Subretinal implantation of a carrier for retinal pigment epithelium in minipigs
Straňák, Zbyněk ; Kuchynka, Pavel (advisor) ; Mahelková, Gabriela (referee) ; Rusňák, Štěpán (referee)
Age-related macular degeneration (AMD) is the leading cause of practical blindness in developed countries. AMD is one of many retinal diseases that arise from the absence and/or dysfunction of the retinal pigment epithelium (RPE). There are two main types of VPMD (wet and dry), which differ in etiopathogenesis and ophthalmological findings. To date, therapy is only possible for the wet form of AMD, where neovascularization permeating from the choroid and subsequent oozing or bleeding in the subretinal space with deterioration of rod and cone cells and other neuronal cells occurs. Neovascularization can be partially prevented by repeated intra-vitreal applications of anti-VEGF agents. Without treatment, the result is a disciform retinal scar and irreversible visual impairment. A current trend in the treatment of AMD is the attempt to introduce innovative surgical procedures that have the potential to improve severe ophthalmic morbidity, particularly in non-responders to pharmacological therapy. The aim of this study was to evaluate the success rate and optimize a surgical procedure that allows for controlled and safe transplantation of RPE cells. The success of transplantation of RPE cells in the form of an epithelium that acts as a selective barrier immediately after transplantation is dependent on the...
Angiogenesis Promoters Determination in the Intraocular Fluid by Means of Multiplex Immunoanalysis
Rusňák, Štěpán ; Raiskup, Frederik (advisor) ; Řehák, Jiří (referee) ; Kolář, Petr (referee)
The objective of the project was to find out whether patients with proliferative diabetic retinopathy (PDR) can be stratified by means of multiplex immunoanalysis - i.e. whether in patients with complicated clinical finding a difference in concentration of selected biomarkers can be found in comparison with patients without complications. Multiplex analysis was used to determine angiogenic, anti-angiogenic and inflammatory cytokines in 59 samples of intraocular fluid aspirated from anterior chamber of PDR patients and 51 cataract patients control samples. The results for patients without complications were compared with patients in the terminal stage of therapeutically unmanageable neovascular glaucoma due to proliferative diabetic changes. It was found that patient stratification according to clinical severity of PDR correlates with pro- angiogenic cytokine concentration in the intraocular fluid. Furthermore, several additional cytokines were found the levels of which were elevated in the intraocular fluid of PDR patients compared to the control group. Patients with clinically more severe finding had higher levels of pro-angiogenic cytokines in the intraocular fluid. Determining pro-angiogenic cytokine concentrations by the means of multiplex analysis is of importance when clinical findings show...

Aspects of Conflict in the Behaviour of Fish and the Protection of a Specially-Protected Species (Phalacrocorax carbo) after 25 years of Breeding the Species in the Czech Republic
Rusňák, Štěpán ; Řezníček, Jan (advisor) ; Andreska, Jan (referee)
The thesis objective was to collect all modern biological knowledge about the presence, migratory and feeding habits of a special grade protected bird species of The Great Cormorant and to collect statistical data on its predatory pressure on fish communities which represents the core of the conflict between nature conservation activists and fish farmers. The findings of ornithological monitoring have been used as input data - winter habitat frequency of the bird species, data on nest distribution of the birds and feedback reports on bird-ringing in the last three decades. Data on selective shooting requests and estimated damage compensation request data relating to the species has been provided, too. An extensive survey in the ornithological community's opinions has been carried out. Using the results of literature research, direct observations and a statistics evaluation, I tried to find an answer to the following questions: - What is the cause of the pan-European species expansion? - What damage does the species cause with its feed behaviour to the fish farmers? - How can the damage claim be specified and provable? - What are the potential methods of protecting the fish communities and the species regulation within the legislation limits? - To what degree is it feasible to protect cormorants in the Czech...
