Inovace PPK GNSS senzoru Titan v posuzování zemětřesné kinematiky

Within the sotware package Trimble Business Center ver. 5.90 (May 2023) was released a novel PPK GNSS processor named Titan. It utilizes all available GNSS constellations bringing improvement over the previous processor. CROPOS as the national permanent GNSS network of Croatia was modernized in summer of 2019 and since than it provides observations of all available GNSS constellations, namely, the GPS, GLONASS, Galielo and BeiDou. On March 22nd 2020, the wider area of Zagreb was hit by the M5.4 earthquake. The closest GNSS station to the epicenter (approx. 9 km) was the station ZAGR in Zagreb downtown and as such it was subject to earthquake shaking. Although the kinematic effects of that earthquake have already been assessed and examined, with this approach and novel PPK GNSS processor we are attempting to provide new insights into the earthquake kinematics involving observations from several CROPOS's GNSS stations in wider area around Zagreb.
Use of Equine Assisted Therapy with children suffering from behavioral disorders and ADHD

Therapeutic horseback riding is one of the most popular and the most used form of animal based therapy. The term "therapeutic horseback riding" itself is very broad and it consists of four basic branches. The most used, yet least described, is the branch that has been called "pedagogical-psychological horseback riding therapy" since 2011. This branch is further divided into two sub-branches, Equine assisted psychotherapy and Horse-based activities. These two sub-branches overlap each other and are usually conducted together. With school-age children they are mostly used in cases of behavioral disorders and ADHD. The goal of the thesis is to evaluate the effectiveness of the methods with children diagnosed as such. The research was realized in "Stredisko hyporehabilitace Pirueta" which operates within the Mental Hospital in Havlickuv Brod. The method of the research was observation of five subjects within the one complete therapy unit. At the beginning and at the end of the unit the tutors of the subjects were given observational scale, which served as supplementary technique for diagnostics. Data obtained from own observation and the data gained from observational scaled matched each other approximately in 90%. The role of phosphatidylinositol-4-phosphate 5-kinase PPK-1 in Wnt signalling regulation in Caenorhabditis elegans

The Wnt signalling pathway is highly conserved signalling among Eukaryotes that regulates many cellular processes. In particular, it plays a role during invidual's development, but it is also important at later stage of life, when it is involved in maintaining homeostasis in the body. Recent studies have shown that phosphatidylinositol-4-phospahte 5-kinase (PIP5K), which is key to the production of phosphatidylinositol (4,5)-bisphosphate in the cell, is also involved in regulation of the canonical Wnt signalling cascade. In mammalian cells, a model has been described in which PIP5K is involved in signal transduction in a Wnt receiving cell. We decided to verify the involvement of PIP5K in the regulation of Wnt signalling also in Caenorhabditis elegans, an excellent model organism for study of signalling pathways, and thus contribute to a better understanding of this evolutionarily conserved pathway. In this work, we found that decreased expression of PPK-1/PIP5K in wild type animals does not result in Wnt signalling disruption. Nevertheless, in conditions, where the activity of Wnt signalling is already reduced, decrease in PPK-1 levels leads to defective migration of the QL neuroblast daughter cell. Zhodnotenie špecializačných skúšok pre kone zaradené do hiporehabilitácie

The diploma thesis deals with the evaluation of the specialization examinations. Each horse for hiporehabilitation purposes is specially selected and trained in a composed Specialized Exam. I'm watching the so-called a selection of effects that affects the results of horses included in the trials and their subsequent use in hiporehabilitation. A database of 195 horses of different breeds was compiled, where different parameters of their body were evaluated on the basis of a linear description, and therefore specialized examinations for horses included in hiporehabilitation were performed. Based on the results of the specialization tests, the collected data were followed by statistical processing and evaluation of the effects of specialization tests for horses in hiporehabilitation. I divided the horses into 6 groups by type to better compare the necessary effects. Then, in the Gretl statistical program, I evaluated the individual parameters of horses that correlate together. I have given both positive and negative values, from which I conclude that one variable is smaller, the other is bigger. Various groups correlate different parameters, depending on the type of the horse. Then, I evaluated the step and trot in the individual groups. In the statistical program I created a so-called thermal maps, where the color correlated degrees of color were, the more red the color, the more the parameters correlate together. Only mares and geldings participate in the specialization tests. The most suitable age for inclusion is 5 years.

