Elimination voltammetry with linear scan (EVLS) represents an almost traditional\nmathematical method that helps to obtain additional and detailed information about the recorded\nelectrochemical signals and therefore, to better understand the analyzed systems. Although it\nhas been substantially developed and expanded during its existence of more than a quarter of a\ncentury and has found many areas of application, like any other mathematical and experimental\nmethod, it exhibits several advantages and disadvantages. In this contribution, we summarize\nits strengths, weaknesses, opportunities, as well as threats.\n
Dotační možnosti realizace rozvoje cestovního ruchu v mikroregionu Horňácko

This thesis evaluates the use of grant funds from various grant schemes for transnational, national, regional and local level in relation to support the development of tourism. The chosen problem area for the evaluation is the micro-region Horňácko. Review of the literature explains the concepts related to the evaluation of strategic documents, programs, projects, contemporary approaches and documentation as well as methods for the evaluation. The thesis also analyzes the main strategic documents related to the development of the monitored area through which the importace of development is confirmed. Especially in the area of tourism there is a potential area specific for such development. There are also included characteristics of the area, history and explanation of the specific municipalities. The usage of financial aids for the development of tourism is evaluated in the analysis of realized projects. Various projects in the field of tourism development in Horňácko are specialy evaluated with objectives of strategic documents and the usage of potential in the area. In the evaluation we find essential persistent problems which are confirmed by the content of the new LAG strategy for the next programming period from 2014 to 2020. There are researched further possibilities of financial aid for new programming period from several sources availible on different levels in the framework of ascertined facts.
Study of Genotoxicity of Organic Compounds Using DNA Biosensors

Proposed dissertation thesis presents novel hybrid electrochemical DNA biosensors based on distinct forms of pyrolytic graphite ("edge-plane" and "basal-plane") and their potential in terms of applicability in the field of DNA electrochemistry. Testing of the applicability of the presented bioanalytical tools was preceded by thorough optimization process involving selection of the optimal values for the most crucial operational parameters of the electrochemical DNA biosensors fabricated by means of electrostatic adsorption - deposition potential (Edep), deposition time (tdep), optimal DNA concentration for electrostatic adsorption (cg(DNA)), and possible involvement of convection during the adsorption procedure. Initially, applicability of presented bioanalytical tools was investigated in relation to monitoring DNA damage by utilization of triplet of external DNA damaging sources - UV light irradiation, one- electron oxidants, and hydroxy radicals. Oxidation Processes of N-methylguanines

The oxidation processes of N-methylguanines were investigated in both experimental and\ntheoretical way. Voltammetric oxidation potentials of guanine and its methyl analogues,\nacquired on polymer pencil graphite electrode (pPeGE), were compared with the HOMO\n(SOMO) energies calculated by density functional (DFT) methods. Our study contributes to\nunderstanding not only the oxidation processes of methylated guanines but also their role in\nepigenetics.
Effect of Suspension Kinematics on Vehicle Handling

This diploma thesis deals with effect of suspension kinematics on vehicle handling. In the first part of the thesis, there are the basic parameters of suspension kinematics introduced. In order to perform the simulations, two multi-body vehicle models are created, one of which is derived from an off-road vehicle, for which there has been a significant change in suspension. For each model, there further is a description and comparison of the kinematic characteristics of different settings variants of their suspension introduced. The main part of the thesis deals with the analysis effect of individual suspension changes on vehicle handling during the selected driving maneuvers. The results of the simulations are qualitatively and quantitatively evaluated, and the resulting findings are in the conclusion summarized.
Novel Approaches in Electrochemical Determination of Xenobiotic Compounds and in Study of Their Interaction with DNA

Presented Ph.D. Thesis is focused on the development of analytical methods applicable for determination of selected xenobiotic compounds and for monitoring DNA damage they can induce. The main attention has been paid to the development and testing of non-toxic electrode materials for preparation of miniaturized electrochemical devices and novel electrochemical DNA biosensors. 2-Aminofluoren-9-one (2-AFN) was selected as a model environmental pollutant, which belongs to the group of hazardous genotoxic substances. Its carcinogenic and mutagenic effects may represent a risk to living and working environment. 2-AFN has one oxo group, where the cathodic reduction occurs, and one amino group, where the anodic oxidation occurs. The voltammetric behavior of 2-AFN in the negative potential region was investigated at a mercury meniscus modified silver solid amalgam electrode (m-AgSAE) representing a non-toxic and more mechanically robust alternative to mercury electrodes. Title of diploma thesis: Modification of Glutathione S-transferase by selected glycating agents Glycation is a non-enzymatic reaction, which leads to the binding of carbonyl compounds to free amino groups of amino acids, peptides, proteins or other biomolecules. This process is closely related to impaired protein function in diabetes mellitus, during aging, and in neurodegenerative diseases. The main purpose of this thesis was to monitor the impact of several glycating agents on the properties of the enzyme glutathione S-transeferase (GST, EC 2.5.1.18) which plays a key role in the detoxification of many exogenous as well as endogenous compounds. Methylglyoxal (0.5-2 mM), glyceraldehyde (0.5-10 mM), glucose (50 mM), and fructose (50 mM) were selected as glycating agents. Development of Novel Electrochemical Methods Using Various Membrane Materials for Monitoring of Selected Anticancer Drugs and Phytochelatins

Present Ph.D. Thesis is focused on the development of electrochemical methods for determination of anticancer drugs using various types of membranes for their preliminary separation. Furthermore, this Thesis reports the study of transport mechanisms of heavy metals in the presence of phytochelatins across biological membranes. Sodium anthraquinone-2-sulphonate (AQS) was used as a model compound for its similar structure with anthraquinone-based (AQ-based) anticancer drugs (doxo/daunorubicin) and also due to its better availability. All these compounds can be easily electrochemically oxidized and/or reduced. Redox behaviour of AQS was investigated by cyclic voltammetry (CV) and differential pulse voltammetry (DPV) in a cathodic region on mercury meniscus modified (m-AgSAE) and polished silver solid amalgam (p-AgSAE) electrodes, Obtained results were used for the development of a micro-volume voltammetric cell (MVVC). Influence of hydroxycinnamic acids on methylglyoxal-induced protein glycation

Author: Hana Vejvalková Publishing place: Charles University in Prague, Faculty of Pharmacy in Hradec Králové Title: Influence of hydroxycinnamic acids on methylglyoxal-induced protein glycation Supervisor: PharmDr. Iva Boušová, Ph.D. The diploma thesis was aimed at potencial antiglycation effect of antioxidants of natural origin (hydroxycinnamic acids) on the modification of bovine serum albumine and asparate aminotransferase by methylglyoxal as a glycating agent. I monitored the effect of hydroxycinnamic acids on generation of glycation products using fluorescent, absorption, and electrophoretic methods. The influence of hydroxycinnamic acids on the glycation of model proteins was ambiguous. The only derivative with proven antiglycation effect on production of non-specific glycation products and pentosidin measured by fluorescence and spectrophotometry was p-coumaric acid in 100 μM concentration. On the other hand, chlorogenic acid 100 μM displayed strong proglycation effects in all methods used for detection of glycation products. The effect of all tested substances was concentration dependent. Results show that the more substituents given acid contained in its molecule, the stronger proglycation effect this compound exerted.
