Study of changes in the chemical composition of coffee during roasting

This bachelor thesis deals with monitoring of changes in acrylamide and hydroxymethylfurfural levels in coffee during the process of roasting. Green coffee was roasted using two separate roasting modes (medium roast: temperature 210 °C for duration of 14 minutes, City roast: temperature 220 °C for 17 minutes). Concentration of acrylamide and hydroxymethylfurfural for various roasting durations was determined by high performance liquid chromatography. For the determination of acrylamide, linear growth of the concentration was observed. The final levels of acrylamide corresponded with levels given by the literature, although the characteristic trend of acrylamide formation was not observed. Typical trend of hydroxymethylfurfural formation was observed in samples roasted with City roast mode, while the first part of the trend was also apparent in samples roasted with medium roast mode. Final levels of hydroxymethylfurfural in roasted coffee were within the limits given by the literature.
Processing of sauerkraut juice to enhance its stability and consumer acceptance

This diploma thesis focuses on the development possibilities of fruit and vegetable juices containing sauerkraut juice, the characterization of their nutritional and sensory qualities and their stability, as well as the potential for enriching selected juices with probiotic culture of Lactobacillus plantarum. The best combinations of various juice mixtures were selected based on sensory analysis and were stored for 30 days at 8 °C. Changes in selected parameters were monitored during the storage. Antioxidant activity, total flavonoids and polyphenols content and content of total water-soluble and reducing saccharides were determined by spectrophotometric methods. HPLC was used for lactic and L-ascorbic acid determination, flow cytometry was used for probiotic cell content determination, culture test was used to observe viability of cells, pH was monitored. Contents of antioxidant activity and of total polyphenols content did not drastically change during the storage. Major changes were observed in total and reducing sugar content and total flavonoids content. It was observed that probiotic viability was preserved for 14 days, decrease occurred afterwards. In order to achieve as high content of probiotics as possible, it would be advisable to store probiotically enriched juices for maximum of 14–19 days at temperature of 8 °C. The highest viability was observed in JHM sample (containing apple, pear, carrot and sauerkraut juice in the same ratio). Even through viability of probiotic cells was determined as low, these presented functional beverages remain nutritionally, visually and taste-wise interesting products, which can help popularize the consumption of non-traditional local foods.

Completion of work and rights and duties resulting from defective work This thesis focuses on the analysis and description of selected aspects of a contract for work which are completion of work and rights and duties resulting from defective work. The selected aspects are dealt with in this thesis according to their regulation after the recodification of civil law in Act no. 89/2012 Coll., the Civil Code. This new legislation is analyzed and at the same time compared with the legislation from which it partly arose and that was abolished by the Civil Code, i.e. Act no. 40/1964 Coll., the Civil Code, as amended, and Act no. 513/1991 Coll., the Commercial Code, as amended. The goal of this thesis is to analyze the current legal regulation of the selected aspects of a contract for work and compare it with the previous legislation, then to briefly summarize some conclusions drawn from existing case law in this area, consider comprehensibility and applicability of the new legislation in question, make some recommendations to parties concluding a contract for work and submit several proposals for a modification of the legislation de lege ferenda. Analysis of hip dysplasia occurrence by selected dog breeds

In my thesis I deal with canine hip dysplasia. The paper consist of two parts. The first part summarizes all the information available about this disease. These include factors influencing the emergence of the disease, clinical signs, evaluation systems, diagnosis, conservative ang surgical treatment, prognosis and prevalence. The second part is methodological. There are summarized data about particular breeds of dogs that were obtained Mastiff, Auvergne Pointing Dog, Beauceron, Fila Brasileiro, Brittany, Cavalier King Charles Spaniel, Czechoslovakian Wolfdog, French Pointing Dog, Italian Pointing Dog, Italian Wirehaired Pointing Dog, Small Blue Gascony Hound, Blue Picardy Spaniel, Newfoundland, Portuguese Pointing Dog, St. Germain Pointing Dog, Tibetan Terrier and Golden Retriever. In the end I gave breeding recommendations regarding to reducing the incidence of high grade of canine hip dysplasia.
Dysplasia and locomotive organs diseases by dogs

In my bachelor paper, I describe some of the genetically determined diseases of the musculoskeletal system of dogs. These orthopedic diseases include hip and elbow dysplasia, Legg-Calvé-Perthes disease and patellar luxation. They represent a significant risk in breeding practise. They are influenced by genetic and environmental factors (size and body weight, nutrition, etc.). Each of these diseases are painful for dogs. These diseases are inherited polygenic. Expression of the trait is controlled by multiple genes. Heritability coefficient is in the low to medium values, but for hip dysplasia can be up to 0,8. Breeder of affected animals can choose between conservative or surgical treatment. Because these are hereditary diseases, should be given the attention to selective programs.
