The diploma thesis deals with the marketing strategy of an international company. The theoretical part presents the characteristics of marketing and selected analyzes. The practical part analyzes the international company WABCO Holding Inc. and includes macro-environment and micro-environment analyzes and research. Finally, proposals compiled using the data obtained from analyzes and research are presented, representing measures to improve the company's marketing strategy.
Taxation of Non-profit Entity - a Case of Selected Accounting Entity
Sklenářová, Petra ; Pěta, Jan (referee) ; Svirák, Pavel (advisor)
Bachelor thesis deals with non-profit organization and subsequently its taxation from the perspective of corporate tax and road taxes. The theoretical part shows the characteristics of individual non-profit organizations, their financing and the conclusion of the theory detailed described corporate tax and road tax. The practical part analyses the Department of Wheelchair League. This chapter specifies the activities of the Organization, the determination of the subject of tax, the tax base and tax calculations themselves. Finally, it includes a tax return.
Case report of physiotherapy care for a patient with a fractured ankle joint
Sklenářová, Petra ; Kučerová, Ilona (advisor) ; Hamouzová, Dita (referee)
Title: Case report of physiotherapy care for a patient with a fractured ankle joint Objectives: The objective of the theoretical part is a detailed introduction to theoretical knowledge and treatment options for ankle joint fractures. The aim of the special part is to become familiar with the course of treatment of a patient diagnosed with a fracture of the ankle joint and with the therapeutic and investigative procedures that were used for the treatment. Methods: The theoretical part was developed thanks to findings from professional literature, professional articles and information gathered during the bachelor's study of physiotherapy at the UK FTVS. The special part was developed on the basis of data obtained during bachelors practice. Key words: ankle joint fracture, ankle joint, physiotherapy
Effect of surfactin on the lipid moiety of Bacillus subtilis cytoplasmic membrane
Sklenářová, Petra ; Seydlová, Gabriela (advisor) ; Lichá, Irena (referee)
Surfactin, a secondary metabolite produced by Bacillus subtilis, is a surface active compound and antibiotic permeabilizing membrane bilayer. The aim of this study was to reveal the self-resistance strategy at the level of the lipid moiety of cytoplasmic membrane, which B. subtilis employs to combat surfactin in concentrations that are lethal for other bacterial species. Non-producing strain B. subtilis 168 was cultivated in the presence of two different sublethal concentrations of surfactin (350 a 650 µg/ml), which was isolated from the culture broth of B. subtilis ATCC 21332. Presence of surfactin in the medium resulted in a concentration dependent lag phase, which took 40 min (350 µg/ml) and 3 h (650 µg/ml), respectively. Afterwards, the culture grew with the altered doubling time of 44 min (350 µg/ml) and 126 min (650 µg/ml), respectively. Surfactin induced substantial changes in the phospholipid composition of the cytoplasmic membrane. The proportion of the major phospholipid component phosphatidylglycerol decreased and inversely, the level of phosphatidylethanolamine increased. Interestingly, the content of phosphatidic acid rose considerably in the presence of surfactin concentration causing stimulation of B. subtilis growth (350 µg/ml). Liposome leakage assay using phospholipids mimicking...

Conformational changes of the membrane protein sensors
Sklenářová, Petra ; Konopásek, Ivo (advisor) ; Fraiberk, Martin (referee)
This work focuses on a structure and a function of prokaryotic signal systems. There are three types of signal systems in bacteria: one-component, two-component and three-component systems. These three systems differ with respect to their structure and number of protein components and domain. I have focused mainly on two-component systems in bacteria despite their occurrence has been confirmed also in lower eukaryotes. Prokaryotic two-component system are the main instruments which help bacteria to differentiate between different stimuli from the environment and to react to them. So far, there were described two types of two-component systems. The first one is the basic system and the second one is the phosphorelay system. Both types of two-component systems contain two indispensable proteins: histidine kinase and response regulator. They are different with respect to their domain structure of proteins and in a mechanism by which they transfer the signal. The signal transmission system consists of two or four steps in case of the basis or phosphorelay systems, respectively. The principle of the signal transmission is the same for both of them, a phosphate group from the histidine kinase is transferred to response regulator. In such manner, response regulator is activated by and is able to regulate the...
