Advanced Separation Techniques and Data Processing in Capillary Electrophoresis

Techniques of capillary electrophoresis are essential for both the quantitative and qualitative analysis of a large variety of compounds. They find application in many areas of chemistry, from pharmacological industry and food processing, up to highly specialized biotechnological laboratories. Over the last hundred years, their mathematical model has been described with precision. Thus, current research mainly aims for the development of more advanced and more specialized applications. During the greatest boom of affinity capillary electrophoresis within the nineties, many authors would describe similar phenomena under different names. The first part of this work focuses on the consolidation and unification of the problematics and terminology of this method. It also discusses the phenomena which can affect electromigration and separation during electrophoretic processes. We will focus on a specific subset of affinity capillary electrophoresis, partial filling capillary electrophoresis, which, to our knowledge, has not been fully theoretically explored yet, and present a mathematical model allowing the determination of complexation constants and state some limitations of this approach. Processing of selected species of edible insects for food purposes

Táto diplomová práca je zameraná na nutričné zhodnotenie vybraných práškov hmyzu zo svrčkov domových, z lariev múčiara obyčajného a potemníka stajňového a jedného potravinového produktu na báze hmyzu, ktorý bol tiež vyvinutý ako jeden z cieľov tejto práce. Okrem základných nutričných analýz boli použité aj ICP-OES pre zistenie obsahu minerálov a GC-FID pre zistenie zloženia mastných kyselín. Tiež bol stanovený obsah hrubej bielkoviny pomocou CHNSO analyzátora a stanovený obsah EAA. Pomocou PCA analýzy sa úspešne podarilo rozlíšiť vzorky jednotlivých hmyzích práškov, ktoré potvrdili rozmanitosť jednotlivých druhov hmyzu a vďaka projekcii do roviny hlavných komponent je možné vidieť, v ktorých nutričných parametroch sa najviac líšia. Bola tiež vykonaná senzorická analýza novo vyvinutého produktu na báze hmyzu s prídavkom prášku z potemníka stajňového v porovnaní s konkurenčnými produktmu. Súčasťou práce je aj rozsiahly spotrebiteľský prieskum na vzorke 2 019 účastníkov, týkajúci sa vnímania entomofágie v Českej republike a na Slovensku, ktorý sa uskutočnil online.
Preparation and characterization of fibrous non-woven textile decorated by silver nanoparticles for water filtration

The environmentally friendly preparation of silver nanoparticles was proposed for the production of antibacterial non-woven textile used in water filtration. The silver nanoparticles were prepared by chemical reduction of silver nitrate using two environmentally friendly reducing agents, such as fructose or ascorbic acid. For comparison also commonly used reducing agent-sodium borohydride was used. The silver nanoparticles in various size and yield were produced by immersion of the plasma pre-treated polypropylene (PP) nonwoven textile in the colloidal solutions for different periods (35, 45 and 135 min). The morphology of the silver nanoparticles was characterized by SEM and EDX analysis. Additionally, the antibacterial activity of the silver decorated PP non-woven textile was evaluated by an agar diffusion test using both Gram-positive Staphylococcus Aureus and Gram-negative Escherichia coli. The results suggest that the type of the reducing agent has major effect both on the morphology of silver nanoparticles and the antibacterial activity.
Study of macerates of selected plant materials in oily bases

The aim of this thesis was in terms of literary research describe the composition, production, impact on human health and diverse uses of vegetable oils and their herbal macerates in human life. Herbal oil macerates are actually a blend of oils and herbs, created to produce a product that combines the beneficial effects of oils with therapeutic effects of herbs. Avocado and almond oil were used in combination with herbs such as black elder, chinese tea tree and ginger. Each herb also affects to some extent the quality of oil, which was observed in the time dependence of acid number in herbal macerates compared to pure oils. The acidity of the oil is also significantly influenced by temperature and therefore one set of samples was macerated at room temperature and the other in a thermostate at 35°C. The time dependencies of the acidity number of the individual oils and macerates were plotted into graphs and compared. In most cases, the thermostate-stored oils and macerates obtained higher acidity than those stored at room temperature. The effect of the herbs on the acidity of the oil varied in the different types of macerates, with the highest acidity values being taken by macerates with black elder flowers and the lowest, most similar to pure oils, by macerates with black elder fruits. At the end of the experiment, total and free fatty acids from the last sampling (after 77 days of extraction) of macerates and oils at room temperature were analysed by gas chromatography. Again, there were some differences in the composition of total and free fatty acids in herbal macerates compared to pure oils. Some herbs affect the composition of individual fatty acids more, others do not affect it almost at all.

Thesis is focused on implementation of project management in the company and on improvement of project realization processes. Improvement ideas are tested on pilot project and compared with another projects (with no project management applied).
Simulation of routing protocols in ns3 simulation environment

The first part of thesis is about networks that are divided based on area. It includes also basic characteristic of Manet networks. The next chapter is about distribution of routing protocols into preactive, reactive, hybrid. In the simulation, there is comparison of 3 routing protocols, which are described in this chapter. The simulation is performed in program called Ns-3, more specifically in version 3.14. In the third chapter, this network simulator is described more closely. Here is also mentioned its continuity on previous version Ns-2. The goal of this simulation is to create a network with implemented routing protocols and with certain number of nodes, which has configured mobility, so the network looks like Manet network as much as possible. There are also 2 applications created, which were used to determine the delay for each routing protocol. Another part of thesis is about implementation of the quality of service (QoS) and energy model for each routing protocol. Energy model was used to calculate the remaining energy of node. Next activity summarizes how the implementation of QoS changed delay for used applications and how it changed the energy consumption of a node. The result of thesis is that the voice applications was preferred after the QoS implementation. The value of delay increased for ftp application. Percentually, the improvement for voice traffic was 30-31 %. The increase of delay was percentually 7-19 %. The difference in remaining energy after QoS implementation was about 1%. The actual amount of battery in the beginning of simulation was 1 J.

The master‘s thesis deals with processing of business plan for the estabilishment of café. The teoretical part describes general requirements and conditions for business estabilishment and general structure of business plan. In the practical part the basic analysis of the interal and external enviroment are performed. Then the whole business plan is described - marketing mix, financal plan and time scheldue of implementation of the planned café are described. At the end the whole plan is reviewed and prepared for implementation.

The Company expanded its offered product portfolio and now is focused to small cogeneration unit market. This work proposed a solution of project management in this new market area. The proposed solution is applied to a specific project of installation of small cogeneration unit.

Diploma thesis describes using discrete simulation using with help Witness simulation software as a tool to support decision-making process and the company ECKELMANN l.l.c. This work briefly introduces business processes modeling and simulation problems. Creating a model of real production process and its subsequent optimization will help us map out the processes that take place in the company, enabling discovery of strengths and weaknesses.
