Vladimír Wsól, Ph.D. Title of diploma thesis: Cloning and expression of human AKR1A1 The aldo-keto reductase 1A1 (AKR1A1) is monomeric cytosolic enzyme from aldo- keto reductase superfamily (AKR) with carbonyl reducing activity. The members of AKR superfamily have an important role in reductive reactions in metabolism of some endogenous substrates and in the first phase of biotransformation of xenobiotics. Fifteen members of this large family have been found in the human body. These proteins are expressed in different tissues within all our organism. The highest concentrations of proteins have been detected in the hepatocytes and in the renal cells. The function of AKR1A1 is to catalyse reductive reactions of aromatic and aliphatic aldehydes to the respective alcohols. Among its important substrates belong mevalonate (endogenous substance) and some drugs like anthracykline antibiotics - doxorubicin and daunorubicin. AKR1A1 also participates in biotransformations of sorbitol and in the development of diabetes complications. The internet and private internaional law

The purpose of my thesis is to analyse the regulation related to the Internet and private international law. The Internet is a quickly developing system of interconnected computer networks. Recent expansion of the internet caused that it becomes almost an essential part of our lives which serves either to communicate or to contract. A growing number of internet-users raised a question of a need for more specialized legislation related to the Internet and e-commerce. The thesis is focused on the regulation applicable to the internet and e-commerce and questions raising when rules intended for real-life obligations are being applied on the Internet. The thesis is composed of six chapters, each of them dealing with different aspects of the topic. Chapter One is introductory. It explains the connection between private international law and the Internet and presents relevant legislation of the Czech Republic and the European Union. Chapter Two and Chapter Three explain where the contracting party can expect to be sued and which jurisdiction's law will be applied. Chapter Four concentrates on e- commerce and contracts in cyberspace. Determination of selected pesticides and their metabolites by LC/MS

Pesticides are the substances designed for destroying, preventing and controlling of pests and protection of plants. Their wide usage in agriculture causes contamination of ground, surface and drinking water, because wastewater treatment plants are not able to remove them. Mostly used pesticides are triazine based pesticides. However their usage is worldwide reduced because of their negative influences on human health and contamination of environment especially with their metabolites, about their toxicity are no information. A method for determination of triazine pesticides atrazine, cyanazine, propazine, simazine, terbuthylazine and their metabolites hydroxytrazine, desethylatrazine, desisopropylatrazine, hydroxyterbuthylazine a desethylterbuthylazine was developed and validated in this diploma work. The determination was realized by LC-MS/MS with electrospray ionization. LOD ranged between 0.5-3.2 pg/ml and LOQ between 1.5-9.6 pg/ml. A test of stability of the maternal substances of these pesticides was performed, in which temperature was found to have the biggest influence on their degradation.
