsequently, own definition of myth is proposed, following and elaborating the theories of P. Ricoeur, L. Coupe, D. Cupitt, and others. Myth is then perceived as a dialectics between narrative and mythic motives which are not unalterable but, in the context created by a story, they obtain always a new meaning. In such manner, myths form chains of interconnected versions that are open for future interpretations in the process of "radical typology." Myths are not restricted to the realm of religion, "radical typology" factors profane context into calculation. Individual links of the chains of myths can represent pieces of literature as well. It is the literary works of 20th and 21st century (M. Bulgakov's The Master and Margarita, G. Orwell's 1984, H. Murakami's Kafka on the Seashore, P. The Natural Law In The Work Of Božena Komárková

Božena Komárková put her brain to a natura law/natural justice in her study of human rights. She said that a natural law is unnatural artificial human aftifact. Komárková was influenced by The Bible and a Biblical way of a justice, law and human institutions and orders. They lost their absolute validity/authenticity, they were relative in The Bible. In The Bible is not exist any natural law. There is not only one absolute Decalog - one ordinary code - in the biblical canon There are more of the Decalogs and each of them is contextualized. The Bible helps us to find new dimension (terrestrial-time, corporeality and interpersonal) of ouf being. Komárková find an influence of biblical way of thinking in The Reformation. There is adiscrepancy between a reformation way of law and a natural law. Komárková advised of a fact that Jean Calvin finished the epoch of traditional natural law thinking. Disagreement of Karl Barth with every kind of a natural law in ethical area is just his objective disagreement with a natural theology. The Theism of Charles Hartshorne

The aim of the thesis is to introduce the American process philosopher Charles Hartshorne and his philosophical theism and to relate his approach and his thoughts to the debate about the relationship of theology and philosophy. After introducing the basics of the career and work of Alfred North Whitehead, the better known representative of process thought, and then the career and achievements of Hartshorne himself, I proceed to make a survey of the main aspects of his neoclassical philosophical concept of the God-world relation. Special attention is paid to one of Hartshorne's specifics - his defense and interpretation of the ontological argument for the existence of God. I then deal with Hartshorne's attempts to engage some classical theistic concepts, namely that of Thomas Aquinas, and of the luminaries of Protestant theology, Paul Tillich and Karl Barth, into a kind of dialogue. Then I introduce briefly "process theology" and some significant examples of applying Hartshorne's ideas in theology, Christology, and moral theory. I survey some polemical reactions to process thought and to Hartshorne by German Protestant theologians and I consider the possibility of a certain kinship between Hartshorne's view of the God-world relation and the theology of nonviolence. Guilty conscience as a way to Goodness

The intention of my thesis is to explain the issue of guilt as a natural part of a human life. I try to consider different approaches to quilt as they have passed during the centuries until now. I also briefly consider the psychological insight into dealing with our own quilt. Starting point to my understanding of guilty conscience as a way to goodness is Jasper's conception of guilt. Jasper's division of guilt into four elements is fundamental for my recognition of four levels of guilt in biblical events of the Old and New Testament. However Jasper's recognition of guilt can not be fully compared to biblical events, it can be considered as a pattern that is actual to current times. My intention is to point out guilty conscience that forms and outlines human determination, gives knowledge of faith that is necessary for dealing with quilt. Guilty conscience that opens a horizon of faith is illustrated especially in biblical events that create pattern situations either for a human of the 21st century. Guilty conscience is actually of future characteristics since it opens a level of life that is formed by religious or transcendence faith. Dynamization of spatial data using location-based services: Tourist potential of industrial heritage

The paper deals with industrial heritage and its potential in tourism. Although the post-industrial landscape is an integral part of all the countries that went through a period of de-industrialization and economic transformation, presentation and publicity of valuable industrial monuments that are linked with post-industrial landscape in many cases are not fully utilized. This paper aims to acquaint readers with a possibility of transformation of data on industrial objects into augmented reality, while using location-based services. The final result is the web-based platform allowing easy data transformation. The paper has thus largely methodological-application character. The outcomes of the testing phase indicate that information about the objects do have their limits, but respecting them brings their effective performance in augmented reality.

Churches and Charta 77

The aim of the bachelor thesis "Churches and Charta 77" is to cover the early years of existence of Charter 77 and its reflection in churches. After a short introduction of the historical and social contexts, the main topic of the thesis focuses on the analysis of the press issued during January and February 1997, including both the papers controlled by the Czech government (Rudé právo) and the press issued by churches (Kostnické jiskry, Český zápas, Katolické noviny). The work also focuses on the analysis of the Synod Council records from the archive files of the Evangelical Church of Czech Brethren from the whole year 1977, on published interviews and private dialogues conducted for the purpose of a research related to the Evangelical Church of Czech Brethren, the Czechoslovakian Hussite Church and the Roman Catholic Church. This work should show the historical and social contexts the Charter 77 entered into and the attitudes that the churches had towards Charter 77 and its authors and collaborators. It also aims to find out if there were any differences between the public manifestation of churches - towards the public administration - and the internal manifestation within a church itself.
