Development of an LC-MS method for determination of new antimalarial drugs in biological matrices.

Artemisinin a representative of Endoperoxide class of drugs and its derivatives particularly artesunate are the most important class of antimalarial drugs used in clinical practice. They are recommended as the first-line treatment of malaria in combination with other longer-acting antimalarial drugs (lumefantrine, piperaquine). As the main skeleton of these compounds lacks UV visible or fluorescent chromophore, earlier methods of detection have used post-column on-line derivatisation or electrochemical detection in the reductive mode. However, these methods suffer from poor sensitivity and selectivity. Within this thesis the whole LC-MS method development for the analysis of artesunate and its major metabolite dihydroartemisin in biological samples from the very beginning was performed. It included tuning of ESI - triple quadrupole MS detector and optimization of chromatographic conditions, particularly mobile phase pH. Artesunate (ARST) and dihydroartemisin (DHA) were assayed in human plasma using artemisisnin as an internal standard. Different approaches of plasma sample treatment, (protein precipitation and liquid- liquid extraction) were optimized and compared. The properties expected from drug product are stability and suitability for manufacturing. All of these properties must be in accordance with therapeutic and regulatory requirements. For this purpose, there are many analytical methods used in the pharmaceutical industry that serve for investigation of stability, quality and possible interactions of active pharmaceutical ingredient with excipients. Accuracy, efficacy, and especially promptness and difficultness of analysis are the criteria for selection of a particular analytical method. Analytical evaluation of drugs and novel drug candidates using HPLC

High performance liquid chromatography (HPLC) is one of the most frequently used analytical techniques for the analysis of drugs. HPLC methods are widely employed in all fields of the modern pharmaceutical analysis - new drugs development, quality control and assurance, the analysis of drugs and metabolites in a biological material (e.g. therapeutic drug monitoring, bioequivalence). The theoretical part of this thesis deals with the main specifics and aspects associated with HPLC analysis of drugs. The experimental part is consisted of six original research papers with appropriate comments divided in to two sections (The analysis of the drug candidates from the group of aroylhydrazone iron chelators and The stability study on selected drugs). The first section concerns with development, validation and application of new HPLC methods in the analysis of drug candidates from the group of iron chelators - pyridoxal isonicotinoyl hydrazone (PIH), salicylaldehyde isonicotinoyl hydrazone (SIH) and pyridoxal 2-chlorobenzoyl hydrazone (o-108). Project of a Database for a Czech Red Cross Critical Evacuation Centre

This bachelor’s thesis deals with design of a database for a critical evacuation center of the Czech red cross. There are theoretical basics described in the first part. It offers basic overview of knowledge, which is important for reaching assigned aims. There is also analysed internal situation of the Czech red cross and defined requirements to the database. Obtained pieces of knowledge are processed in the practical part and they are utilised for the main proposal. Suggested design of the database is summed up in the final part.

Management of Information Flow of Small Software Company

This diploma thesis deals with the management of information flow of a small company using Business Intelligence tools and the data mart. There is defined the problem of working with information from the point of view of selected company in the first part. The next part presents selected theoretical background on the basis of which was the aim achieved. The fourth part analyses the current situation of the company. There is recommended complex improvement of the current situation in the practical part. Selected information management problem is accomplished factually. There are also introduced suitable software tools, which were used for the solution.

METODIKA PRÁCE BUSINESS ANALYTIKA NA DLOUHODOBĚ TRVAJÍCÍCH PROJEKTECH

Dlouhodobě trvající projekty patří do kategorie projektů majících zvláštní charakteristiky. Tato diplomová práce obsahuje mimo jiné studii situace příkladu existujícího dlouhodobě trvajícího projektu a poukazuje na důležité okolnosti, které ovlivňují práci role business analyst (byznys analytika). Na základě studia konkrétního případu je prezentován koncept nové metodologie pro roli byznys analytika, jehož cílem je práci této role zpřehlednit a zefektivnit. Byznys analytik jako role je zkoumán z hlediska postupu jeho činností, vztahu k ostatním projektovým rolím a také k zákazníkovi a ostatním rolím zainteresovaným na projektu. Nový návrh metodiky je zaměřen na minimalizaci problémů a rizik identifikovaných v rámci studia případového projektu. Definuje rozsah činností byznys analytika a techniky, jakými jsou tyto činnosti vykonávány. Celá metodika je podpořena specifikací dokumentů relevantních pro práci byznys analytika, což představuje zejména dokumenty pro samotnou správu požadavků, řízení toku nových požadavků, specifikaci řešení pokrývajících požadavky a hodnocení naplnění zákazníkových očekávání. Všechny části práce jsou zpracovány s ohledem na specifika prostředí dlouhodobě trvajících projektů, kde je zvýšená míra fluktuace členů projektového týmu a nemožnost dlouhodobě alokovat zkušené profesionály.
