Unravelling molecular mechanisms of the abasic crosslink DNA repair

4 Abstract DNA as the primary carrier of genetic information guarantees organisms to live, grow, develop and reproduce. However, this most vital molecule in the cell is subject to various damages every moment. If it is not repaired, the cell and the organism will eventually succumb to inevitable destruction. One of the most serious damages is abasic site interstrand crosslink (Ap-ICL). Ap-ICL is formed spontaneously when an abasic site covalently pairs with an adenine on the opposite strand. The lack of information on repair mechanisms, the influence of the local sequence and its stability leads to questions about the fate, toxicity and occurrence of these lesions in cells. During evolution, several mechanisms have evolved to repair these and other damages to ensure the organism's survival. A recently discovered pathway known to repair Ap-ICL is named after the DNA glycosylase responsible for removing Ap-ICL. NEIL3 DNA glycosylase is recruited to Ap-ICL by ubiquitylation of DNA helicase, which is part of the DNA replication complex. NEIL3 glycosylase contains several zinc-finger domains, that bind to the damaged DNA and ensure its catalytic function. Apartment building

The subject of this bachelor thesis is the elaboration of project documentation for the construction of an apartment building. It is a detached basement building, which is located on a rectangular plot with flat terrain. The building is designed with four floors and one underground floor. This new building is equipped with a lift for 8 people. In the underground floor there is a technical background of the building and a storage cellars for individual apartments. There are 12 flats with different disposition and size of floor areas. The load-bearing structural system is diagonal. The lower structure is made of a waterproof concrete, the so-called "white tub" which consists of a reinforced concrete foundation slab and reinforced concrete basement walls. The upper structure is built of Porotherm system, which is insulated with ETICS facade contact system with basalt wool insulation boards. Ceiling structures are designed as monolithic reinforced concrete slabs and the staircase is built as a reinforced concrete monolithic three-armed. The façade is shaped according to the partially opened loggias. The last floor recedes from the face of the building and creates spacious terraces. Holes are filled with wooden windows. The roofing of the apartment building is solved by a single-layer flat roof with a classical layer arrangement and a hydro isolation layer made of asphalt strips.
Car showroom with service

The subject of this master thesis is the elaboration of project documentation for the construction of a car showroom & service. This building is located in Brno, cadastral area of city Brno: Královo Pole. It is a detached building without a basement built on a trapezoid shaped area with a flat terrain. Object is divided into two functional parts: the car showroom and the car service. These two parts are constructionally and operationally connected. The car showroom is designed as a two-storey object, which serves as car exhibition, technical and hygienic facilities and mainly the administrative parts of both objects. The car service is designed as a single-storey object without a basement, which serves as a workspace of the car service and a warehouse. The load-bearing structural system of the car showroom is made of reinforced concrete pillars with ceramic masonry. The load-bearing structural system of the car service is made of reinforced concrete pillars with light sandwich panels. The foundation slab consists of prefabricated stepped foundation pad, prefabricated reinforced concrete grade beam and concrete strip footing. Insulation system of the building is made of insulation boards from basalt wool cladding by ventilated facades with perforated steel panels. Insolation in the car showroom is secured by glass curtain wall in combination with plastic windows and doors. Ceiling structures are designed as prestressed ceiling panels Spiroll. Roofing structure of both objects is supported by prestressed reinforced concrete roof girder in combination with trapezoidal sheet and insulation boards from basalt wool and PIR boards. The roof of the objects is flat with ordinary layer order with waterproof layer from softened PVC.

This master's thesis deals with intelligent wiring Foxtrot and use in the field of lighting. The theoretical part is comparsion of classical and intelligent wiring. This section provides an overview of the most widely used bus system for building control in our market. Thesis is devoted to the description of the Foxtrot system and development environment mosaic. In the development environment Mosaic is designed program for lighting control DALI interface. WebMaker enabled program visualization and his remote contorol. The result of practical part is a lab manual.
Implementation of the Roofing of the Stable for Cattle in Studenec

The topic of the bachelor’s thesis is the elaboration of a construction technological project for the roofing of a cattle stable in Studenec. The work contains a technical report focusing on the installation of the roof structure, the construction situation with broader relations of transport routes, technological regulation for the roofing stage, item budget with area statement, site equipment design with technical report site equipment, time schedule, machine design. Furthermore, the work addresses the control and test plan, occupational safety. The last point of the thesis deals with the implementation of roofing of buildings with a larger span.

18 metres long drawing on canvas, describing my own six years long live on high school and in my home town or my cottage. Trophies, the most important theme for my art, is growing in this drawing. The canvas is reeled in wooden machine and it is scrolling with the handle. In this installation there are some of my trophies and founded fragments of nature.

Indicates the proportion of various types of renewable energy sources in electricity production and their representation in the electrical load profile. Thesis describes the various storage systems, which can be used to stabilize the renewable energy sources. Evaluate the applicability of these storage systems in the energy sector.
Structural studies of selected protein complexes involved in signal transduction

Protein-protein interactions are critical for most physiological and pathophysiological processes. Detailed characterization of these interactions is therefore essential not only to understand the nature of these events, but also to design strategies to target these interactions. This work focuses on the study of the structure and interactions of several proteins and their complexes. Apoptosis signal-regulating kinase 1 (ASK1) is a mitogen-activated protein kinase kinase kinase (MAP3K) that activates the p38/JNK protein kinase pathways, thereby directing cells toward an inflammatory response or apoptosis. ASK1 interacts with thioredoxin (TRX), a small dithiol oxidoreductase, which inhibits ASK1, but the mechanism of this inhibition has not been clarified. CaMKK1 and CaMKK2 are Ca2+ /calmodulin (CaM)-dependent protein kinases that regulate cellular energy balance, memory, and inflammation, among others. Both are inhibited by 14-3-3 proteins, but despite their domain and sequence similarities, the extent of 14-3-3 protein- mediated inhibition is different. Estrogen receptor alpha (ERα) is a nuclear receptor involved in breast cancer. Neuronal cell culture in vitro

Neuronal cell cultures are in vitro cultures of dissociated neurons that have become an essential part of many neurobiological experiments in the last century. Cultured neurons not only allow to answer questions about their physiology under complex in vivo conditions, but also can serve as a model of neurodegenerative diseases. Neuronal cells can either be isolated directly from the nervous tissue of animals at the prenatal or adult stage of development, or they can be obtained through targeted manipulations of stem cells and secondary cell lines that lead to their neuronal differentiation. Primary neurons are considered the gold standard of neurobiological research not only because of their long tradition of cultivation, but also because primary neurons retain typical neuronal properties under in vivo conditions. There are several disadvantages associated with primary neurons, including the fact that fully differentiated neurons do not proliferate and are relatively demanding in terms of culture conditions For this reason, their role is often replaced by mitotically active secondary cell lines or stem cells.