Secondary metabolism and its regulation in Streptomyces ambofaciens: the study of cryptic secondary metabolite biosynthetic gene clusters

I. ABSTRACT in English The presented work is focused on secondary metabolism and its regulation in Streptomyces ambofaciens and Streptomyces lividans with special interest in new biosynthetic pathways. The sequencing of bacterial and fungal genomes revealed that the number of their secondary metabolite biosynthetic gene clusters greatly exceeds the number of produced secondary metabolites. Further studies showed that at least some of the newly discovered clusters, called cryptic since no product had been associated to them, were expressed in certain conditions and that they directed the biosynthesis of exploitable secondary metabolites (Gottelt et al., 2010; Gross et al., 2007; Pang et al., 2004). Therefore, these cryptic clusters have been considered as one of the promising reservoirs of new bioactive molecules. Using different approaches I studied the cryptic secondary metabolite biosynthetic gene clusters of Streptomyces ambofaciens, a strain exploited industrially for the production of the antibiotic spiramycin. Evaluation of growth and fertility in chosen columnar apple-tree varieties

This paper deals with the evaluation of growth and fertility in selected apple varieties. I will discuss the range of columnar apple trees, which represent a further step in the development of growing shapes. Eleven different varieties grafted onto two different rootstocks was assessed. Planting was establish in the spring of 2013, the clamp 3 x 0,8 m. The trees were planted as a two-year vaccinees. The aim of the study was to evaluate and compare the increase in trunk diameter, the intensity of growth and yield of selected columnar apple varieties, before and after the end of their vegetation. It was necessary to evaluate the cross-sectional area strain in (cm2), increase in cross-sectional area strain in (cm2), average fruit weight (g), crown height (m), absolute yield (kg) per tree, specific yield on (cm2) cross-sectional area, yield (kg) on increase in area strain, area yield (t / ha) and specific yield on the crown height (kg / m). In the theoretical part I will deal with the history of apple growing and growing development of various shapes, characteristics of apple trees, their significance and composition. Further I will discuss demands on the growing the apple tree and its treating. At the same time I will mention the major diseases and pests. At the end of this part I will describe harvesting methods and proper storage. The practical part is focused on the experiment itself and its subsequent evaluation. I will mention the location and characteristics of the habitat where the apple tree grew. I will describe specific columnar varieties and rootstocks used. I will characterize the specific location of the varieties grown and the activities that were implemented during the evaluation period of the experiment. In conclusion, the results have been evaluated, to which I arrived during the entire measurements. This part contains the observed values displayed in two tables. For a better overview of the data obtained and the results they are displayed graphically. At the end there is a complete summary of the whole work. For the most of the results it has been proved that a base affects the intensity of growth and yield. The biggest growth area strain (cm2) had a variety 'Cumulus' on rootstock M 26, but on the rootstock M 9 the increase was significantly lower. When assessing absolute yield the highest values were observed in a variety 'Red Spring' on rootstock M 26, followed by the yield of variety 'Slendera' on rootstock M 26. The lowest yield was observed in cultivar 'Cumulus' on M 9. In these cases, we can confirm the influence of rootstock of variety. When evaluating the specific yield on increase strain the highest yield was at varieties 'Red Spring' on M 26, followed by a variety 'Slendera', where the yield was greater on the rootstock M 9. Here it is impossible to unequivocally confirm that the rootstock M 26 has a higher yield than the rootstock M 9.
Use of stable hydrogen and oxygen isotopes in hydrology of small catchments in the Czech and Slovak Republic

The contribution summarizes the development of isotope hydrology and hydrogeology in the Czech and Slovak Republic over the past 40 years. Records of 18O and 2H in nine experimental catchments and their interpretation provide a description of runoff component mixing from various soil and rock environments, as well as a calculation of mean water residence time of 7 to 25 months. The 18O and 2H content in two profiles of the Jizera River and two boreholes in the lower reaches shows little communication of river water with the groundwater supply resources in the Lower Jizera area; the communicaton increases with pumping intensity. The 18O and 2H content in wetlands and groundwater in two catchments of the Jizera Mts. shows the differences in recharge to the wetlands via groundwater or precipitation. The isotope hydrology and hydrogeology perspectives in the ČR and SR include capacity building, cooperation of laboratories and conjunctive use of izotopic and hydrogeochemical approaches.
