Pospíšil, Petr ; Beneš, Jan (referee) ; Topolánek, David (advisor)
The theme of this master‘s thesis is distribution network development and configuration in the part of Brno – Trnitá. There is expected to be investments there in the next few years. The thesis consists of two parts. For better understanding of this problem, the first theoretical part concerns with the description of electrical network (subdivision in accordance with voltage levels, conductor types, network types and other electrical network parts), solution of grid’s steady state (as a linear task and also non-linear task, elimination of the balancing node, the iterative method) and Brno distribution network characteristics. The second part concerns with distribution network development and configuration depending on anticipated load development in Trnitá. The result is proposal for transformer station construction Opuštěná fitted with three transformers of nominal output SN = 40 MVA and network configuration change required both for providing new consumption supply as well as sustaining current one.
Price analysis of building with changes of construction materials
Pospíšil, Petr ; Vovesný, Jan (referee) ; Aigel, Petr (advisor)
The following bachelor thesis concentrates on the analysis of construction expenditures dutiny the process of introducing material and construction changes. We have selected four variant of the given construction systems supplied by four different providers. This has been conducted under the following condition: all the four variants has a common heat transfer coefficient. U= 0,14 [W/m2K]. Among the selected systems we have included Heluz, Velox, Porotherm and Ytong. Under Heluz system we have chosen Heluz Family, ground surface, thickness 500 mm. This system does not involved contact insulation. As for Porotherm, we have decided for ceramic breeze blocks Porotherm 30, thickness 300 mm, to follow the obligatory common heat transfer coefficient, therefore we have also suggested contact insulation as of 220 mm. The third system, Velox permanent boarding, VELOX WS EPS PLUS thickness 420 [mm]. This system fully complies with the heat transfer coefficient requirements. Finally, we have used Ytong breeze blocks 300 mm in thickness. For this solution there also arose a need to design the contact insulation to follow the heat transfer coefficient requirements. For ech project variant we have designed project documuntation as a the basis for further budget item calculations pertinent to each single project variant. The item budget calculation was made using KROS PLUS programme. The overall budget prices and variables in each project variant have been comapred with each other, the data processed into several graphs accordingly. Among the variables we have included ground works, founding, vertical and complete constructions, horizontal constructions, surfaře finishing works, flooring and filling works, thermal insulations. Furthermore, purchasing prices for each invividual type of material, based on provider prices list, have been compared with prices as given by KROS PLUS programme.
Endless Cave
Pospíšil, Petr ; Pečiva, Jan (referee) ; Milet, Tomáš (advisor)
The goal of this thesis is to implement an application showing an endless cave. The basis of this cave is simplex noise method. On the noise produced by this function is afterwards applied thresholding. Produced grid of points is used like input for marching tetrahedrons algorithm. This algorithm converts volumetric data to boundary representation. Phong reflection model and Bump mapping were used in the application, too, in order to improve the visual quality. The application is based on OpenGL library. The first part of the technical report contains theoretical description of mentioned methods, the second part contains description of implementation.
Influence of Wind Farm Integration to Power System
Pospíšil, Petr ; Belatka, Martin (referee) ; Malý, Jiří (advisor)
This bachelor’s thesis is divided into four main parts. The first point includes a characterization of the origin of wind, a principle and a division of the wind – mill motors and a characterization of a wind power plant. The second part analyses effects which the wind power plans have on the power system and also analyses grid problems with large amount of installed wind – mill motors. In the other point national grid codes are analysed (such as Germany, Denmark, Ireland). On the bases of these analyses the suggestions of the modifications of the Czech distribution grid code are summarized.
Fluorescence Spectroscopy: Advanced methods and their defined applications in protein science
Pospíšil, Petr ; Hof, Martin (advisor) ; Heřman, Petr (referee) ; Polívka, Tomáš (referee)
The hydration and dynamics of the biomolecules appear to be vital for their proper biological functioning. In the presented thesis, various fluorescence techniques were developed and applied to access these properties and their changes upon the mutual interactions of the biomolecules. Initially, the solvent relaxation method based on recording time-dependent fluorescence shift (TDFS) was used to map DNA interactions with proteins and lipids by the newly synthesised fluorene dye covalently bound to the DNA. Secondly, copper-transporting ATPase was probed by Badan attached to the copper-binding cysteine-proline-cysteine motif. The variations in hydration were found to be crucial for the proper ATPase function. Third, a detailed study on quenching of Badan/Prodan fluorescence by tryptophan revealed the limitations of the TDFS method for protein studies, which is essential finding for further applications of TDFS. Fourth application involves investigations of heavy atom effects on the excited state relaxation processes by up-conversion approach in iodinated metallocorroles, which are promising dyes for biological imaging. The obtained findings shall help in further tuning of the optical properties of the corroles desired for the variety of applications. Finally, fluorescence correlation spectroscopy...
Self-assembly and excited energy transfer in bacteriochlorophyll aggregates
Pospíšil, Petr ; Pšenčík, Jakub (advisor) ; Vácha, František (referee)
Bacteriochlorophyll c (BChl c) belongs among photosynthetic pigments. It is found in lightharvesting complexes of green photosynthetic bacteria in the form of self-assembled aggregates. It is possible to prepare BChl c aggregates also in vitro in aqueous solutions, in the presence of certain non-polar molecules (e.g. lipids or carotenoids). In this work, artificial light-harvesting antenna was prepared composed of BChl c and selected azulene derivative. The aggregation of BChl c with azulene enhances the absorption in the near UV region compared to BChl c. Fluorescence spectroscopy was used to study an efficiency of energy transfer from azulene to BChl c. The procedure used was based on fluorencence excitation spectra measurement, and a comparison with absorption 1-T spectra. The measurements were complicated by anomalous fluorencence of azulene, which occurs from the S2 state. Furthermore, the reference diode in the fluorescence spectrometer did not correct properly the signal in the near UV region, where azulene absorbs. The problem was partially solved by preparing the new correction files using selected laser dyes. Excitation spectra demonstrate the presence of the energy transfer from azulene to BChl c, however, it was not yet possible to satisfactorily quantify its efficiency.
Variances in the Blue Card Directive Transposition across the European Union
Pospíšil, Petr ; Young, Mitchell (advisor) ; Šlosarčík, Ivo (referee)
This work attempts to analyze causes of divergent transposition of the Blue Card directive, regulating the conditions of immigration of highly-skilled workers from third countries, between individual EU Member States. The differences are analyzed on two levels. Firstly, in terms of timeliness and correctness of transposition, secondly, from a perspective of a different manner of transposition of the discretionary clauses entailed in the Blue Card directive (whether the Member States opted for an open or for a restrictive way of transposition of these provisions). Scholarly literature about transposition compliance and highly-silled immigration policies creates the foundation, by means of which the author identifies several variables and articulates hypotheses aiming at providing explanation of the divergent transposition of the directive in the respective Member States. The author classifies the Member States in groups and clusters according to their transposition performance and its results embody a point of departure for a subsequent analysis. From each group, certain number of countries is selected for a detailed country-level analysis in order to ensure universality and geographical representativeness of the outcomes. To review how the individual hypotheses confirmed in the Member States, the...
